HLA and obesity.

A possible association between HLA and obesity was investigated in 42 unrelated obese children from Hessen, Germany. A family study was possible in 11 families with a total of 26 children. No significant deviation was observed for one of the HLA ABC antigen frequencies in the sample group compared with controls (n = 220). The linkage analysis in families: a) ruled out a close linkage (less than 10 cM) between HLA and a hypothetical obesity gene (assumption of a dominant mode of inheritance), b) did not exclude recessive inheritance so far, since the importance of penetrance is unknown and a simple gene model is improbable, c) yielded also negative lod scores (which did not reach the elimination level of -2) for an intermediary genetic model.

Pigeon breeder's lung: association with HLA-DR 3.

52 symptomatic (SPB) and 64 asymptomatic (APB) pigeon breeders were investigated for the HLA-A, -B, -C, -DR; C2, C3, C4 and Bf systems, and C3, C4 and factor B serum concentrations. HLA-DR3 and Bf S frequencies were significantly higher in the SPB than the APB group. Mean factor B concentrations were lower in the SPB than in the APB group. A positive two-way association between HLA-DR3 and the disease was found. It is concluded that a gene or genes responsible for type III and IV allergic reactions leading to the disease is associated with HLA-DR3. The increase of Bf S allotype and low mean factor B concentrations, however, can be explained by the strong linkage disequilibrium between BfS and HLA-DR3.

IgG and IgA heavy chain allotypes in Type 1 diabetes.

IgG and IgA heavy chain allotypes were determined in the sera of 483 Caucasian Type 1 diabetes patients and 503 Caucasian healthy controls. There was no significant difference between patients and controls neither on the level of Gm phenotype frequencies nor on the level of Gm three-locus and two-locus haplotype frequencies. A selective IgA deficiency was found in 14 patients (2.9%) but in none of the control individuals (P less than 10(-4].

Concentrations of placental proteins (HPL and SP1) in maternal serum throughout normal pregnancy.

Throughout 65 normal singleton pregnancies 332 blood samples were obtained and analyzed for pregnancy-specific beta 1 glycoprotein (SP1) and human placental lactogen (HPL). The measurement of SP1 in maternal serum was made using radial immunodiffusion, that of HPL by using radioimmunoassay. There was wide variation in the number and timing of blood samples obtained from patients, and therefore the results could not be used to construct a regression curve by the usual methods applied to independent measurements. Consequently only one value per patient was chosen at random as a basis for calculations. A second degree polynomial for the logarithm of the serum concentrations yielded an optimal fit for both parameters. Tolerance intervals of 90% were calculated for use in future studies.

Immunogenetic studies of the platelet-specific antigen P1A1 (Zw(a)).

The phenotype frequency of the platelet-specific antigens P1A1/A2 ( = Zw(a,b)) in the German population (N = 711) is 97.75% and 2.25% respectively. The corresponding gene frequencies are P1A1 0.85; P1A2 0.15. The P1A1 determinant is codominantly inherited (19 families; 151 family members). There is a strong gene dosage effect between P1A1 homo and heterozygous individuals. No definite linkage of the locus P1A1 versus the loci HLA-ABC, ABO, or Rh was found.

Population, formal genetics, and linkage relations of the phosphoglycolate phosphatase (PGP)--E.C.3.1.3.18.

One hundred and sixty-seven blood donors, 26 families with 72 offspring and 12 motherchild couples were studied for the phosphoglycolate phosphatase polymorphism. In hemolysates, the isozymes are stable for at least five weeks. The distribution of observed phenotypes in the population study did not diverge from the expected values according to Hardy-Weinberg law. In the family study, the formal genetic model of three alleles--PGP1, PGP2 and PGP3 at one autosomal locus-could be confirmed. Among 33 individuals from a Mongoloid population PGP1 was observed in 100%. This observation lead us to the conclusion, based also on recent data in Negroid populations (Barker and Hopkinson 1978), that phosphoglycolate phosphatase may be a more recent polymorphism of Caucasoid populations. Linkage studies with the hp alpha locus an chromosome 16 resulted in 19 meiotic divisions of 4 informative families in a lod score peak of 0.23 at theta 0.25 being inconclusive. The inclusion of the PGP system in paternity testing is also discussed.

Age-related association of insulin-dependent diabetes mellitus with BfF1 and the HLA-B18, BfF1 haplotype.

Five hundred and ninety-nine patients with insulin-dependent (Type 1) diabetes mellitus were typed for Bf (factor B) polymorphism, and 318 of them for HLA-A, B and C antigens. Bf and HLA antigen frequencies were compared with those in 536 normal controls. A significant positive association between Type 1 diabetes and the rare factor B variant BfF1 was found (p less than 10(-3)), but this was present only in patients aged under 16 years at onset of the disease (p less than 0.005). There was a strongly positive linkage disequilibrium between BfF1 and HLA-B18 in the diabetic patients. This, too was especially pronounced in the juvenile onset cases, in whom it was significantly stronger than in controls (p less than 10(-3)). The known positive associations between Type 1 diabetes and HLA-B8, -B15 and -Cw3 were confirmed.

HLA-A, B, Bf three point association of 1,072 haplotypes in a German population.

Altogether 1,072 HLA-A, B, Bf haplotypes in 536 parents of 268 German families were analyzed by gene and haplotype counting in order to investigate the two and three point association of HLA-A and B antigens with Bf alleles. In agreement with previously published data (Albert et al. 1975, Bender et al. 1977) significant positive linkage disequilibria were found for HLA-A3 and BfF, B7 and BfS, B8 and BfS, Bw35 and BfF, whereas significant negative Delta values appeared to exist for HLA-A3 and BfS, B7 and BfF, B8 and BfF, B12 and BfS, as well as Bw35 and BfS. Significant positive three point Delta values were found for the following haplotypes: HLA-A11, Bw35, BfS, HLA-Aw23, Bw35, BfS, HLA-A28, B12, BfS, HLA-A2, B17, BF, HLA-A3, Bw35, BfF, HLA-Aw24, b12, BfF and HLA-A29, B12, BfF. According to the large number of comparisons performed calculating the level of significance of the three point associations, these data have to be corroborated by further investigation. The three point Delta values, obtained by calculation from the patients' phenotypes differed markedly from those results obtained by haplotype counting.

[Pilocarpin eye drops with a novel Pilocarpin polymeric salt (author's transl)].

A new pilocarpin salt was obtained by chemically linking pilocarpin to a polymere substance. By galenic processing to the respective emulsion, eye drops containing this pilocarpin salt were produced, which enable a retarded pilocarpin delivery in vitro. With respect to glaucoma therapy, clinical tests showed that frequency of application can considerably be reduced. In the present investigations on a collective of 30 normal volunteers the effect of a commercial aqueous pilocarpin solution on the IOP, pupillary diameter and accommodation was compared with that of the novel pilocarpin drops.
