AhR and PPARalpha: antagonistic effects on CYP2B and CYP3A, and additive inhibitory effects on CYP2C11.

The aryl hydrocarbon receptor (AhR) is a ligand-activated transcription factor that mediates a spectrum of toxic and biological effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and related compounds. The peroxisome proliferator-activated receptor alpha (PPARalpha) is a member of the nuclear receptor super-family of ligand-activated transcription factors and it functions as an obligate heterodimer with retinoid X-receptor alpha RXRalpha. The aim was to investigate whether the negative cross-talk recently proposed by the present authors between AhR and PPARalpha on CYP4A and CYP1A has any impact on other cytochrome P450 enzymes. Treatment of male Wistar rats with a PPARalpha ligand clofibric acid (CA) induced CYP2B1/2 and CYP3A proteins, activities, and the mRNA expression of CYP2B1, CYP2B2, CYP3A1 and CYP3A2, and suppressed CYP2C11 protein, activities and mRNA expression. AhR ligand Sudan III (S.III) treatment decreased basal and CA-induced CYP2B, CYP3A and CYP2C11 protein, activities and mRNA expression. To the best of the authors' knowledge, this is the first study showing the presence of mutual effects of AhR and PPARalpha on CYP2B and CYP3A and an additive inhibitory effect on CYP2C11 in the livers of male rats.

Human scalp skin and hair follicles express neurotrophin-3 and its high-affinity receptor tyrosine kinase C, and show hair cycle-dependent alterations in expression.

BACKGROUND: Neurotrophin (NT)-3 and its high-affinity receptor tyrosine kinase C (Trk C) are essential for nervous system development. These members of the NT family are also involved in murine hair morphogenesis and cycling. However, their role in human hair follicle (HF) biology remains to be elucidated. OBJECTIVES: To explore the role of NTs in human skin and HF biology. METHODS: The immunoreactivity (IR) of NT-3 and Trk C was studied in human scalp skin and HFs by immunofluorescent and light microscopic immunohistology. Skin biopsies were obtained from normal human scalp containing mainly anagen VI HFs from women (age 53-57 years) undergoing elective plastic surgery. RESULTS: Both NT-3 and Trk C showed prominent, yet distinct, IR patterns in human scalp anagen HFs (anagen VI), whereas they were weakly expressed in catagen and increased again in telogen HFs. Within HF compartments, NT-3 IR was prominent in the outer root sheath, inner root sheath, dermal papilla and connective tissue sheath. Trk C IR was prominent in all HF epithelial and mesenchymal compartments. Outside the HF, both NT-3 and Trk C showed prominent IR in the epidermis, sebaceous glands and sweat glands. CONCLUSIONS: These observations provide the first indication that NT-3 and Trk C are expressed in human scalp skin and HFs, and suggest that Trk C-mediated signalling is involved not only in murine but also in human HF biology. They may be useful in determining therapeutic strategies for the treatment of hair cycle and skin-related disorders.

Regional permeability of coenzyme Q10 in isolated rat gastrointestinal tracts.

The objective of the study was to identify the region with the maximum permeability for low bioavailable coenzyme Q10 (CoQ) in the gastrointestinal tract. To evaluate the regional differences in permeability, male Sprague-Dawley rats, 250-300 g, were anesthetized and the gastrointestinal segments were isolated. Stomach, duodenum, jejunum, ileum and colon tissues were mounted on a Navicyte side-by-side diffusion apparatus. Radiolabeled CoQ (1 microM in DMEM, pH 7.4, 37 degrees C was added to the donor side and the samples withdrawn from the receiver compartment at predetermined time intervals were analyzed using a scintillation counter. Membrane integrity was monitored by 14C-mannitol permeability. The apical to basal permeability coefficients (Papp x 10(-6), cm/s) were 0.32 +/- 0.13, 3.14 +/- 0.89, 1.36 +/- 1.4, 0.83 +/- 0.40, and 1.59 +/- 0.13, for CoQ through rat stomach, duodenum, jejunum, ileum, and colon tissues respectively. The basolateral to apical permeability coefficients (Papp x 10(-6), cm/s) were 1.6 +/- 0.2, 2.2 +/- 1.2, 0.88 +/- 0.12, 1.6 +/- 0.42, and 1.9 +/- 0.41 respectively. Therefore the region of maximum CoQ permeability is duodenum followed by colon and ileum. Jejunum and stomach regions also have fairly high permeability. Therefore CoQ formulations should be made with an aim to target the duodenum to get maximum dosage effect.

