Effect of carrier on the immunogenic capacity of synthetic cholera vaccine.

In the design of the synthetic antigens and synthetic vaccines, primary consideration should be given to the choice of the carrier. Since small peptides which are being used as the relevant antigenic determinants are likely to be poor immunogens as such, the augmentation of their immunogenic capacity by the carrier or any other means is crucial for the induction of immunity. In the present study, we explored several approaches for the enhancement of the immune response towards synthetic peptides derived from the B-subunit of cholera toxin. The results indicate that the use of tetanus toxoid as a macromolecular carrier, polymerization of the peptide without any external carrier and the conjugation of dipalmityl side chain had comparable effects in enhancing the immune response to several synthetic peptides. This effect was manifested both at the level of antibodies produced and in their capacity to neutralize the biological activity of the cholera toxin. Prior exposure to the carrier resulted in a dose-dependent suppression against the synthetic epitope attached to it.

Modified stock-eye ocular prosthesis.

A technique for the use of modified prefabricated eyes was described. When properly used, many of the limitations attributed to prefabricated eyes can be overcome. This technique allows the proper positioning of the iris while simultaneously recording an impression of the ocular defect tissue bed. This technique permits the finished ocular prosthesis to generate an equal distribution of pressure throughout the defect, while increasing its movement due to excellent adaptation to the walls of the socket. Although a custom-eye technique may still be necessary for some, most patients can be treated with the modified prefabricated-eye technique with excellent results.

The covalent linking of two nucleotide analogues to antibodies.

Two anticancer drugs, antagonists of nucleic acids, were covalently linked to antibodies specifically reactive with B leukemia cells and thus with a potential possibility of drug targeting to the tumor site. The drugs cytosine 1-beta-D-arabinoside and 5-fluorouridine, competitive inhibitors of enzymes involved in DNA synthesis, were linked to antibodies via a dextran bridge. Cytosine 1-beta-D-arabinoside was linked to periodate-oxidized dextran and fluorouridine to dextran hydrazide. The dextran derivatives were in turn linked to antibodies that recognized a specific membrane IgM on B leukemia cells. The drug-antibody conjugates maintained most of the original antigen-binding capacity of the antibody and the full pharmacological activity of the drugs.

Prosthetic treatment of earlobe keloids.

An ear clip prosthesis has been developed for maintaining pressure on earlobe keloids before and after surgical removal. The prosthesis includes an ear clip to which heat-polymerized acrylic resin is attached, which covers the keloid area.

Major salivary glands in branchial arch syndromes.

In five patients with branchial arch syndromes the presence of the salivary glands was demonstrated. Clinical, sialographic, and scintigraphic examinations revealed morphologic defects and diminished function of the parotid glands. Hyperfunction of the submandibular glands was assumed in the three patients in whom scintigraphic studies were performed. The findings may be of diagnostic value in patients in whom the clinical signs of the syndrome have remained occult.

Both cholera toxin-induced adenylate cyclase activation and cholera toxin biological activity are inhibited by antibodies against related synthetic peptides.

The immune response against six synthetic peptides corresponding to various segments of the B subunit of cholera toxin was evaluated. Conjugates in which the peptides were covalently linked to tetanus toxoid served for immunization of rabbits. As previously reported, four of these conjugates elicited antibodies cross-reactive with intact cholera toxin. We report here that antisera against two of these synthetic peptides inhibit the entire spectrum of activities of the intact cholera toxin. This is manifested both on the biochemical level (adenylate cyclase induction) and on the biological effect (intestinal fluid secretion). These results indicate that these peptides may serve as suitable candidates for preparation of a synthetic anticholera vaccine.

HLA antigens A, B, C and DR in hay-fever families of similar environmental conditions.

Investigation of 98 members (healthy and affected) belonging to 17 hay-fever families, for clinical picture, total and specific IgE and HLA A, B, C and DR is presented. There was no significant correlation between hay-fever, total or specific IgE and a certain HLA antigen or haplotype. There was, however, an association between hay-fever and same haplotype within 4 of the 17 families.

Survey of immunoglobulin E levels in atopic families in Israel.

Two hundred and ten members belonging to two consecutive generations of 41 atopic families (healthy and affected by clinical atopic manifestations) were investigated for basal immunoglobulin E (IgE) levels and clinical picture. The results show overall increased IgE levels, above standard values in the whole population studied, and a significant correlation between clinical atopy and high IgE levels in the children of the group. Within these families recessive inheritance of abnormally high IgE levels suggests that their high IgE levels are due to a genetically transmitted defect in IgE suppression.
