Glycosaminoglycan composition of PC12 pheochromocytoma cells: a comparison with PC12D cells, a new subline of PC12 cells.

PC12D cells, a new subline of conventional PC12 cells, respond not only to nerve growth factor but also to cyclic AMP by extending their neurites. These cells are flat in shape and are similar in appearance to PC12 cells that have been treated with nerve growth factor for a few days. In both cell lines, we have characterized the glycosaminoglycans, the polysaccharide moieties of proteoglycans, which are believed to play an important role in cell adhesion and in cell morphology. Under the present culture conditions, only chondroitin sulfate was detected in the media from PC12 and PC12D cells, whereas both chondroitin sulfate and heparan sulfate were found in the cell layers. The levels of cell-associated heparan sulfate and chondroitin sulfate were about twofold and fourfold higher in PC12D cells than in PC12 cells, respectively. Compared to PC12 cells, the amounts of [35S]sulfate incorporated for 48 h into chondroitin sulfate were twofold lower but those into heparan sulfate were 35% higher in PC12D cells. The amount of chondroitin sulfate released by PC12D cells into the medium was about a half of that released by PC12 cells. The ratio of [35S]sulfate-labeled heparan sulfate to chondroitin sulfate was 6.2 in PC12D cells and 2.2 in PC12 cells. These results suggest that there may be some correlation between the increase in content of glycosaminoglycans and the change in cell morphology, which is followed by neurite outgrowth.

The GTP-binding proteins, Go and Gi2, of neural cloned cells and their changes during differentiation.

The levels of the alpha-subunits of two GTP-binding proteins, Go and Gi2, were determined in neural and nonneural cloned cells by immunoassays. Go alpha was detected in all neural cells and some of nonneural cells, but not in HL-60 leukemic cells and PYS-2 teratocarcinoma-derived cells. The level of Go alpha was highest in the PC12 pheochromocytoma cells. Gi2 alpha was present in all types of cells tested, and its level was highest in the HL-60 cells and relatively high in glioma cells. Treatment of PC12 cells and neuroblastoma x glioma hybrid NG108-15 cells with nerve growth factor and forskolin, respectively, caused the extension of neuronal-like processes and increase in the level of Go alpha by 60-80%, but small changes in the levels of Gi2 alpha.

Surgical treatment of tumors in the anterior skull base using the transbasal approach.

Five patients with tumors in the anterior skull base were surgically treated using the transbasal approach, which permits removal of the tumor, repair of the dura mater, and reconstruction of the skull base in a one-stage procedure. By using autologous materials for the bone graft and pedunculated pericranial flap for the reconstruction, the intracranial structures are separated from the air-filled nasal and paranasal cavities. No postoperative complications such as wound infection and leakage of cerebrospinal fluid were encountered. The use of this surgical technique makes it possible to extirpate brain tumors that heretofore have been considered unresectable.

[One-year chronic oral toxicity study of propiverine hydrochloride in dogs followed by one-month recovery].

The chronic oral toxicity of propiverine hydrochloride (P-4), a new anti-pollakiuria agent, was studied in beagle dogs. Groups of 6 males and 6 females were treated with P-4 at doses of 0, 0.3, 1, 3, 9 mg/kg/day for one year and thereafter 2 animals of both sexes in each group were placed on withdrawal for one month. During administration and recovery period, no death occurred in any dosed animals. As a toxic sign, only the frequency of vomiting was increased in animals of 1, 3 and 9 mg/kg/day groups. Body weight, food and water consumption were not affected by the P-4 administration. In serum chemical examinations, gamma-GTP activity was increased in both sexes of 9 mg/kg/day group at 6 month of administration. Further decrease in total and free cholesterol, triglyceride and phospholipid, increase in GPT activity were detected in some animals of 9 mg/kg/day group at 12 month of administration. In addition decreasing tendency in levels of albumin was noted in males of 9 mg/kg/day group at 9 and 12 month of administration. And also, a gradual increase in total protein level and a gradual decrease in alkaline phosphatase activity were seen in control group, but in females or males of 9 mg/kg/day group, those changes were mild. Urine pH rised slightly in females of 3 mg/kg/day group and in both sexes of 9 mg/kg/day group. No specific findings attributable to P-4 treatment were detected in ECG, heart rate, funduscopy, hematology, fecal occult blood test and necropsy. The absolute and/or relative liver weight in males of 3 and 9 mg/kg/day groups were significantly increased. Light-microscopically, the hypertrophy of hepatocytes characterized by homogenization and enlargement of cytoplasmic space, and concentric inclusions in hepatocytic cytoplasm were detected in both sexes of 3 and 9 mg/kg/day groups. Corresponding to these microscopical findings, the following changes were observed electron-microscopically, the proliferation of smooth surfaced endoplasmic reticulum in hepatocytes in both sexes of 1, 3 and 9 mg/kg/day groups, lamellar bodies in hepatocytes in females of 3 mg/kg/day group, and in both sexes of 9 mg/kg/day group, and annulate lamellae in hepatocytes were detected in one female of 9 mg/kg/day group. After the recovery period, the above mentioned abnormalities were markedly attenuated or disappeared except the changes in hepatocytes. From these results, it seemed that 9 mg/kg/day of P-4 might be safety dose.(ABSTRACT TRUNCATED AT 400 WORDS)

