Regulated expression of repetitive sequences including the identifier sequence during myotube formation in culture.

We have isolated and characterized a cDNA of 1183 bp, pL6-411, from rat L6 muscle cells. This cDNA contains repetitive sequences - including two inverted copies of the previously described identifier sequence - as shown by sequence analysis. Repetitive sequences from pL6-411 characterize a family of RNAs which is specifically induced during L6 myotube formation. Another part of the pL6-411 sequence, existing at low-copy number per haploid rat genome, hybridized to two RNAs of 5 kb and 2 kb from L6 myoblasts as well as from L6 myotubes. A third pL6-411-related RNA of 150 bases was detected which hybridized with the repetitive sequence but did not hybridize with the low-copy number part of pL6-411. It appears that the 'identifier' sequence in this population of small RNAs is complementary to one of the 'identifier' copies in the pL6-411-related RNA. Finally, we identified on cDNA pL6-411 the recognition site for the TGGCA-binding protein and in both orientations a total of four putative promoters for RNA polymerase III.

Isolation and partial characterization of the most immunologically reactive antigen from Mycobacterium tuberculosis H37Ra culture filtrate.

The culture filtrate of Mycobacterium tuberculosis H37Ra was fractionated by ammonium sulphate precipitation, diethyl aminoethyl (DEAE) cellulose-Sephadex A-50 column chromatography, preparative isoelectric focusing and polyacrylamide gel electrophoresis to isolate the most antigenic tuberculoprotein. One protein, designated P2, was isolated and determined by immunodiffusion to be the most immunologically reactive antigen present in H37Ra culture filtrate. This protein had a molecular weight of about 22 000, a sedimentation constant of 2.2S and was present in Seibert's A, B, and C proteins.

Thermodynamic and kinetic studies of the interaction between T5-phages and their receptor, isolated from Escherichia coli b, in the absence and presence of Triton X-100.

Particles with receptor activity for T5-phages were isolated from the outer membrane of Escherichia coli B. We describe the interaction of these particles with T5-phages as a two-step chemical reaction. The rate constants were estimated from the inactivation kinetics. The transition-state theory permits the calculation of the entropy, enthalpy, and Gibbs free energy of activation. In the absence of Triton X-100, the reaction can be described with one set of thermodynamic constants for the temperature range from 10 degrees to 40 degrees C. The addition of Triton, which results in the splitting of receptor particles and in the building of mixed micelles, causes a complicated dependence on temperature. In this case, a subdividing of the temperature range measured into two parts yields two sets of thermodynamic constants that permit a good description of experimental kinetics.

Proteolysis and recycling of Mycobacterium tuberculosis culture filtrate tuberculoproteins.

The culture filtrate of Mycobacterium tuberculosis H37Ra has proteolytic enzyme activity which can hydrolyse extracellular tuberculoproteins. Some of the released products are transported into the cells and are incorporated into cell protein, and finally reappear in the culture filtrate as constituents of secondary tuberculoproteins. Continued extracellular proteolysis and utilization of the resultant products could produce a cyclic source of culture filtrate tuberculoproteins.

Paraganglioneuroma of the duodenum. Report of a case with radiographic findings, angiographic findings and a review of the literature.

A 59-year old Cuban male had seven episodes of gastrointestinal bleeding. Angiographic and radiographic studies revealed a polypoid mass in the second portion of the duodenum. The mass proved to be paraganglioneuroma. A review of the literature reveals only 25 previously recorded such cases. Eighty-eight per cent of the lesions were located in the second part of the duodenum: 84% of the patients were symptomatic. The two main symptoms noted were abdominal discomfort (45.5%) and bleeding (47.6%). None of the lesions was a malignant or functional tumor.

Further characterization of Mycobacterium ulcerans toxin.

