Production of mycotoxins by Fusarium species isolated in Germany. 2. Time course of deoxynivalenol and 3-acetyldeoxynivalenol formation by Fusarium graminearum in different liquid media.

Several semisynthetic liquid media were examined for the large-scale production of deoxynivalenol (DON) und 3-acetyldeoxynivalenol (AcDON) by Fusarium graminearum 183. Only in three of the eight media used could high toxin yields of DON and AcDON be detected. The maximum levels of DON in a medium according to Miller were 3 mg/l and of AcDON 32 mg/l. In glucose-yeast extract-peptone (GYEP) medium containing 1% glucose, the AcDON concentrations reached 33 mg/l and the DON yields were 19 mg/l. In a rice flour liquid medium, however, the mean levels of AcDON and DON increased to 170 mg/l and 9 mg/l, respectively. The maximum amounts observed were 480 mg/l for AcDON and 65 mg/l for DON. The addition of trifluoracetic acid sodium salt or malonic acid, which are suggested to cause an accumulation of acetyl-CoA by inhibiting enzymes of the tricarboxylic acid cycle, did not stimulate the toxin formation.

Beta-thromboglobulin in pre-eclampsia.

Beta-thromboglobulin was tested in blood and urine in a study group of pre-eclamptic patients. The control group comprised a subgroup of normal non-pregnant women and a second subgroup of normal pregnant women. The results of the plasma beta-thromboglobulin blood test revealed no significant difference between the groups, whereas the urine of the pre-eclamptic group showed a significantly increased concentration of beta-thromboglobulin. This difference may reflect renal involvement in the pre-eclamptic patients; the turnover of platelets may be increased but the plasma level of beta-thromboglobulin is apparently maintained by the increased renal clearance.

[Resistance of Staphylococcus aureus to methicillin].

Data on methicillin-resistant Staphylococcus aureus have not been presented before in Argentina. Due to non adequate microbiologic techniques, this phenomenon may be overlooked. Heterogeneous strains are only detected with 5% NaCl agar, low temperature (30 degrees C) or prolonged incubation and/or high inocula (10(6) C.F.U.). Minimal inhibitory concentrations (MICs) of methicillin were studied by the agar dilution technique in antibiotic medium number 4 with and without 5% salt. This study was performed for 107 strains at 37 degrees C, after 24 and 48 h incubation, with an inoculum size of 10(6) CFU. S. aureus strains were considered resistant whenever MICs were greater than or equal to 8 mg/l. Forty seven (44%) methicillin-resistant strains were detected. Thirty one were high level resistant mutants and sixteen showed the heterogeneous phenomenon. Due to the high frequency of resistant strains among clinical specimens in this country, the routine use of elaborate screening tests with special media and or prolonged incubation time is justified.

Allergic reactions to enzymes used in plant cloning experiments.

We report a case of rhinitis and asthma resulting from exposure to enzymes used in an experimental plant pathology laboratory. Immediate skin-test responses were elicited to both "Cellulase" and "Macerozyme" products, and a large late onset skin reaction occurred at the Cellulase site beginning in 6 hr. Special radioallergosorbent test (RAST) substrates were prepared that showed a strong positive response to Cellulase. By the RAST method other laboratory workers were screened and another symptomatic individual was found who also showed evidence of type I hypersensitivity to both enzymes. Skin biopsy and immunodiffusion data are presented. We conclude that enzymes used to digest cell wall structures of plants are capable of eliciting both immediate and late onset skin-test reactions and positive RAST responses in susceptible persons. These reactions are thought to be mediated by IgE antibodies and can be associated with symptoms of rhinitis and asthma.

Müllerian malformations and simultaneous pregnancies in didelphys uteri. Review and report of a case.

