Effect of Ba2+ on the K+ conductance pathways in the frog cornea.

Two types of transepithelial potential difference (PD) responses have been observed in the bullfrog, Rana catesbeiana, when the K+ concentration is changed in the aqueous solution. (1) A normal response, that is, a decrease in the positivity of the aqueous solution when the K+ is increased in this solution. (2) An anomalous response, that is, an increase in PD when K+ is increased from 0 to 4 mM in the aqueous solution. In present experiments 2 mM Ba2+ results in a significant decrease in transepithelial PD and an increase in resistance (R), consistent with the well-known effect of Ba2+ on the K+ conductance in other biological membranes. In the presence of Ba2+ compared to its absence the normal PD responses were decreased when K+ was increased from 4 to 20 or to 79 mM in the aqueous solution. Barium enhanced, but not significantly, the anomalous PD response (PD increase) when K+ was increased from 0 to 4 mM. An anomalous PD response (PD decrease) was obtained with Ba2+ when K+ was changed from 4 to 0 mM while in its absence the response was normal (PD increase) or did not change. These findings support the concept that anomalous PD responses as a result of the electrogenic (Na+ + K+)-ATPase may be obtained when the resistance of the simple K+ pathway is increased.

Evidence for HCO3- conductance pathways in nutrient membrane of bullfrog antrum.

The effect of changing the nutrient HCO3- concentration on potential difference (PD) and resistance in bullfrog antrum bathing in CI- media was determined. Changes in HCO3- concentration were from 25 mM to several lower concentrations and back to 25 mM. A plot of /delta PD/ versus log [HCO3-] gave a linear relation for changes of HCO3- concentration from 25 down to 3.1 mM and back to 25 mM but deviated to some extent for changes to 1.6 mM. In these experiments, changes from higher to lower HCO3- concentrations gave a less rapid initial PD response than those in the reverse direction. This result eliminated H+ conductance pathways as being predominant. Experiments were done in which in the first part changes were made in nutrient solution from 5 percent CO2 and 25 mM HCO3- to 0.6 percent CO2 and 3 mM HCO3- and in the second part the same changes with a simultaneous changes of secretory solution from 5 percent to 10 percent CO2. The magnitude of PD decrease was greater by 4.5 mV in the second part. This result indicated that HCO3- conductance pathways rather than OH- conductance pathways are predominated . There was no evidence of HCO3-, OH-, and H+ conductance pathways in secretory membranes.

Human X chromosome markers and Duchenne muscular dystrophy.

Two DNA markers, a random DNA fragment 754 and the cDNA sequence encoding the gene for ornithine transcarbamylase (OTC) have been studied in kindreds segregating for Duchenne muscular dystrophy. 754 and OTC are located close physically to the mutation in the region Xp21 below the breakpoints in two Duchenne females. The genetic distance was found to be approximately 10cM between 754 and DMD (two crossovers in 26 meioses) and to be approximately 10cM between OTC and DMD (two crossovers in 26 meioses). Physical data suggest the order DMD-754-OTC. The frequency of recombination compared to physical distance between these markers and DMD suggests that there may be a hot spot of recombination. The relevance of these observations for the isolation of the DMD mutation and clinical use of these probes is discussed.

Regenerating fish optic nerves and a regeneration-like response in injured optic nerves of adult rabbits.

Regeneration of fish optic nerve (representing regenerative central nervous system) was accompanied by increased activity of regeneration-triggering factors produced by nonneuronal cells. A graft of regenerating fish optic nerve, or a "wrap-around" implant containing medium conditioned by it, induced a response associated with regeneration in injured optic nerves of adult rabbits (representing a nonregenerative central nervous system). This response was manifested by an increase of general protein synthesis and of selective polypeptides in the retinas and by the ability of the retina to sprout in culture.

Computed tomographic analysis of brain morphometrics in 30 healthy men, aged 21 to 81 years.