Nematodes and water pollution in eastern part of Nile Delta.

Examination of some represented water sites revealed presence larvae and eggs of the nematodes infecting man and animals. Eggs in a descending order of abundance were Trichostrongylus, Toxocara canis, T. vitolorum, and Ascaris sp., then Trichocephalus and Ancylostoma sp. Larvae in a descending order were Strongyloides, Trichostrongylus and Ancylostoma sp. No doubt contaminated or polluted water plays an important role as nematode-borne source.

No alteration in serum leptin levels during acute endotoxemia in sheep.

To determine the role of leptin in endotoxin-induced anorexia in ruminants, circulating leptin levels were measured during acute experimental endotoxemia in sheep. Injection of bacterial lipopolysaccharide (450 ng/kg, i.v.) induced anorexia accompanied with fever and increases in serum levels of cortisol, insulin and glucose which are known to stimulate leptin secretion in rodent and human, while it did not affect serum leptin levels at all. These results indicate that serum leptin levels in sheep during acute endotoxemia are differentially regulated from those in rodent and human, and that leptin might not be involved in the endotoxin-induced anorexia in sheep.

Study of soluble adhesion molecules in some intestinal and tissue helminthes.

Soluble adhesion molecules namely soluble intercellular adhesion molecule-1 (sICAM-1) and soluble E-selectin (sELAM-1) were assayed in hydatid patients with or without complications. It was found that sICAM-1 was significantly increased in patients with hydatid cysts as compared to the control group. In lymphoedemic filariasis cases both sICAM-1 and sELAM-1 showed highly significant increase more than the control group. However, non of both soluble adhesion molecules were significantly elevated in patients with ascariasis, hymenolepiasis, heterophyiasis and strongyloidiasis as compared to controls. The results indicated that SICAM-1 & S ELAM-1 are useful markers for hydatidosis and filariasis, but not for ascariasis, hymenolepiasis, heterophyiasis and strongyloidiasis

A study on Trichomoniasis vaginalis and female infertility.

A total of 280 patients, 240 infertile and 40 pregnant were subjected to thorough history taking, general and local examination for exclusion of organic lesion, laboratory investigations to exclude parasitic, bacterial and fungal infections. Sterile vaginal swab from the posterior fornix was taken, and examined by wet smear preparation, Giemsa staining and cultivation on C.P.L.M. medium for trichomoniasis infection. The mean age of the infertile group was 25.75+/-3.92, and of the control group was 21.6+/-2.38 (in years). The mean duration of infertility was 2.81+/-1.51 (years). Out of 240 infertile women, 18.75% complained of discharge, 17.5% itching, 15.42% dysuria, 14.58% dyspareunia, and 10% had cervical lesion. Of the 40 controls, 5% complained of discharge, 2.5% complained of itching, dysuria, dyspareunia, but none had cervical lesion. Of the total cases (280), 36 (12.9%) had T. vaginalis. The clinical data observed were significantly higher among the infertile group than the control group. Cultures were positive in 14.58% of the infertile group and 2.5% in the control group. The difference between the 2 groups was statistically significant. No doubt, T. vaginalis plays an important role in female infertility.

Studies on patients with Schistosomiasis mansoni, HCV and/or typhoid fever.

Two groups of patients were studied. First one included 50 schistosomiasis mansoni patients, 30 with simple infection, 10 with splenomegaly and with ascites. Second group included 111 patients of whom 20 with pure S. mansoni, 27 with pure HCV infection, 54 with mixed infection of schistosomiasis and HCV and 10 with schistosomiasis, HCV and typhoid fever. Serum transaminases and anti-HCV antibodies performed, showed anti-HCV raised levels in 10% of simple schistosomiasis, 60% in splenomegalic patients, 80% in ascites patients, and 7.1% in controls. Liver function tests in first group were within normal range except in those with ascites. In second group, liver function tests was norma in pure schistosomiasis patients, in pure HCV patients serum bilirubin was normal in 22.2%, AST, ALT and alkaline phosphatase were higher. In mixed infection, serum bilirubin was normal in 18.5%, serum transaminases were higher and alkaline phosphatase was normal among 77.7%. Patients with typhoid fever, HCV and schistosomiasis (12.6%) showed significant increase of liver function as compared with each of pure HCV or HCV and schistosomasis. Results were discussed.

