Synthesis and antiinflammatory activity of indole carboxylic acids and esters.

1-Phenylalkylindole-3-carboxylic acids 1-4, indole-1-acetic acids/esters 5-10 and the hydrazones 11-15 were prepared and submitted to the rat paw edema test using carrageenin. In the first two groups of compounds, the 2-chloro indoles were more active than the corresponding indole derivatives. In the third group the activity seemed to be determined largely by the substituent at the 1-position.

Human HOX genes are differentially activated by retinoic acid in embryonal carcinoma cells according to their position within the four loci.

We studied the expression of 33 human homeobox genes belonging to four complex HOX loci in embryonal carcinoma NT2/D1 cells. These cells can be induced to differentiate by culturing them in media containing retinoic acid. Northern blot analysis reveals that no expression of these genes was detectable in NT2/D1 stem cells, whereas 22 HOX genes are well expressed in NT2/D1 cells treated with 10 microM retinoic acid for 14 days. The 11 HOX genes the expression of which remained undetectable in NT2/D1 cells after this treatment are located at the 5' end of their loci: four in HOX1, five in HOX3 and two in HOX4. The boundary between induced and silent genes roughly corresponds to the HOX genes constituting the homology group 5, related to the Abdominal-B homeotic gene of Drosophila. All nine identified HOX2 genes are well expressed in fully induced NT2/D1 cells and none of them maps 5' genes of this homology group. We conclude that HOX genes are differentially activated by retinoic acid in these cells according to their physical location within the four chromosomal loci.

Detection of microorganisms in clinical specimens using slides prestained with acridine orange (AOS).

Slides prestained with acridine orange (AOS) were used for direct examination of microorganisms in clinical specimens. The method is simpler than a commonly used acridine orange stain (AO) and has the advantage of being a supravital stain. Results indicate that the AOS method is sensitive, and allows rapid detection of microorganisms, even in the specimens containing few microorganisms, e.g. cerebrospinal fluid. Moreover AOS may be the choice method for detection of Trichomonas vaginalis.

A simple and rapid method to evaluate lymphocyte response to mitogens.

A new method to evaluate lymphocyte response to mitogens is proposed. The method is based on the observation in fluorescence of the stimulated cells after their supravital staining on slides prestained with acridine orange in alcoholic solution. The results are expressed as Blast Index (IB). The method is very practical, easy to reproduce and shows a good correlation with 3H-thymidine uptake.

Importance of showing HBsAg and HBcAg positivity in the liver for better aetiological definition of chronic liver disease.

Serum and tissue hepatitis B virus (HBV) markers were compared in 35 alcoholic and 23 non-alcoholic subjects affected by chronic liver disease. Seventeen point one per cent of alcoholic and 21.7% of non-alcoholic subjects had HBV tissue markers, but not serum markers, for this virus. It is therefore concluded that showing the presence of HBV tissue markers permits a better aetiological definition of hepatitis B surface antigen (HBsAg) negative chronic liver disease, both in alcoholic and non-alcoholic subjects.

Hyperthyroidism and Addison's disease in a patient with myotonic dystrophy.

A 53-year-old man had myotonic dystrophy, hyperthyroidism, and Addison's disease, an association not previously reported, to our knowledge. In the literature, at least five cases of hyperthyroidism associated with myotonic dystrophy have been described, but none also had Addison's disease. The presence of thyroid anti-microsomal antibodies and anti-adrenal antibodies suggests that the two endocrine disorders may be autoimmune. In our case, the treatment of the two endocrinopathies caused a reduction of myotonic symptoms.

Potential antitumor agents X: Synthesis and antitumor activity of two nitrogen mustard derivatives related to ketocaine.

The synthesis of two nitrogen mustard derivatives (VIII and IX) related to the well-known local anesthetic ketocaine (III) is reported. These compounds were tested in mice implanted with Ehrlich ascites tumor cells, and the antitumor activity was compared with that of two previously synthesized analogues (I and II) lacking the nitro group and with that of doxorubicin. The monofunctional compound IX was inactive, but the bifunctional compound VIII showed potent antitumor activity (%T/C greater than 254 at 20 mg/kg).
