[Use of monoclonal antibodies against human HLA II antigens for the detection of bovine B lymphocytes and macrophages].

The crossreactivity of mouse monoclonal antibodies (MoAbs) (Tab. I) prepared against human HLA-DR and HLA-DP antigens was studied in various bovine cells: lymphocytes from lymph nodes and peripheral blood, adherent (B) and nonadherent (T) lymphocytes, monocytes, granulocytes and platelets. In the immunofluorescence test, MoAbs Bra13, Bra14, Bra20, Bra22, Bra30, Bra70, HL-38 reacted with bovine B lymphocytes and monocytes, but not with other tested cells (Tab. III, IV). These antibodies, except Bra22, were positive with B lymphocytes in the complement dependent cytotoxic test (Tab. II). The similarity of the bovine antigens and HLA-DR antigens determined by used MoAbs was also proved by immunoblotting. Monoclonal antibodies Bra38 and BraFB6 did not react with the bovine cells and separated antigens. The epitope (HLA-DR) recognized by the antibody Bra38 is probably absent in cattle. The presence of HLA-DP analogue determined by the antibody BraFB6 has not been confirmed. The crossreactive MoAbs could be used for the detection of B lymphocytes and macrophages in veterinary immunology.

Unilateral cerebellar hypoplasia.

In a 48-year-old man, marked unilateral cerebellar hypoplasia was diagnosed by CT scans and angiographic examinations, later confirmed on autopsy. Clinical findings in cerebellar hypoplasia/aplasia are discussed here, along with the pathogenesis of cerebellar hypoplasia. In this case, several grand mal seizures and persistent headache were the only neurological symptoms. Repeated neurologic explorations did not reveal any abnormality. The clear demonstration of cerebellar hypoplasia associated with ipsilateral hypoplasia of the vertebral artery favors the concept of a vascular genesis of cerebellar hypoplasia/aplasia.

Nurses' health, age and the wish to leave the profession--findings from the European NEXT-Study.

BACKGROUND AND OBJECTIVES: In many industrialised countries the number of workers with low health is expected to increase in the nursing profession. This will have implications for occupational health work in health care. The European NEXT-Study (www. next-study. net, funded by EU) investigates working conditions of nurses in ten European countries and provides the opportunity to evaluate the role of health with respect to age and the consideration of leaving nursing. METHODS: 26,263 female registered nurses from Belgium, Germany, Finland, France, England, Italy, Netherlands, Poland and Slovakia were eligible for analysis. RESULTS: In most countries, older nurses considered leaving the profession more frequently than younger nurses. 'Health' was--next to 'professional opportunities' and 'work organisational factors'--strongly associated with the consideration of leaving nursing. However, more than half of all nurses with low health wanted to remain in the profession. This group reported rather positive psychosocial working conditions--but also the highest fear for unemployment. CONCLUSIONS: The findings indicate that 'the nurse with low health' is reality in many health care settings. Both positive supporting working conditions but also lack of occupational alternatives and fear of unemployment may contribute to this. Current economic, political and demographic trends implicate that the number of active nurses with low health will increase. Occupational health surveillance will be challenged by this. But NEXT findings implicate that prevention also will have to regard work organisational factors if the aim is to sustain nurses' health and to enable nurses to remain healthy in their profession until retirement age.

Peritoneal mesothelium in vitro: an electrophysiologic study.

OBJECTIVE: In vitro electrophysiologic characterization of the peritoneal mesothelium as a barrier for ion transport during peritoneal dialysis. DESIGN: Experiments were performed on rat's mesentery and diaphragm. Electrophysiologic parameters of the isolated tissue were assessed in the modified Ussing system. RESULTS: We have shown that there are significant differences between electrophysiological parameters of the parietal and visceral peritoneum. The value of the potential difference across the parietal mesothelium lining the abdominal part of the diaphragm (PDmpt) was 0.26 +/- 0.03 mV (mean +/- SEM), with a positive charge recorded from the apical side of the mesothelium. Electrical resistance of the mesothelial monolayer (Rmpt) was Rmpt = 13.6 +/- 1.3 omega.cm2 and 1.4 +/- 0.2 omega.cm2 for the tissue isolated from the diaphragm and mesentery, respectively. Both the mechanical and chemical procedures used for the preparation of the peritoneal membrane model from the diaphragm have been shown to produce similar values of electrophysiological parameters. CONCLUSION: The electrophysiologic approach presented here may be a useful tool for studies of highly permeable biological membranes such as the peritoneal mesothelium. The mesothelial tissue isolated from the rat diaphragm has been shown to be a suitable model for electrophysiologic studies of the peritoneum as assessed by the values of the PDmpt and Rmpt parameters.