Toxoplasma antibodies in normal and complicated pregnancy.

A total of 70 women with complicated pregnancy and 20 ones with normal pregnancy were selected. They were all RH positive and free from syphilis, brucellosis and malignany. Both groups were subjected to detection of anti-Toxoplasma antibodies levels by the indirect haemagglutination tests (IHAT-IgG) and by ELISA (IgG and IgM). The results showed that all controls (normal pregnancy) were IHA-IgG and ELISA-IgM negative. However, two of the controls (10%) were ELISA. IgG positive. As to the women with complicated pregnancy 68/70 or 97.1% were IHAT positive. When using ELISA, 57/70 or 81.4% were IgG positive and 42/70 or 60% were IgM positive. The results revealed a significant difference between the complicated cases and controls. The diagnosis of abortion, stillbirth, premature labour, or neonatal death due to Toxoplasma infection needs careful and laborious evaluation. The use of ELISA (IgG & IgM) is a must, as it is more sensitive and specific than IHAT.

Evaluation of soluble adhesion molecules in the diagnosis of amoebiasis, giardiasis and toxoplasmosis.

A total of 47 patients with toxoplasmosis (21 cases) with amoebic liver abscess (14 cases) and with giardiasis (12 cases) as well as 14 healthy control were subjected to thorough history taking, clinical examination, stool & urine analysis, complete blood picture, ESR, C-reactive protein, ASO, widal test, blood cultures, liver function tests, serum creatinine, hepatitis viral markers, rheumatoid factor, auto-antibodies, stool culture, rectal snip, chest X-ray, abdominal sonar, level of serum adhesion molecules (sICAM-1, sELAM-1), ELISA detection of Toxoplasma antibodies in serum, liver biopsy, detection and counting of Giardia cysts. In toxoplasmosis group, highly significant increase in serum levels of sICAM-1 (P<0.01) and significant increase in serum levels of sELAM-1 (P<0.05) in comparison to control. However, only sICAM-1 levels were significantly increased in IgM cases more than in IgG cases. In amoebic liver abscess group, both sICAM-1 and sELAM-1 significantly increased when compared with control. In giardiasis group, highly significant increase of serum levels of sELAM-1 was noticed than in control group (P<0.01), while sICAM-1 showed no significant difference (P>0.05). There was no correlation between sELAM-1 and number of cysts in the stool (intensity of infection). Soluble forms of adhesion molecules especially sICAM-1 have the potentiality as good markers of endothelial damage, severity of disease and to less extend load of infection.

IL-10 gene expression as a marker for detection of protective immunity levels with irradiated cercarial vaccine against experimental Schistosoma mansoni infection.

Cercariae were obtained in a large number from the maintained life cycle of S. mansoni. They were attenuated at different doses (20 Kr, 50 Kr, 60 Kr, 70 Kr and 80 Kr) of gamma radiation. Laboratory bred Swiss Albino mice were classified into 7 groups. Five groups were immunized with +/-500 S. mansoni cercariae. Two groups were used as positive and negative controls. All animals were sacrificed after 8 weeks. Spleen cell proliferative responses to Phytohaemagglutinin (PHA) were assessed in all groups. IL-10 was measured by ELISA in serum and splenic cells secretion in-vitro. RNA extracted from freshly isolated liver cells was analyzed for detection of mRNA of IL-10 by reverse transcriptase polymerase chain reaction (RT-PCR). The results showed augmentation of proliferative cell from the spleen in all vaccinated groups except with 80 Kr, irradiated cercariae group. The highest percentage of lymphocytes transformation was recorded among the mice immunized with 60 Kr, irradiated cercariae. After challenge, splenic responses in all groups declined progressively to the control level. IL-10 secretion from spleen cells of all vaccinated groups increased after challenge with the least level in 60 Kr, immunized challenged group. IL-10 mRNA expression was higher among 60 Kr, immunized with irradiated cercariae mice group than 70 Kr, one, but with no expression among 80 Kr, cercariae immunized ones.

Coccidiosis among immuno-competent and -compromised adults.