Computed transverse axial tomography (CT) was employed to examine brain morphometrics in 30 healthy men, aged 21 to 81 years. Seven consecutive CT slices 30 to 80 mm above the inferior orbitomeatal line were analyzed. CT numbers in gray and white matter regions were not correlated significantly with age (p greater than 0.05), nor were right/left ratios for derived parameters. The volume of gray matter was correlated negatively with age (p less than 0.05), and the volume of cerebrospinal fluid correlated positively with age, in the seven slices. The volumes of the lateral and third ventricles were elevated in the elderly, and volumes of the thalamus and lenticular nucleus were reduced. The results demonstrate that brain atrophy, evidenced by a loss of gray matter and by dilatation of cerebrospinal fluid spaces, occurs in the healthy elderly, and provide baseline CT-derived brain morphometric data for healthy men in relation to age.

Segregation analysis of a marker localised Xp21.2-Xp21.3 in Duchenne and Becker muscular dystrophy families.

A DNA marker C7, localised Xp21.1-Xp21.3, has been studied in kindreds segregating for Duchenne muscular dystrophy (DMD) and Becker muscular dystrophy (BMD). In DMD families four crossovers were observed in 38 informative meioses between C7 and the DMD locus (theta = 0.12, z max = +2.72). In BMD families no recombinants were observed in the 16 informative meioses studied. These data are consistent with the localisation of the mutations in these disorders being in the same region of Xp21. Studies in families also segregating for the DNA marker 754 support the previously reported physical order of these loci as X centromere-754-DMD-BMD-C7-X telomere. A recombination fraction of 0.11 (z max = +5.58) was found between DMD-754 by combining our previously published data with the data presented here. C7 and 754 thus provide good bridging markers for the diagnosis of DMD and BMD.

Computer-assisted categorization of brain computerized tomography pixels into cerebrospinal fluid, white matter, and gray matter.

A computer-assisted method was employed to estimate the amounts of cerebrospinal fluid (CSF), white matter, and gray matter in individual computerized tomography (CT) scans of brains. By means of an image processing procedure (DMORPH), the means +/- SD CT numbers of "pure" CSF, white matter, and gray matter were determined in each scan and stored. A CATSEG program used these means to define ranges for CT numbers for each of the three tissues on each scan, and to assign each pixel in a scan to one of the three categories. Summing over seven serial scans provided volumetric estimates of CSF, white matter, and gray matter in a brain segment. For 10 subjects aged 21 to 43 years, CSF volume equaled 1.4 to 4.7% of the total segment volume, white matter equaled 37.5 to 48.2%, and gray matter equaled 50.2 to 58.9%. Image processing hardware and software which allow standardized sampling from CT images for the evaluation of surface areas and CT numbers are described. These procedures, as applied to CT scans of the human brain, can be used to estimate the volumes of CSF, white matter, and gray matter in a selected intracranial segment.

A new transorbital surgical approach to the rabbit's optic nerve.

This work describes a surgical approach which establishes the rabbit's visual system as an experimental model for studying CNS regeneration. Using this model, the optic nerve, its cell bodies, and the axons, are easily accessible through an orbital approach, without the need for craniotomy and brain retraction. This surgical approach allows transplantation and 'wrap around' implantations of nerve segments from xenogeneic and syngeneic systems and diffusible substances derived from them, respectively. Furthermore, it enables studies aimed at determining deficiencies in mammalian CNS and investigating methods of augmentating mammalian CNS regeneration.

Indirect effects of the 3'-5' cyclic adenosine monophosphate binding protein (CAP) on the transcription of the malPQ operon in Escherichia coli.

Uninduced malPQ transcription, as followed by measuring beta-galactosidase expression in a strain carrying a malP-lacZ hybrid gene and grown in the absence of maltose, requires the presence of CAP. However this requirement is lost when the expression of malT, positive regulator gene of the maltose regulon, is rendered independent of CAP by a mutation in the malT promoter. This result suggests that the effect of CAP on uninduced malPQ expression is mediated through a modulation of MalT protein synthesis. The effect of CAP on the induced expression of malPQ is presumably mediated, in addition, through a modulation of the synthesis of the maltose transport system and, hence, of the entry of the inducer. Therefore the effect of CAP on malPQ expression seems to be merely indirect, and this is surprising since a CAP binding site is present at the malPQ promoter.
