The effect of the thromboxane A2/prostaglandin endoperoxide receptor antagonist SQ 30,741 on myocardial infarct size and blood flow during myocardial ischemia and reperfusion.

The effect of the thromboxane A2 (TXA2) receptor antagonist SQ 30,741 on infarct size and myocardial blood flow during coronary occlusion and reperfusion was determined. In anesthetized dogs, the left circumflex coronary artery (LCX) was occluded and after 10 min a continuous infusion of SQ 30,741 (1 mg/kg + 1 mg/kg/h, i.v.) or saline was begun. After 90 min of LCX occlusion, the LCX was reperfused for 5 h and infarct size was then determined. Myocardial blood flows before, during, and after occlusion were determined using radioactive microspheres. SQ 30,741 resulted in a significant decrease in infarct size (34% +/- 6% of left ventricular area at risk) compared to controls (60% +/- 9%). Cardioprotection was also found with SQ 30,741 when infarct size was normalized for both area at risk and predrug collateral flow. The protective effect of SQ 30,741 occurred without an increase in collateral flow. At 1 h postreperfusion, subendocardial flow was significantly higher in SQ 30,741-treated animals (109 +/- 15 ml/min/100 g) compared to controls (71 +/- 16 ml/min/100 g). SQ 30,741, in the dose resulting in infarct size reduction, produced a 95% inhibition of platelet TXA2 receptors throughout the experiment as measured by dose-dependent inhibition of the ex vivo platelet shape change response to U-46,619, a TXA2 mimetic. Thus, a dose of SQ 30,741 that results in TXA2 blockade also results in myocardial salvage without changes in collateral flow.

Interspecies cross-reactivity of bovine lymphocytotoxic sera with pig lymphocytes.

Forty-three bovine BoLA antisera were tested on pig lymphocytes by a microlymphocytotoxicity test. Twenty-five were found to be cytolytic. Fifteen sera detected the A blood group antigen on porcine lymphocytes but showed no reaction with the J antigen on bovine lymphocytes. Six BoLA reagents reacted with all pig cells tested. Cross-reactions with SLA antigens were observed in only four sera, the highest correlation being recorded with SLA-W7 (r = 0.87). Bovine alloantisera are not of value for SLA typing.

Lesional keratinocytes express OKM5, Leu-8 and Leu-11b antigens in lichen planus.

Biopsy specimens of involved and uninvolved skin were studied in 16 patients with lichen planus using murine monoclonal antibodies against several cell surface markers of effector and/or accessory cells of the immune system. In contrast to uninvolved skin, keratinocytes were found to express HLA-DR, OKM5, Leu-8 and Leu-11b antigens in all biopsies from involved skin. We demonstrate here for the first time the in vivo expression of several effector and/or accessory cell markers on keratinocytes in lichen planus.

B cell acute lymphoblastic leukemia (ALL) in donor cells following bone marrow transplantation for T cell ALL.

Leukemia recurred in a child with acute lymphoblastic leukemia after a bone marrow transplant. The pre-transplant leukemia was of T cell origin whereas the post-transplant relapse leukemia was of B cell origin. The recurrence was shown to be in donor cells by analysis of chromosomes and restriction fragment length polymorphisms. Although the mechanism of leukemia induction is uncertain, the possibilities of donor relapse due to EBV infection or chromosomal exchange were disproven. An alternative mechanism must account for leukemia induction in this patient.

Recombinations between IRP and cystic fibrosis.

A candidate gene for cystic fibrosis was recently isolated by selective cloning of HpaII-tiny-fragment islands; it maps considerably closer to CF than does MET or D7S8 (pJ3.11), and DNA polymorphisms from this region are in marked disequilibrium with CF. cDNA cloning has shown that this protein has a growth factor-like structure and shows homology to the murine and human proto-oncogene int-1; it is designated IRP (int-1-related protein). DNA sequences from the IRP locus that recognize RFLPs are proving to be highly informative for prenatal diagnosis. We report five crossovers that have been identified which occur either within the IRP locus or between IRP and CF; these recombinants demonstrate that CF maps between the DNA markers D7S8 and KM.19.

[Preparation of monoclonal antibodies against cell surface antigens in cattle].

