Saliva level of ethynylestradiol in presence of ethynodiol diacetate after oral administration.

In the presence of ethynodiol diacetate the ethynylestradiol concentration in serum and saliva after oral administration was measured by a rapid radioimmunoassay method developed by the authors. By means of the saliva/serum quotient, and from the concentration of ethynylestradiol in the saliva samples, it was possible to infer the ethynylestradiol content of the serum. Attention is called to the relation between the quotient and time, which should be taken into account when values are calculated. 2, 4 and 12 h after oral administration, the saliva/serum quotients for an ethynylestradiol dose of 0.05 mg were 0.27, 0.76 and 0.40, respectively.

[Gonadotropin treatment in corpus luteum insufficiency-induced functional sterility].

The authors report on gonadotropin treatment of 91 women who were infertile due to luteal dysfunction. As an effect of treatment there was an improvement of the corpus luteum function in 77 patients (84.6%), and 30 of them (33.0%) became pregnant. Application of HCG is recommended in cases if clomiphene treatment proved ineffective previously. Application of FSH and HCG is recommended at low level of estrogen.

[Treatment of corpus luteum insufficiency-induced functional sterility with clomiphene].

Two hundred and fifty-six women were treated with clomiphene citrate. As a consequence of treatment preovulatory FSH concentration of the serum increased. There was an improvement of the luteal function in 84.0% of the patients, and 53.9% of them became pregnant. Based on the above data the authors consider clomiphene citrate suitable for treatment of luteal dysfunction. In a number of cases clomiphene treatment proved to be effective even after failure of the gonadotropin treatment.

Prolactin synthetizing capacity of the human decidua during the first trimester of pregnancy.

In vitro prolactin (PRL) synthesis by human decidua of early pregnancy (5-12 weeks gestation) was investigated. Tissues obtained during therapeutic abortions were incubated in oxygenated Gey's buffer supplemented with 20% fetal calf serum at 37 degrees C for 24 h. PRL production increased rapidly to 42.3 mU PRL/100 mg tissue wet weight until 8 weeks gestation and then slightly declined but these differences were not significant. There was not PRL production when chorion was incubated under similar conditions. A significant correlation was found when the initial PRL content in the decidua prior to incubation and the capacity of decidua to synthesize PRL were compared (r = 0.76, P less than 0.001). These data provide further evidence that human decidua can produce PRL in vitro. The finding that the rate of the PRL synthesis increases progressively in early pregnancy is in correlation with the intensive decidualization of the endometrium.

[Results of treatment with bromoergocryptine in functional infertility caused by corpus luteum insufficiency].

Eighty women were treated with bromocryptine (Parlodel, Sandoz). They were infertile in consequence of luteal insufficiency. The luteal function got normalized i 83.6% of the patients, and 31.3% of them became pregnant. The dopaminergic treatment was most successful in the case of those hyperprolactinemic women, who were infertile due to luteal dysfunction only. The treatment was less effective in case of normoprolactinaemia, and in case of luteal dysfunction with other reasons of infertility, too.

Parlodel treatment of patients with pathospermia.

The response to bromoergocryptine (Parlodel, Sandoz) was examined in 42 pathospermic patients. The authors conclude that Parlodel did not influence the spermiogram considerably. Serum prolactin levels were lowered significantly, while the FSH, LH, testosterone, 17-alpha-hydroxyprogesterone and 17-beta-oestradiol concentrations did not change. Improvement was reported in potentia coeundi in one third of the patients.

Serum FSH and LH levels in men after administration of a long-acting LH-RH preparation.

The effects of the natural gonadotropin releasing hormone (LH-RH) and of a long-acting analogue, desgly10-D-leu6-LH-RH ethylamide, on the serum FSH, LH, testosterone and oestradiol-17 beta levels were studied in healthy men. The basal value for FSH was 3.58 +/- 0.47 mU/ml (SE), for LH 6.33 +/- 1.19 mU/ml. Following an intravenous dose of LH-RH the values increased by 3.40 +/- 0.88 and 25.99 +/- 5.67 mU/ml, respectively. After intravenous administration of the long-acting analogue, the peak FSH exceeded the basal value by 12.89 +/- 6.95 mU/ml, while LH increased by 41.27 +/- 4.72 mU/ml. The effect of the analogue on FSH and LH lasted 693 min and 862 min, respectively. Neither preparation changed the serum testosterone or oestradiol-17 beta levels essentially. The results suggest that the long-acting variant of LH-RH is more suitable than the natural hormone for examination of the FSH and LH reserves in men.

Peripheral concentrations of gonadotrophins and sexual steroids in infertile males.

Relationships between the spermiogram and sex hormones in 104 andrologic patients were studied. Endocrine connections in normal and in pathologic spermiogenesis have been studied. A rather close correlation was found between FSH and ICSH levels. ICSH increases parallel with the increase of FSH level. There is also a close relationship between sperm concentration and FSH level. Between ICSH and sperm count the correlation is not significant. Between steroid hormone level and sperm concentration no correlation ws found. It is only the plasma FSH level which show a close relationship with the disorder of spermiogenesis and it has diagnostic significance.