[von Recklinghausen's disease associated with malignant lymphoma].

A 59-year-old man was admitted to the Saga Medical School Hospital in November 1986 because of unconsciousness and lymph node swelling. He had café au leit spots and multiple neurofibromas since his thirties. The leukocyte count was increased. The level of serum LDH and Ca were extremely elevated. His bone marrow aspiration showed the invasion of lymphoma cells. Lymph node biopsy from his left neck and the study of surface markers were performed. And then, his illness was diagnosed as non-Hodgkin's lymphoma (diffuse, medium sized cell type, B cell type, stage IV B.) following von Recklinghausen's disease. After the admission, his general condition improved by chemotherapy, but he had a relapse 6 months later. And he died in November 1987. The case of von Recklinghausen's disease associated with malignant lymphoma in rarely reported. We reviewed the literature and discussed von Recklinghausen's disease associated with malignant lymphoma including this case.

[Changes of high molecular weight kininogen in various states].

Changes of high molecular weight kininogen (HMW-K) clotting activity, antigen and cleavage in the plasma in the health and various diseases were studied. In 20 healthy individuals clotting activity of HMW-K, as measured by APTT one stage method, was 99 +/- 12% (male) and 84 +/- 15% (female). Antigen as measured by Laurell method were 106 +/- 24% (male) and 91 +/- 21% (female). In 35 patients with disseminated intravascular coagulation (DIC), both activity (78 +/- 33%) and antigen (69 +/- 31%) were statistically lower than those in normal individuals (p less than 0.01). In DIC both activity and antigen of HMW-K was correlated with serum albumin level. These results suggest that the cause of the lower level of HMW-K in DIC especially with septicemia is the result of lower production rather than consumption. In vivo cleavage of HMW-K was detected in plasma of a patient with septicemia and DIC by immunoblotting. The change of HMW-K was also assessed in other pathological states including liver cirrhosis, collagen disease, cardiopulmonary bypass and pregnant women.

[Acute biphenotypic leukemia followed by two color flow cytometry].

In this paper is reported a case of acute biphenotypic leukemia who was treated by chemotherapy and pursued its effect by two color flow cytometry. A 33-year-old male patient was admitted due to fever and general fatigue and diagnosed as acute leukemia by hematological findings. Surface markers were investigated to find positive reaction of Leu 12 (CD19), J 5 (CD10), My 7 (CD13) and My 9 (CD33), in which Leu 12 and My 9 were simultaneously expressed on the same blast cells by two color flow cytometry. He was treated with daunorubicin, enocitabine, mercaptopurine, vincristine, and prednisolone to obtain partial remission. Then, he was administered L-asparaginase, doxorubicin, vincristine and prednisolone to reach complete remission. The effect of chemotherapy was investigated by not only bone marrow puncture, but also by two color flow cytometry. From the findings in this case, the two flow cytometry was proved to be a useful tool for not only diagnosis of acute mixed leukemia, aut also the judgement of the effect of treatment.

[Effect of inorganic anions on the activity of dental pulp lactate dehydrogenase].