The BALB/c mice were immunized three times with bovine lymphocytes and thrombocytes in vivo. Besides, dissociated spleens of the mice were immunized with bovine lymphocytes in vitro. Hybridomas producing monoclonal antibodies were formed as a result of the fusion of the spleen cells with the murine myeloma cells. On the whole, four fusions were performed after immunization in vivo and three fusions after immunization in vitro, and 70 stable hybridoma clones were obtained. Five monoclonal antibodies exhibited an identical specific reaction only with bovine thrombocytes, two antibodies reacted only with a certain limited population of bovine spleen cells. The remaining monoclonal antibodies exhibited no tissue specificity and were bound to the lymphocytes of peripheral blood, lymph nodes, thymus, and spleen, to thrombocytes, liver cells, and spermatozoa, but never to erythrocytes. As for the amount of the obtained hybridomas and specific antibodies, no significant difference was observed in the effectiveness of in vivo and in vitro immunization.

[Differential diagnosis of jaundice and fever of unknown origin: ruptured aortic aneurysm with obstruction of the bile ducts].

A 65-year-old man with fever of unknown origin developed progressive extrahepatic cholestasis. Radiological examination documented a suprarenal abdominal aortic aneurysm. Fatal intraabdominal bleeding occurred, leading to death. Post-mortem examination revealed an earlier retroperitoneal rupture of the aortic aneurysm and massive perianeurysmal inflammation. The blood masses, reaching the porta hepatis, caused progressive obstruction of the extrahepatic bile ducts. Surgical treatment, delayed because of cholestatic and febrile complications, could have prevented the outcome.

[HLA typing in patients with Melkersson-Rosenthal syndrome].

27 patients suffering from Melkersson-Rosenthal syndrome (MRS) and 46 of their first degree relatives were investigated with special reference to histocompatibility standardization. In contrast to a control group of 223 healthy German volunteers, our MRS patients revealed high-yet statistically not significant-levels of HLA-B16 and HLA-Cw3 antigens.

The mitochondrial reading frame RF3 is a functional gene in Saccharomyces uvarum.

The yeast mitochondrial genome contains three reading frames, RF1, RF2, and RF3, which are related to the group I maturases, though they are not intronic sequences. In the Saccharomyces cerevisiae strain D273-10B/A, these reading frames are interrupted by G + C-rich sequences (GC clusters) which break the frames. In the present work we described a Saccharomyces uvarum strain which possesses a RF3 continuous sequence devoid of GC clusters. Moreover, our results strongly suggest that in the same strain RF2 and RF1 are also continuous sequences. As all three RFs belong to transcription units which are highly expressed, it is most reasonable to assume that RF1, RF2, and RF3 are functional genes. Furthermore, we have discovered a rule which seems to explain the transposition of GC clusters, considered as mobile elements, in the mitochondrial genome.

Chronic liver iron overload in the baboon by ferric nitrilotriacetate. Morphologic and functional changes with special reference to collagen synthesis enzymes.

Two baboons receiving intramuscular injections of ferric nitrilotriacetate over a two-year period were compared with two control baboons. The results indicate that in iron-overloaded animals: liver iron excess was major (maximal liver iron concentration values of 42 mumol/100 mg dry weight for both animals vs 1.3 +/- 0.2 (mean +/- SD) in controls) and chronic (for 15 months liver iron concentrations were higher than 15); iron deposition, although less abundant than in sinusoidal cells, was pronounced within parenchymal cells; serum transaminase activities were markedly increased; rare foci of perisinusoidal fibrosis were observed in areas of massive iron overload; and a dramatic decrease in hepatic 4-prolyl-hydroxylase activity was found, in contrast with unchanged glucosyltransferase and galactosyltransferase activities. In conclusion these findings suggest that, in our model, chronic liver iron overload: exerts a marked biochemical cytolytic effect; and does not produce significant hepatic fibrosis, possibly related to an inhibiting effect of ferric nitrilotriacetate complex on 4-prolyl-hydroxylase activity.

A 20S particle ubiquitous from yeast to human.

We have purified and characterized a particle sedimenting at 20S from the postribosomal fraction of yeast, wheat germ, Drosophila melanogaster tissue culture cells, chicken embryo fibroblasts, rabbit reticulocyte lysate, and HeLa cells. Most of the protein constituents of the 20S particle have molecular weights of 20-35 kd and differ between species; however, some do have similar molecular weights and isoelectric points, suggesting they are related. Several low-molecular-weight RNAs, distinct from tRNAs, co-purify with the particle isolated from all these species and show increasingly more complex patterns ascending the arbitrary order from yeast to human (yeast, plant, insect, bird, and mammals). In Drosophila, we present evidence that these small RNAs are tightly associated with this 20S structure.
