Evaluation of a rapid latex test for direct detection of Streptococcus agalactiae in various obstetrical and gynaecological disorders.

Cervical samples collected during speculum examination in 413 women were tested for the presence of Streptococcus agalactiae by Gram staining, a new latex agglutination test and standard cultures. The results of each method are reported in correlation with the clinical circumstances of isolation. Gram stain and the latex agglutination test had a sensitivity of 30% and 78%, and a specificity of 89% and 98% respectively compared with culture. The sensitivity of the latex agglutination test was higher in cases of premature rupture of membranes (90%), presumed chorioamnionitis (86%), presumed post-partum endometritis (86%) and presumed salpingitis (87%). This latex agglutination test appears to be useful for identifying patients who are heavily colonized with Streptococcus agalactiae and are at a high risk of infection.

Preclinical antitumor activity of a soluble etoposide analog, BMY-40481-30.

BMY-40481-30 is a new, water-soluble derivative and probable prodrug of etoposide characterized by the presence of a phosphate group in position 4' of the E ring of the etoposide molecule. The compound was only weakly cytotoxic in vitro and, consequently, an investigation of its antitumor activity was conducted in several murine and human tumor (xenograft) models. Etoposide was administered ip or po whereas BMY-40481-30 was given ip, po or iv. The potency of the derivative, when administered parenterally, as defined on the basis of maximum tolerated dose (MTD), was less than the parent compound on a weight (mg/kg) basis in some experiments but comparable to etoposide in other instances. Comparison at the MTD of the two compounds showed that BMY-40481-30 administered ip was as active as etoposide against ip P388 leukemia. BMY-40481-30 given iv was more active than etoposide given ip in two of five experiments versus iv P388 leukemia, but the two compounds were comparably active in the other three studies. Of particular interest was the finding that the derivative was more active than the parent compound at many of the comparable (on a mg/kg basis) dose levels of both evaluated po versus iv P388 leukemia; MTD levels were not achieved, and hence not compared, for either compound using the po route of administration. Both etoposide and BMY-40481-30 yielded comparable maximum effects against ic P388 leukemia, ic L1210 leukemia, and sc B16 melanoma, but etoposide was more efficacious versus sc M5076 sarcoma.(ABSTRACT TRUNCATED AT 250 WORDS)

Inhibition of methanogenesis in pure cultures by ammonia, fatty acids, and heavy metals, and protection against heavy metal toxicity by sewage sludge.

The effect of ammonium chloride, sodium butyrate, sodium propionate, and the heavy metals nickel, zinc, and copper on methanogenesis by pure cultures of Methanospirillum hungatei, Methanosarcina barkeri, Methanobacterium thermoautotrophicum, and Methanobacterium formicicum at pH 6.5 was studied. The latter three strains were resistant to greater than 60 g/L of the volatile fatty acids and to greater than 10 g/L of NH3 N. Methanospirillum hungatei was somewhat more sensitive with 50% inhibition of methanogenesis occurring at 4.2 g/L NH3 N, 27 g/L butyrate, and 41 g/L propionate. All strains were very sensitive to both copper (1-5 mg/L) and zinc (1-10 mg/L), but much more resistant to nickel. Zinc and copper concentrations 30 to 270 times higher were required to cause inhibition of Msp. hungatei incubated in sewage sludge compared with buffer, indicating a strong protective environment was afforded the methanogens against heavy metal toxicity in the sludge.

[Latex agglutination test and counterimmunoelectrophoresis in the diagnosis of Haemophilus influenzae meningitis].

Authors have searched for Haemophilus influenzae by culture and by detection of capsular antigens in CSF of 1.700 bacterial meningitis in Dakar. Culture has been positive in 71.9%, counter-immunoelectrophoresis in 94.9% and latex (slidex meningitis kit) in 91.2% of cases. Diagnosis results are improved of 39% comparatively with bacteriologic culture. False positive results caused by antigenic cross-reactions have been observed only in 1.3% of others bacterial meningitis. Latex-test is allowing a diagnosis in three minutes and a quantitative determination of antigens (useful for pronostic).

[The latex agglutination test for the diagnosis of meningococcal A and C, pneumococcal and H. influenzae b meningitis. A study of 920 purulent cerebrospinal fluids (author's transl)].

The cerebrospinal fluids of 920 Senegalese patients with purulent meningitis wee examined by latex particle agglutination (LA) and the results were compared with those of conventional bacteriology and of counter immuno electro phoresis (C.I.E.). For all three organisms taken globally, CSF culture was positive in 72.7% of the patients, the LA test in 78.8% and the C.I.E. test in 89.0%. The LA test was positive in 17.9% of subjects with negative CSF culture. When the results wee analysed for each organism separately, the LA test was positive in 69.3%, 74.7% and 85.9% respectively of patients with meningococcal, pneumococcal and H. influenzae b meningitis. False agglutinations were found in 6.7% of meningitis due to other pathogens. Cross-reactions and autoagglutinations were rare (0.4% and 3.7% respectively). The sensitivity, specificity, simplicity and rapidity of the LA test make it extremely useful for the diagnosis of purulent meningitis, particularly in Africa.