Pig dental pulp lactate dehydrogenase (EC. 1.1.1.27; LDH) isozymes were separated by DEAE Sephadex A-50 column chromatography, and LDH-1 isozyme was purified by blue dextran affinity chromatography. The specific activity of pig dental pulp LDH-1 was 193.2 units/mg protein, and the purified enzyme showed a single protein band on disk electrophoresis. In this paper, we studied the effect of inorganic ions on the activity of pig dental pulp LDH-1 isozyme, and compared it with that of affected by anions. Dental pulp and heart muscle LDH-1 isozymes were inhibited at a low pyruvate concentration; otherwise, at a high pyruvate concentration, each enzyme activity was enhanced by the anions. But the effect of SO2(2-) was different from that of the other anions: SO2(2-) did not activate the enzyme at a high pyruvate concentration. In the dental pulp, the Km values for pyruvate were 1 order of magnitude smaller by SO2(2-), and 1 order higher by other anions than the control value. In the heart muscle, the Km values were not changed by anions, except by SO2(2-).

[Genetic markers and thrombin reaction in a family of Bernard-Soulier syndrome].

A family with Bernard-Soulier syndrome (BSS) was investigated with reference to the genetic markers and thrombin reactions. The proband was a 24-year-old man with a life-long history of epistaxis and gingival bleeding. His parents were first cousins; furthermore, his father was born to parents of second cousins. His father also had bleeding tendency and was also diagnosed as having BSS. However, his mother and elder brother were normal. Genetic marker analysis among the family members suggested that the 16th chromosome was associated with the development of BSS, because only the haptoglobin genotype coded on the 16th chromosome was the marker in both the proband and his father. In addition, they both exhibited decreased thrombin-induced platelet aggregation at a low dose, but an almost normal reaction at a high dose of thrombin.

[Acute myeloblastic leukemia associated with chronic thyroiditis].

A 57-year-old woman who suffered from acute myeloblastic leukemia during the course of chronic thyroiditis, is described. The patient was diagnosed as having chronic thyroiditis in 1984 when she was 53 year-old, and was treated with L-T4.Na. She admitted in July 1988 because of general fatigue, fever, cough and sore throat. On admission, hematological examination in the peripheral blood showed marked anemia and increased leukocytes with 20.5% leukemic cells positive for peroxidase staining. Bone marrow aspiration showed 38.8% leukemic cells. She was diagnosed acute myeloblastic leukemia. She reached complete remission after combination chemotherapy. The case of acute myeloblastic leukemia associated with chronic thyroiditis is rarely reported. We reviewed the literature and discussed acute myeloblastic leukemia associated with chronic thyroiditis including this case.

[Surgical treatment in dissecting aneurysm of the infrarenal abdominal aorta--report of two cases].

In this paper, we present two cases of dissecting aneurysm in the infrarenal abdominal aorta and a review of this type of lesion. DeBakey's classification has found wide acceptance since it combines both anatomical description and a basis for management. However, there is another type of the aneurysm, omitted in this classification, which involves the infrarenal segment of the abdominal aorta, the intimal tear being distal to the renal arteries. Its clinical manifestation, therefore, differs from dissecting aneurysm of the thoracic aorta. The incidence of dissecting aneurysm in the lower abdominal aorta in the literature is 2-14%. Sixteen cases of atraumatic dissecting aneurysm in the abdominal aorta, including our two, have been reported in Japan. Radioimaging techniques such as ultrasound, computerized tomography with contrast enhancement and conventional angiography, allow diagnosis of dissecting aneurysm. Computerized tomography with contrast enhancement has led to more frequent preoperative diagnosis of dissecting aneurysm in the abdominal aorta. However, precise visualization of the intimal defect together with the site of entry is a prerequisite of operation. Angiography remains the most suitable method of achieving this end. Although both abdominal and thoracic aortic dissection share a common management in respect to hypotensive therapy, we believe that surgical intervention is required, especially in dissection of the abdominal aorta, with prosthetic replacement of the infrarenal segment and obliteration of any proximal or distal false lumen.

[Differentiation induction of myeloid leukemia cells by glucocorticoid--the differentiation induction effect in vitro and in vivo].

