Amplification of c-yes-1 proto-oncogene in a primary human gastric cancer.

Abnormalities of cellular oncogenes in 22 cases of human primary gastric cancer were screened by Southern blot analysis using 16 onc probes. Human cellular sequences related to the v-yes oncogene of Y73 avian sarcoma virus (human c-yes-1 gene) were found to be amplified in a primary gastric cancer. The degree of amplification was about 4- to 5-fold. Another v-yes-related cellular gene (human c-yes-2, a pseudogene) was not amplified in this tumor. The normal stomach tissue adjacent to the tumor tissue in the same patient showed no amplification of c-yes-1 gene, suggesting that the amplified c-yes-1 is involved in the onset or the progress of this carcinoma.

[Laser acupuncture therapy for pain and vague complaints in neurosurgery].

The value of diode laser acupuncture for pain and other vague neurological complaints has been studied in chronic patients of head trauma and of cerebrovascular disease or neuralgia in head and neck regions. Gallium-Aluminium-Arsenide (Ga-Al-As) diode laser pain attenuator PANALAS-4000 manufactured by Japan Medical Laser Laboratory was used in the following specifications; wave length: 830 nm, power: 20-40 mW, frequency: 5-50 c.p.s., pulse duty: 0.9. The laser acupuncture was applied to some of the representative Chinese meridian points, local pain points and related nerves for 1 to 7 times in a week. It was given in 310 patients in our neurosurgical department. 175 of 227 patients who complained of pain and 12 of 39 patients who complained of numbness showed improvement within 24 hours, another 24 patients with pain and 16 patients with numbness showed improvement after 24 hours. No side effect was noticed. The duration of the effectiveness was about 2 or 3 days. It is very useful to apply the laser acupuncture for pain and vague complaints in neurosurgery.

Localization of angiotensin converting enzyme (dipeptidyl carboxypeptidase) in swine sperm by immunofluorescence.

Testis and epididymis are known to have high amounts of angiotensin converting enzyme (dipeptidyl carboxypeptidase, EC 3.4.15.1). We investigated the localization of the enzyme in these tissues by an immunofluorescent technique and found that the enzyme was localized in the spermatids and residual bodies in the Sertoli cells of the testis. Furthermore, the enzyme was shown to be present in the cytoplasmic droplet of epididymal sperm and also in detached cytoplasmic droplets in semen. The enzyme was not detected in the interstitium of testis and epididymis except for the endothelial cells of the vessel.

Revertants and partial transformants of rat fibroblasts infected with Fujinami sarcoma virus.

Fifteen revertants were isolated from three independent clones of rat fibroblasts transformed by Fujinami sarcoma virus (FSV). Three revertant clones resulted from the deletion of the one copy of the FSV provirus, and one encoded an enzymatically inactive, transformation-defective protein. The remaining revertant clones were characterized by a transcriptional block of the provirus. Digestion of chromosomal DNA with MspI and HpaII revealed that the FSV provirus was hypermethylated in these revertants, whereas proviral DNA of their spontaneous retransformants was hypomethylated. Furthermore, the revertants had lost DNase I-hypersensitive sites in and around the FSV provirus. The effect of transcriptional regulation of the FSV provirus was further analyzed in clones showing various degrees of phenotypic transformation. We quantitated v-fps mRNA levels in these cells by liquid hybridization and found that increasing levels of viral RNA correlated with a more pronounced transformed phenotype. These results suggest that transcription of FSV proviral DNA is under both viral and cellular control and that transformation by FSV is a function of the dosage of v-fps mRNA.