Congenital anomalies originating in the mullerian ducts are among the causes of gynecological and obstetrical disturbances that are commonly overlooked. These cases reach the rate of 1,1--3.5 per cent in hysterographic examinations. Some authors have stressed the need to keep this type of lesion in mind and to try to diagnose these conditions correctly, especially when the patients are examined for trivial complaints in the non-pregnant state, as well as during the prenatal period, or after delivery. The case presented in a multiparous Beduin woman with simultaneous pregnancies in both cavities of an unexpected didelphys uteri. The first twin, female 2,000 gram, was born at home, and the second, female 2,250 gram by cesarean section, following unsuccessful attempts at vaginal delivery. The decision to undertake surgery was taken for common obstetrical reasons like inertia uteri and high presentation. Proper early diagnosis should result in adequate management and the earlier end of labor.

Determination of palladium in gasoline by neutron activation analysis and automated column extraction.

Palladium in gasoline was determined by means of neutron activation analysis (NAA) and selective sorbent extraction. Unleaded gasoline consistent with DIN EN 228, RON 95 was irradiated at a thermal neutron flux of phith = 1.68 x 10(13)s(-1)cm(-2) and an epithermal neutron flux of 3.32 x 10(11)s(-1)cm(-2) for t(irr) = 12 h. The irradiated gasoline was digested with nitric acid and palladium was then separated as N,N-diethyl-N'-benzoylthiourea complex by an automated column pre-concentration procedure. The eluate of 50 microL was dried on a filter paper and the 88.03 keV photons resulting from the decay of 109Pd were detected in a low level HPGe spectrometer with an efficiency of 35.5%. Severe interferences with other matrix constituents, especially 82Br could be overcome and the detection limit for palladium was improved to 3.4 ng/L at a confidence level of 90%. Although the analytical procedure applied yielded the lowest detection limit for palladium obtained in gasoline up to now, no indications for the presence of palladium were found.

Survey of injuries and protective gear worn by in-line skaters in public parks.

In-line skating, also known as rollerblading, has become popular as a recreational activity as well as a mode of transportation. Increased participation has been accompanied with a marked rise in major and minor injuries. The objective of this study was to survey active in-line skaters and to describe their demographic features, formal training, protective gear used, and a history of incurred injuries. A questionnaire was distributed to randomly selected in-line skaters in a large public park in New York City. Of the 223 skaters who responded, 128 were male and 95 were female. Ages ranged from 12 to 64 (mean, 29) yr. Some kind of injury was reported by 87 (39%) of the participants. The majority of those injuries occurred when the skaters were beginners (46). Skin abrasions and musculoligamentous injuries were the most common. Only 15 (17.2%) sought medical treatment, and of these, 5 were treated for fractures: 3 at the wrist and 1 each at the ankle and knee. The knee was the most commonly injured part of the body (24.7%), followed by the elbow and wrist. Approximately 5% of all injuries involved the head. Many skaters did not wear helmets or knee pads, even though they owned this equipment. There was a strong preference for wearing wrist pads, either alone or with other protective gear. This suggests that skaters have learned that the wrist is particularly vulnerable to serious injury and should be protected.

Apoptosis of cultured neuroblastoma cells is induced by ceramide and not by ligation of the Fas/Apo-1/CD95 receptor.

The expression of the Fas/APO-1/CD95 receptor on the neuroblastoma cell lines IMR-32, Kelly, SK-N-SH, LS and SiMa was investigated. The induction of apoptosis was attempted by incubation with Fas antibody IgM and IgG, Fas ligand and with ceramide. All neuroblastoma cell lines proved to be positive for Fas/APO-1/CD95 receptor expression by FACS. However, propidium iodide staining in FACS showed that neither incubation with the Fas antibody nor with the Fas ligand resulted in convincing apoptosis of the neuroblastoma cells. On the other hand, incubation with ceramide rapidly led to all signs of apoptosis morphologically and in the FACS. Therefore, other pathways than Fas/Apo-1/CD95 ligation must be of significance for the apoptosis of these neuroblastoma cell lines.