The effects of glucocorticoid on the differentiation of myeloid leukemia cells were examined. Dexamethasone at 10(-6)M or 10(-7)M revealed marked effects not only on leukemic blasts' survival but also on its' differentiation in vitro. In 10 of 17 cases of myeloid leukemia, the obvious morphological and functional differentiation induction effects were observed in vitro. The direction of differentiation were differed in leukemia cell lineage and in cases. Granulocytic differentiation in M2 cells, mono-macrophage differentiation in M5 cells and either granulocytic or monocytic differentiation in M4 cells were induced. A AML (M2), it's leukemia cells were induced into granulocytic pathway by dexamethasone in vitro, was treated with prednisolone (40 mg per day).Ara-C (15 mg per day). The increase in peripheral leukocyte count and the decrease in immature cells were observed simultaneously. The peripheral leukocytes mainly consisted of intermediate forms of granulocytes and Pelger-Huët like neutrophils probably originated from leukemia cells. After that course, abnormal clone was eliminated from bone marrow. A AMoL, it's leukemia cells were induced into macrophage like cells completely by dexamethasone in vitro, was treated with prednisolone (30 mg per day) and complete remission was obtained without passing through a hematological nadir. It is indicated that anti-tumor effects of glucocorticoid on myeloid leukemia cells are closely related to it's differentiation inducing effect and glucocorticoid can be used as the drug intending for the differentiation induction therapy of acute myeloid leukemia.

In vivo efficacy of levamisole against larval stages of Angiostrongylus cantonensis and A. costaricensis.

Anti-larval effects of levamisole were examined on A. cantonensis in rats and A. costaricensis in mice. 1) In rats inoculated with 40 infective larvae of A. cantonensis: Compared with a non-treated control group, a significant reduction in number of worms recovered was seen in the group receiving a single dose of 1.0 mg/kg or more. A significant decrease in host lung-body weight ratio was seen in the group receiving drug of 3.0 mg/kg or more. 2) In mice inoculated with 20 infective larvae of A. costaricensis. In the non-treated control group, a severe loss in body weight and death of host animals were observed. A single dose of 30 mg/kg on 3, 4 or 5 days post-infection remarkably inhibited these changes. At 30 mg/kg for 3 or 7 days levamisole was more effective than a single dose of the drug. These results suggest that levamisole has conspicuous in vivo effects against larval stages of A. costaricensis as well as A. cantonensis.

[Arterial infusion cancer chemotherapy in liver tumor using implantable reservoir].

Four primary liver tumors and 19 secondary ones (8 gastric, 8 colorectal, 2 esophageal and 1 gall bladder cancer) were treated by intra-arterial infusion chemotherapy intermittently using implantable reservoir. Drugs used in this series were 5-Fluorouracil, Mitomycin C, Adriamycin, and Cis-platinum. They were infused every 2 weeks for outpatients. The antitumor efficacy was evaluated in terms of tumor regression measured by CT scan. The toxicity was slight and temporary. The total response rate was 23.1%, 50% survival period was 7 months, and 1-year survival rate was 24.2%. The response rate was 33.3%, and the 50% survival period was 7 months for 8 gastric cancer patients with liver metastasis. The result was not good, and we must improve the criteria of indication and devise a regimen. But the implantable drug delivery system has made it possible to lengthen the time that patients may stay home and assure good quality of life because of the freedom of movement, normal physical appearance, protection from infection, and reduction of mental burden, due to subcutaneous placement.

[Urinary cytodiagnostic abnormality in a patient with malignant lymphoma].

A 69-year-old man was admitted due to chronic subdural hematoma. He had lymphadenopathies in the neck. The diagnosis of non-Hodgkin lymphoma (diffuse, large, B cell type) was made by a cervical lymph node biopsy. The renal failure by obstruction of the urinary tract developed gradually, and the tumor cells were found by urinary cytodiagnosis. He was treated immediately with chemotherapy. After the treatment, the urine volume increased and the general conditions improved. It was indicated from this case that the urinary cytodiagnosis is useful for the detection of urinary tract infiltration by lymphoma cell. The urinary cytodiagnostic abnormalities in patients with non-Hodgkin lymphomas were discussed from the literature, including this case.
