Amino acid sequence of a human leukocyte interferon.

The primary structures of three major species of human leukocyte interferon differ from the structure predicted from the DNA sequence of recombinants containing leukocyte interferon-coding regions. Compared to the recombinant interferon produced in bacteria, three of the purified natural proteins isolated from leukocytes lack the 10 COOH-terminal amino acids suggested by the DNA sequence.

Production of hybridomas secreting monoclonal antibodies to the human leukocyte interferons.

Thirteen monoclonal antibodies to human leukocyte interferon have been obtained. They exhibit different patterns of binding to purified leukocyte interferon species that are consistent with the structural multiplicity of the human leukocyte interferons. These antibodies will be useful as probes into the structure of the human leukocyte interferons, for their purification, and for rapid assay of leukocyte interferon.

Closed-chest electrode-catheter technique for His bundle ablation in dogs.

A modified quadripolar electrode catheter that had two-thirds of the distal surface insulated with high-voltage plastic was inserted in 10 dogs. After a His bundle potential had been recorded, a synchronized direct-current electrical discharge was delivered between the electrodes showing the largest His bundle deflection using a standard direct-current defibrillator, and a metallic plate was positioned over the dog's back. Complete atrioventricular (AV) block was induced in 9 of 10 dogs, which were followed for 3 mo before being killed. During AV block, the QRS complex was broad and not preceded by a His bundle deflection. The mean control cycle length during AV block was 1,441 +/- 223 ms and decreased to 1,151 +/- 181 ms after exercise, a response that was usually abolished by beta-blockade. Overdrive pacing resulted in pacemaker suppression with gradual rate stabilization after 10-20 beats. There was no evidence of myocardial or valvular damage. This technique provides for a stable model of complete AV block and is suitable for experiments in which heart rate control is required. In addition, this technique may be of value for patients with tachycardia requiring His bundle section.

Effect of emotional stress on hearing.

An experimental model for emotional stress is described. Emotional stress can affect hearing if severe enough or if it lasts long enough. The noxious effect on the ear can be explained by the high level of blood catecholamines and exaggerated activity of the cochlear sympathetic innervation.

Amino-terminal amino acid sequence of human leukocyte interferon.

We report the amino-terminal sequence of the first 22 amino acids of human leukocyte interferon. These and other results indicate that human leukocyte interferon consists of many individual species. We, therefore, postulate that diversity in this protein is routinely present and that the human leukocyte interferons represent a multigene family.

Augmentation of human K-cell activity with interferon.

Our laboratory recently reported that interferon (IF) could augment human antibody-dependent cellular cytotoxicity (ADCC) and cytotoxicity by natural killer (NK) cells. Although there is general agreement that IF can boost the activity of human NK cells, two laboratories have reported a failure to boost ADCC. We have therefore examined in more detail the effects of IF on human ADCC. We have found, by several lines of evidence, that IF can indeed appreciably augment the activity of K cells, the subpopulation of lymphocytes that mediate ADCC: (1) human leucocyte IF, purified to homogeneity, boosted ADCC: (2) IF augmented ADCC against target cells that were completely resistant to NK activity; and (3) the ADCC activity of leukaemic cells from a patient with chronic lymphocytic leukaemia (CLL) which lacked any detectable NK activity was boosted by IF. These results are consistent with the hypothesis that human NK and K cells are the same subpopulation of lymphocytes and that the mechanisms regulating their activity are similar. However, the absence of NK activity in the CLL patient strongly supports the idea that not all FcR-positive cells are NK and that ADCC effectors devoid of NK may exist. However, further evidence is required to analyse this point.

Randomized double-masked trial of sodium oxyferriscorbone for the treatment of gastric ulcers.

A double-blind 4-week trial of sodium oxyferriscorbone versus placebo (distilled water) was conducted in 46 outpatients with endoscopically confirmed gastric ulcer. Ulcer healing occurred in 15 of 20 patients receiving sodium oxyferriscorbone (75%) and in 7 of 20 patients receiving placebo (35%). Patients receiving sodium oxyferriscorbone experienced less pain and required less antacid than those receiving placebo (P less than 0.05). Side effects were reported in 12 patients, 7 while receiving sodium oxyferriscorbone and 5 while receiving placebo. Six patients did not complete the study due to ulcer complications. Routine laboratory tests revealed no persistent abnormalities that could be related to the treatment. Five placebo-treated patients that were therapeutic failures were switched to sodium oxyferriscorbone and healing was observed within 3 weeks. It is concluded that sodium oxyferriscorbone is effective in enhancing healing of gastric ulcers.

Human leukocyte interferon: production, purification to homogeneity, and initial characterization.

A method of fractionating proteins by high-performance liquid partition chromatography has been developed and used for isolation and purification to homogeneity of one of the species of human leukocyte interferon. The homogeneous interferon exhibited a sharp peak on high-performance liquid chromatography and a single narrow band on sodium dodecyl sulfate/polyacrylamide gel electrophoresis in the presence of 2-mercaptoethanol. Extraction of the gel gave a single sharp peak of antiviral activity coinciding with the protein band. The specific activity of pure interferon was found to be 2--4 X 10(8) units/mg, based on amino acid analysis. The molecular weight is 17,500--18,000.

Purification to homogeneity of camel pituitary pro-opiocortin, the common precursor of opioid peptides and corticotropin.

Pro-opiocortin was purified from camel pituitaries by procedures including high-performance liquid chromatography. The precursor relationship of the pure protein to the opioid peptides and to corticotropin was confirmed. Partial chemical analysis consisting of amino acid analysis and tryptic peptide mapping was carried out with the aid of sensitive fluorescence detection.