Changes of the corneal endothelium after ultraviolet-B exposure in previously photokeratectomized eyes.

The photorefractive effect of excimer lasers is based on an interaction between the 193-nm ultraviolet-C laser beam and the stromal chromophore molecules. Recently, in some patients an increase of subepithelial haze and a regression of refractive effect has been observed following suntanning (UV-B exposure). The aim of the study was to find out the possible endothelial damage caused by photoablation with increasing depth and the effect of subsequent UV-B exposure on previously photokeratactomized eyes. Altogether 12 chinchilla rabbits were treated. Four animals received a -5.0 D PRK; four animals a -15.0 D PRK and four animals a -30.0 D PRK treatment. The endothelial average number, size and variation were determined two weeks post-PRK. Three weeks following PRK, a half of the animals received a 1 J/cm2 ultraviolet-B radiation in a constant dermatological UV-chamber. The endothelial morphology was measured the same way with automated specular microscopy two weeks after UV-B irradiation. After PRK treatment there was no statistically demonstrable change in endothelial morphology. On the other hand, after UV-B radiation all eyes showed a decrease in endothelial number and an increase in size and variation. The ratio of hexagonality decreased, and endothelial rosette formation appeared. The early morphological changes resembled the physiological aging changes. Conditions (deep stromal photoablation, cumulative effect of suntanning or solarium treatments) may exaggerate the physiological aging processes leading to subsequent pleomorphism, polymegatism and cell loss. This may accelerate corneal dysfunction.

Web-based training modules in dermatology.

With the goal of using the full power of the World Wide Web for computer-based education, we developed an interactive system for training in dermatology with a database of several thousand color images. The system consists of lecture scripts, case reports with recorded sound, an online atlas, and a quiz system--all relying on the underlying image database. The images used have been reviewed and rated by a senior dermatologist. At present, they are described with a hierarchical key system that permits searching for images by their content. We have evaluated our statistics on access and the results of an open user survey based on an adaptive questionnaire. The responses to 200 questionnaires completed in a two-month study period are encouraging. The online resources are being used not only for training in dermatology but also for continuing education and reference.

Immunohistochemical characterization of the differentiation state of basal cell carcinomas with special interest for infiltrating relapsing tumors.

A retrospective histological and immunohistochemical study was performed on 66 basal cell carcinomas (BCC). To determine the differentiation stages of epithelial cells in these BCC, three monoclonal antibodies directed against cytokeratins K1, K2, K9 and K10-11 (EE21-06), to cytokeratins K1 to K19 (F12-19), and to corneodesmosin (G36-19) were used in indirect immunofluorescence on paraffin-embedded sections. Three histological groups of BCC with specific cytokeratin immunohistochemical features were distinguished: (1) superficial BCC were unlabelled, (2) nodular and variant (keratotic, adenoid) BCC showed an homogeneous labelling, and (3) infiltrative aggressive-type BCC showed a heterogeneous cell to cell labelling. Some nodular BCC cells presented characteristics of granular keratinocytes, i.e. they were labelled by the anticorneodesmosin antibody. All the clinically recurrent tumors were found to be of the infiltrative aggressive-type. If these aggressive forms of BCC were not identified by specific marker, their topographic patterns of labeling with antibodies directed to cytokeratins allowed them to be distinguished. We suggest that an immunohistochemical analysis with antibodies specific for different stages of keratinocyte differentiation is an efficient complement to histological diagnosis of BCC.