Mycobacterium ulcerans produces an exotoxin in culture which, when inoculated into guinea pig skin, causes inflammation, necrosis, edema, and other histopathological changes resembling those in infections of humans. The toxin was resistant to heat and to alkalies and was moderately acid labile. Toxic activity was destroyed by Pronase, phospholipase, lipase, amylase, and glucosidase but not by trypsin, collagenase, cellulase, lysozyme, hyaluronidase, or neuraminidase. Toxic activity was resistant to treatment with 2-mercaptoethanol, urea, guanidine hydrochloride, p-chloromercuribenzoate, ethylenediaminetetraacetate, and sodium deoxycholate but was destroyed by sodium m-periodate and sodium dodecyl sulfate. The toxin was precipitated by a wide range of ammonium sulfate concentrations. Extraction with chlorofrom-methanol or petroleum ether destroyed its activity. Isopycnic density gradient ultracentrifugation in KBr produced a high-density lipoprotein layer with a 24-fold increase in specific activity. The results indicate that this toxin is a high-molecular-weight phospholipoprotein-polysaccharide complex.

[Acute stomach lesions and changes in histamine level of various organs in immobilization stress].

In the glandular part of the stomach, the histamine content was significantly increased during stress by immobilization. Following 8 hours of restraint these alterations were observed only in lung tissue, but not in other organs investigated. Although the maximum of stress ulcer formation did not correspond with the maximum increase in gastric histamine content as far as the time course is concerned, a role of histamine in stress ulcer disease cannot be excluded.

Diagnostic criteria for computer-aided electrocardiographic 15-lead system. Evaluation using 12 leads and Frank orthogonal leads with vector display.

The criteria for the diagnosis of myocardial infarction and ischaemic heart disease by an automated 15-lead computer-aided electrocardiographic system were examined using electrocardiograms of 543 patients. Errors in the electrocardiographic diagnosis were evaluated for each lead system (Frank orthogonal 3-lead, 12-lead, and hybrid 15-lead) using clinical and catheterization data for definitive diagnosis before review of the electrocardiograms and their reports. The effects of combinations of these diagnoses and additional ventricular conduction defects were also studied. Myocardial infarction and left ventricular hypertrophy were more reliably diagnosed using 3-lead and 12-lead systems together than with either system alone. The most sensitive criteria for anterior infarction were a Q/R ratio in Z less than 0-1 and loss of the first 20 ms of anterior forces in the horizontal and sagittal planes of the vectorcardiogram. However, false positive results were frequent, particularly in association with left ventricular hypertrophy, non-specific intraventricular conduction defects, and left bundle branch system block. Our V lead criteria were more specific whether or not these associated conditions were present. No single criterion with an acceptable false positive rate could be found to be sensitive for inferior infarction in all situations. Our most sensitive criteria were those based on the limb leads, and the presence of superior forces for the first 30 ms in the frontal plane of the vectorcardiogram, but these were better in combination. Limb lead criteria were the most specific. False positive results for inferior infarction were more frequent in the presence of left ventricular hypertrophy or ventricular conduction defects other than left anterior hemiblock. ST and T wave abnormalities were more apparent in the 12 leads than in the orthogonal leads. Specificity and sensitivity of criteria were poor, and specificity was decreased and sensitivity was not significantly improved by combining 3-lead with 12-lead criteria. Because of frequent measurement errors of ST, T, and also Q waves by the computer programme, in practice we have achieved increased sensitivity in the diagnosis of ischaemia and infarction with the combination of 3-lead and 12-lead systems. It is concluded that errors of diagnosis by a computer-aided system can be reduced by using multiple leads and that both 12-lead and orthogonal 3-lead systems are necessary for optimal computer diagnosis of left ventricular hypertrophy, myocardial infarction, and ischaemia.

[The course of zonular fiber. A further electronenmicroscopic study (author's transl)].

Zonular fibers could be shown between the basement membrane of the pigmented epithelium of pars plana and the elastic tissue of Bruch's membrane. A connection of zonular fibrils between the unpigmented epithelium and the basement membrane of pigmented epithelium of the pars plana could not be observed. It is possible to discuss two independent courses of zonular fibers.