Gamma-aminobutyric acid-induced contraction of the dog basilar artery.

gamma-Aminobutyric acid (GABA) produced a biphasic action, that is, a relaxation followed by a contraction in the isolated dog basilar artery. Repeated applications of each concentration of GABA (10(-6), 10(-5), 10(-4), 10(-3) M) at regular intervals under resting conditions caused a stepwise increase in the contractile response. Marked reproducible contractile responses occurred (ED50: 1.3 X 10(-5) M), after at least 8-10 applications. The GABA-induced contraction as well as relaxation was blocked by bicuculline and picrotoxin. Pretreatment with inhibitors of prostaglandin synthesis such as aspirin (3 X 10(-6) to 3 X 10(-4) M) and indomethacin (3 X 10(-6) M) reduced the contractile response to GABA in an irreversible manner and without affecting the relaxation induced by GABA. These inhibitors increased the resting tone but reduced the tone in strips subjected to repeated applications of GABA. 15-Hydroperoxyarachidonic acid at concentrations of 1 X 10(-6) and 1 X 10(-5) M also attenuated the contractile response to GABA in a dose-dependent fashion. These observations suggest that GABA may modulate the contractile effect by inducing the production of contraction-causing prostaglandin(s) mediated through GABA receptors and/or by some as yet undefined mechanism.

[Randomized systemic chemotherapy using two or three drug combination for small cell lung cancer (SCLC)].

Randomized trials were sequentially performed in SCLC patients using a two-drug combination (ACNU . VCR, ADM . VCR) and a three-drug combination (ACNU . CPA . VCR, ACNU . ADM . VCR). The response rate for the two-drug regimen was 36.4% (8/22 cases) and that for the three-drug combination was 60.0% (15/25 cases). The median survival time of the two groups was 7 and 9 months, respectively. The group with PS. 0-1 showed a better prognosis than that with PS. 2-3 (p = 0.03). No fatal side effects were experienced in this protocol. These data suggest that the three-drug combination (ACNU . VCR . CPA) represents an active regimen for SCLC.

Triphenyltetrazolium and its derivatives are anisotropic inhibitors of energy transduction in oxidative phosphorylation in rat liver mitochondria.

Triphenyltetrazolium and its derivatives inhibited energy transduction in mitochondria but not in submitochondrial particles, which are inside-out relative to the membranes of mitochondria. Triphenyltetrazolium incorporated into the inside of submitochondrial particles inhibited ATP synthesis in the particles. Triphenyltetrazolium also inhibited the reduction of NAD by succinate coupled with oxidation of succinate by O2 and hydrolysis of ATP. Energization of mitochondrial inner membranes with succinate and with ATP induced sites on the membranes for triphenyltetrazolium and its derivatives. The maximum amounts of energy-dependent binding sites for triphenyltetrazolium on membranes energized with succinate and ATP, respectively, were 14 and 4 nmol/mg protein. Triphenyltetrazolium also induced H+ ejection from the energized membranes. The maximum amounts of H+ ejection from membranes energized with succinate and ATP, respectively, were 4 and 2.4 nmol/mg protein. Triphenyltetrazolium also decreased the membrane potential up to about half the control value and caused shrinkage of mitochondria in an energy-dependent fashion. Comparison of the Hammett's sigma constants of triphenyltetrazolium derivatives with various substituents on the 3-benzene ring showed that lower concentrations of triphenyltetrazolium derivatives with a stronger positive charge were required for inhibition of energy transduction. The present findings show that triphenyltetrazolium and its derivatives act as anisotropic inhibitors of energy transduction by binding to negative charges created on the outer side (C-side) of energized mitochondria, and that the positive charge of these inhibitors is one of important factors for their inhibitory activity. These negative charges may be an essential part of the H+ pump.

The analysis of cytostatic activity of human peripheral blood granulocytes and its augmentation with Nocardia rubra cell wall skeleton (N-CWS).

Granulocytes of normal volunteers showed substantial levels of cytostatic activity against tumor-cell-derived cell lines. Spontaneous cytostatic activity of the granulocytes was observed after 24 h incubation and reached the maximal level within 48 h. Lysates of granulocytes also showed significant cytostatic activity. The cytostatic activity of the granulocytes was significantly augmented by in vitro incubation with Nocardia rubra cell-wall skeleton (N-CWS). Maximal augmentation of granulocyte-mediated cytostasis was achieved with 2.5 micrograms N-CWS per ml after 24 h incubation when the granulocyte-to-tumor-cell ratio was 10:1. The N-CWS itself showed no direct effect on the uptake of 3H-thymidine by tumor cells.
