Biochemical and histopathologic comparison between blood and saline storage of canine veins.

Patency of vein grafts may be influenced by the medium in which they are stored temporarily. We compared saline solution vs blood on prostanoid production and maintenance of endothelium in canine veins after 1 hour of storage at 23 degrees C with 0.2 mg/ml of papaverine. Spontaneous and arachidonate-stimulated prostaglandin levels were measured by radioimmunoassay. Endothelial integrity was analyzed by light and electron microscopy. Prostaglandin production in blood vs that in saline solution was 1821 +/- 1264 and 1259 +/- 719 pg/cm2/min at control and 6705 +/- 3702 vs 6264 +/- 3409 pg/cm2/min, respectively, after stimulation. There were no statistically significant differences between the groups at any time point. Thromboxane levels were also indistinguishable between groups. Microscopy revealed 70% endothelial loss in blood vs 95% for saline solution. We conclude that endothelial preservation is enhanced by blood storage, that the medial layer produces substantial amounts of prostacyclin, and that additional storage solutions need to be investigated.

Natural and synthetic forms of insulin-like growth factor-1 (IGF-1) and the potent derivative, destripeptide IGF-1: biological activities and receptor binding.

Insulin-like growth factor-1 (IGF-1), whether recombinant, chemically-synthesised or purified from bovine colostrum, was equipotent in radioreceptor assays with IGF-1 or insulin-like growth factor-2 (IGF-2) as radioligand as well as in its ability to stimulate protein synthesis in L6 myoblasts. The N-terminal truncated, destripeptide derivative of IGF-1 was approximately 7 times more potent than IGF-1 in the protein synthesis bioassay. This increased activity occurred equally with the peptide purified from bovine colostrum as with chemically-synthesised material. The higher potency of the truncated form was not associated with an increased ability to compete for IGF-1 binding to L6 myoblasts.

Anogenital ratio: measure of fetal virilization in premature and full-term newborn infants.

To provide normative data, we measured anogenital distances in 115 infants of 25 to 42 weeks gestational age and 10 pregnant women, including anus to fourchette (AF), anus to base of the clitoris (AC), and fourchette to base of the clitoris (FC). All infant measurements showed positive and significant correlations with body surface area, weight, length, and gestational age (P less than 0.001). However, the anogenital ratio (AF/AC) followed a normal distribution and did not correlate with any of the anthropometric variables or age. Mean (+/- SD) value in infants was 0.37 +/- 0.07, and in adults, 0.36 +/- 0.07. An anogenital ratio greater than 0.50 falls outside the 95% confidence limits, suggests labioscrotal fusion, and indicates a need for further evaluation. Because it is independent of body size and gestational age, the anogenital ratio should be useful in diagnosing androgen-induced labioscrotal fusion in both premature and full-term female infants.

Intraocular large cell lymphoma presenting as massive thickening of the uveal tract.

A 46-year-old woman with a history of biopsy-proven extranodal large cell lymphoma of the maxillary sinus presented four months after this condition was diagnosed with signs of unilateral iritis. After an initial response to topical corticosteroid therapy, the iritis relapsed, the eye rapidly becoming blind and painful. Histopathological examination of the enucleated globe showed a diffuse infiltration of the entire uveal tract by malignant lymphocytes consistent with large cell lymphoma. Systemic examination revealed no concurrent foci of malignancy. This dramatic presentation of ocular involvement is quite unusual for large cell lymphoma.

Report of a panel discussion on how to run an effective Animal Care and Use Committee.

There was a consensus between the Panel and the participants in the audience as follows: All animal use should be reviewed by an Animal Care Committee. The policies and procedures of that Committee must, in the letter and in the spirit of federal guidelines, be developed by each institution, or group of institutions, mindful of its own operational practices and should apply to all research using animals, irrespective of the source of funds. The ACC will be accountable to the public at large for the conduct of animal research. To obtain the support of the community, the ACC must have the active support of the institution, both administratively and financially, and its own academic community who must be involved in ACC activities and, where possible, in the development of its policies and procedures. Education is essential. The Committee must understand its obligations and duties especially as they relate to confidentiality and internal freedom of discussion (this applies particularly to lay members). The faculty, staff, and students must understand the protective role of the Committee for both the community and animal subjects. The institutional community must be prepared to accept the responsibilities of animal use and so accept the need for education and updating in appropriate techniques and experimental design. Individual institutions and organizations, such as the Scientists Center for Animal Welfare, can and will provide information and guidance to those seeking assistance.

Consolidated Omnibus Budget Reconciliation Act of 1985.

This article briefly describes the legislative history of the old-age, survivors, and disability insurance (OASDI) and supplemental security income (SSI) provisions, as well as related Medicare and Medicaid provisions, of the Consolidated Omnibus Budget Reconciliation Act of 1985 (Public Law 99-272). It includes a chronology outlining the interaction of the budget reconciliation process with the development of the OASDI minor and technical changes bill (H.R. 2005) and the development of other social security related legislation resulting in proposals that ultimately combined in the overall Consolidated Omnibus Budget Reconciliation Act of 1985. The article also provides a detailed summary of the provisions of the legislation.

3H-dopamine clearance from the intravascular compartment of the maternal and fetal rhesus monkey.

The clearance of epinephrine and norepinephrine from the intravascular compartment has previously been demonstrated in both pregnant and nonpregnant models. With the use of the pregnant rhesus monkey, this study describes the clearance of dopamine from the fetal and maternal intravascular compartments. In both, the dopamine clearance was biphasic with an initial half-life of 1.5 minutes in fetal blood and 1.0 minute in maternal blood. These studies also confirmed that dopamine does not significantly cross the placenta in an intact form. These observations are consistent with those made for the other two major catecholamines.
