Purification and some properties of Candida albicans exo-1,3-beta-glucanase.

An exo-1,3-beta-glucanase was purified from blastoconidia of Candida albicans 1001. The purified enzyme appeared as a single protein band by PAGE, and split into two subunits (Mr approximately 63,000 and 44,000) when analysed by SDS-PAGE. The pI of the enzyme was 4 and a Km of 1.7 mg ml-1 was estimated for laminarin as substrate. Despite its very reduced activity on the synthetic substrate p-nitrophenyl beta-D-glucoside, C. albicans exo-1,3-beta-glucanase hydrolysed 1,3-beta-glucan by an exo-splitting mechanism and was inhibited by glucono-delta-lactone and by Hg2+ and Ag+ cations. The active exo-glucanase was mainly located in the periplasm, but it was also present inside the cytoplasmic membrane in small amounts and was secreted into the culture medium. The electrophoretic mobility of the enzyme from all three locations was the same.

A map of barley chromosome 2 using isozymic and morphological markers.

The F2 progeny of a cross between a chromosome 2 multiple marker stock and an adapted cultivar of barley were analyzed for four morphological markers and electrophoretic patterns of eight leaf isozymes. The Idh-2 locus was linked to the Per-5 locus (27.96 +/- 5.07 cM) and to the e locus (10.26 +/- 3.13 cM). Also, the Per-5 and e loci were located on the short arm of chromosome 2. In addition Idh-2 was also located on barley chromosome 2 and was linked to the v locus (13.18 +/- 3.56 cM), which is located on the long arm of chromosome 2. Two other marker genes, li and wst,,B, were linked (26.50 +/- 5.24 cM) on chromosome 2 but segregate independently of the other loci evaluated.

Long-term effects of MPA on human progeny: intellectual development.

Tests of verbal and spatial ability were done on 450 boys and 537 girls in their late teens of whom 73 and 97, respectively, had been exposed in utero to MPA. Exposed boys achieved higher raw scores than controls on verbal and spatial tests but the differences were explained by their more favorable demographic and social characteristics. Exposed girls did not differ from controls. Although, mothers of exposed boys reported that their offspring talked and walked later than controls, our results support the hypothesis that intrauterine exposure to MPA at contraceptive doses has no long-term effect on intellectual development.

Slow infusion of vincristine in the treatment of refractory thrombocytopenic purpura.

Eight patients with idiopathic thrombocytopenic purpura (ITP), who were refractory to glucocorticoid therapy, were given slow infusions of vincristine (VCR) over a 4- to 6-hour period at weekly intervals for 4 weeks. Three patients showed a return to normal platelet counts maintained for 3 months or longer. A transient recovery was observed in 1 patient and a partial response was observed in 3 patients. All patients tolerated therapy well, without side effects. In conclusion, therapy with slow infusion of VCR can be effective in refractory ITP.

Yeast beta-glucanases: a complex system of secreted enzymes.

The complement of glucan-hydrolysing enzymes produced by yeast species is considered to be relevant to morphogenesis, since this polysaccharide is the major structural component of the yeast cell wall. beta-glucanase systems of different species consist of both 1,3-beta- and 1,6-beta-hydrolases, which exert their action by either an exo- or an endo-splitting mechanism. The diversity of beta-glucanase systems, even among related species, is striking. The enzymes are produced for export, and the active forms of some of them seem to be generated upon secretion. Genetic approaches and the cloning of the glucanase genes are being used to clarify the biological role of the different enzymes. These also offer a possibility for genetic analysis of the functions related to cell wall dynamics with potential applications in biotechnology.

Influence of some inhibitors of arachidonic acid metabolism on oxytocin contractions in the isolated testicular capsule of the rat.

Oxytocin (50-750 nM) contracted the isolated testicular capsule of the rat. Mepacrine, a phospholipase A2 inhibitor (3 X 10(-5) M) and the lipoxygenase inhibitor nordihydroguaiaretic acid (10(-5) M) inhibited this response whereas the cyclooxygenase inhibitor, diclofenac sodium (10(-5) M), and the thromboxane synthetase inhibitor imidazole (10(-5) M) did not. It appears that metabolites of arachidonic acid dependent on lipoxygenase are involved in the contractile response of the rat testicular capsule to oxytocin.

Characterization of monoclonal antibodies directed against HLA class II molecules.

Four MAbs recognizing HLA class II antigens were produced by immunization with human leukemic cells, and were characterized through immunoprecipitation and cell distribution studies. They were tested against a panel of normal and leukemic cells, Epstein-Barr virus (EBV)-transformed homozygous typing cells (HTC)s, deletion mutant cell lines, and 11 other cell lines. The immunochemical studies revealed the presence of two bands of 34 and 28 kD, corresponding to the alpha- and beta-subunits of HLA class II antigens, respectively. The cell distribution studies led to the conclusion that GRB1, GRB2, and GRB3 MAbs recognize the HLA-DR monomorphic antigens, and that the GRB4 MAb seems to recognize basically the DR molecules but shows also a cross-reaction to DP molecules.

Periodontal treatment needs in 12-16-yr-old children from Santo Domingo.

The purpose of this study was to determine the periodontal treatment needs, using the CPITN, in 12-16 yr-old children from private and public schools from Santo Domingo, Dominican Republic. A total of 1080 children was studied (540 in private schools and 540 in public schools). More children from private schools were seen without signs of periodontal disease than in public schools. More children from private schools showed no need for treatment. There was no statistical difference between children from private and public schools regarding the treatment need categories I and III. However, for the treatment need II category (oral hygiene-scaling), the children from public schools showed a slight increase in need over the private schoolchildren. Combining private and public schools, 12.9% of the 1080 children examined needed no treatment, 5.9% needed treatment I, 80.8% needed treatment II, and 0.4% needed treatment III.

Remineralization by a monofluorophosphate dentifrice in vitro of root dentin softened by artificial caries.

Artificial caries-softened dentin lesions in root surfaces were treated with 50% slurries of a monofluorophosphate (MFP) dentifrice or a placebo dentifrice twice daily for ten days. Between dentifrice treatments, the lesions were kept in a remineralizing solution. Both treatments resulted in statistically significant remineralization, with the MFP dentifrice being significantly better than the placebo. Deposition of new mineral from the MFP dentifrice occurred about equally at or within the surface zone and at the deepest part of the lesion. Mineral deposition from the placebo appeared to be more uniformly distributed within the lesion.
