Effect of inoculation of Zea mays with Azospirillum brasilense strains under temperate conditions.

Seven strains of Azospirillum brasilense were compared for their effect on the growth of Zea mays grown under temperate conditions in sand--vermiculite pot cultures. Inoculation with all seven strains tested, including Fix(-) mutant strains, increased dry weight and total nitrogen content of shoots, but nitrogen concentrations were unaffected. Low and variable rates of acetylene reduction activity were observed from excised roots of inoculated plants without preincubation. Estimates of N2-fixing A. brasilense associated with inoculated roots showed differences between strains in establishing themselves in the rhizosphere and endorhizosphere. In some strains enrichment in the endorhizosphere of roots occurred following inoculation, but the relative numbers and location of the strains did not appear to affect the yield response.

Effects of pH, Ca2+, temperature, and protease pretreatment on interkingdom fusion.

The incubation of carrot protoplasts and cultured Xenopus cells in a protease solution has been shown to enhance their subsequent interkingdom fusion by a high pH/high Ca2+ method. The effects of Ca2+ concentration, pH, and temperature on the frequency of heterokaryon formation have also been studied. Potentially viable heterokaryons have been repeatedly produced at high frequencies (consistently greater than 10%), far exceeding those so far achieved in PEG-mediated fusion. Cell aggregates are readily dispersed after this method of fusion, permitting the accurate estimation of fusion frequencies.

Effect of Erythrogen on post-thaw recovery of cryopreserved cell suspensions of indica rice (Oryza sativa L.).

This study shows that adding haemoglobin solution (Erythrogen) to post-thaw medium of Indica rice (Oryza sativa L.) cells enhances survival following cryopreservation. Haemoglobin (1:50 - 1:200 v:v) had a beneficial effect on post-thaw viability and subsequent cell growth. A key finding was that the successful recovery from cryopreservation of cell suspensions of the Indica rice cvs. BR26 and Pajam, and their re-establishment in AA2 medium, reflected a requirement for such supplementation of the post-thaw recovery medium with Erythrogen.