There was significant increase in coccidia positive cases among Hodgkin lymphoma receiving chemotherapy as compared to control group (p < 0.05). The increase was highly significant in the group of acute lymphocytic leukaemia receiving chemotherapy as compared to control group (P < 0.01). The total percent of cases with single type of coccidia were 12.2% compared to multiple coccidial infections (4.3%). The most commonly associated parasite was Cryptosporidium. The total cryptosporidial infections (single and mixed with other coccidia) were 12.1% of the total studied cases. In the immunocompromised the prevalence was 14.3%. In the immunocompetents with diarrhoea, was 8.3% and in control was 5.6%. Microsporidial infections (single and mixed with other coccidia), being the least detected among the four intestinal spores forming coccidia, were 2.4% in the immunocompromised groups. No Microsporidium was detected in the immunocompetent or adult control groups. The total cyclosporal infections (single and mixed with other coccidia) were 6.3% of the total studied cases. In the immunocompromised groups, the prevalence was (8.3%) where in immunocompetent group, was 2.8%. None was detected in the control subjects. Single cyclospora was 3.0% while mixed Cyclospora represented 3.3% of all studied cases. The Isospora infections (single and mixed) were 3.8% of total patients. The prevalence among immunocompromised group was 5.2% and in immunocompetent patients was 2.8%.

Laboratory evaluation of the insecticidal activity of camphor on the development of Oestrus ovis larvae.

The essential oil of E. globulus leaves or camphor against the maturation of Oestrus ovis larvae was evaluated at the laboratory conditions. Camphor at concentrations 1:0 and 1.1 showed 100% mortality rate. At concentrations of 1:2-1:6 the mortality rate ranged between 45-98%. On the other hand, 38 or 27.5% of the developed pupae emerged to adults but only 36.8% of them were fertile. Camphor is safely used in Medicine. So, it is recommended in controlling the zoonotic myiasis producer, O. ovis.

Experimental infection of rabbits with Fasciola gigantica and treatment with triclabendazole (TCBZ) or BT6: parasitological, haematological and immunological findings.

The responses of rabbits experimentally infected orally with 25 Fasciola gigantica metacercariae each, to treatment with triclabendazole (TCBZ) in a single oral dose of 10 mg/Kg body weight or with the newly synthesized drug (BT6) in repeated oral doses of 500 mg/Kg for 3 consecutive days, were monitored by faecal egg and fluke counts and the enzyme-linked immunosorbent assay (ELISA), both treatments at 4-, 8- and 12- weeks of infection. In all infected rabbits, eggs appeared in the faeces between 10 and 16 weeks post-infection (P1) and Were egg-negative in the TCBZ-treated animals. Compared with the infected untreated control (group B), the reduction in the fluke burdens were 93.75,100 and 100% in TCBZ-treated rabbits (groups C, D and E), respectively and 33.33, 27.08 and 27.08% in the BT6 treated groups F, G and H, respectively. Total red blood corpuscle (RBCs) counts, haemoglobin (Hb) concentration and packed-cell volume (PCV) values in rabbits (group B) indicated the presence of moderate anaemia from the start of infection with a significant decrease from 10-12 weeks onwards. RBCs count, Hb and PCV values from groups C, D and E were significantly higher (p < 0.001) than in those from group B on week 10, 8 and 10 PI, respectively. The increasing was continued till week 16 PI. By two weeks PI there was significant eosinophilia in infected rabbits, maintained at 4 and 6 weeks PI then dropped up to 8 weeks but still statistically higher than the control values. The ELISA detected antibodies against F. gigantica as early as week 2 PI, rising to high levels 6, 10 and 12 weeks later in groups C, D and E, respectively, whilst peaked at 10 weeks in BT6 treated animals, then tended to drop but remained positive through 16 weeks. We conclude TCBZ is a potent fasciolicidic drug. Eosinophilia and anaemia might be indicators for fascioliasis. The ELISA with adult fluke excretory - secretory (ES) antigens could be a feasible method for the diagnosis of experimental fascioliasis in rabbits and post-treatment monitoring.

Use of capillary electrophoresis with UV detection as a screening method to determine histamine in fish samples.

Histamine levels in fish, extracted with methanol, were determined by capillary electrophoresis (CE) using phosphate buffer pH 2.5 and U.V. detection at 210 nm. Histamine was well separated from the other co-extracted components under the given CE condition without any cleanup of the methanol extract. The average recovery of spiked histamine in various types of fish samples was 96%. Using the same methanol extracts from various fish samples, we then compared histamine concentration obtained by CE and fluorometric methods.
