[Pharmacological studies of FUT-175, nafamstat mesilate. IV. Effects on coagulation, platelets and fibrinolysis].

Inhibitory effects of FUT-175 (nafamstat mesilate) on coagulation, platelets and fibrinolysis were examined. FUT-175 prolonged activated partial thromboplastin time, thrombin time and prothrombin time in rabbit plasma. FUT-175 prolonged these coagulation times in human plasma at lower concentration than in rabbit plasma. FUT-175 inhibited platelet aggregation induced by a variety of aggregation agents in rabbit platelet-rich plasma (PRP). In human PRP, FUT-175 inhibited platelet aggregation induced by a variety of aggregation agents at lower concentration than in rabbit PRP. Lipopolysaccharide induced a dose-dependent platelet aggregation in dog PRP. FUT-175 showed an inhibitory effect on this aggregation. FUT-175 inhibited clot retraction in rabbit plasma. The fibrinolysis activity was measured on fibrinolysis of rabbit plasma activated by urokinase. FUT-175 prolonged this fibrinolysis time. Inhibitory effects on coagulation and fibrinolysis were also found ex vivo. FUT-175 prolonged bleeding time in mice. These results indicate that FUT-175 has potent inhibitory effects on coagulation, platelets and fibrinolysis.

[Effect of catecholamine on reperfused myocardium following hypothermic global ischemia].

This study was aimed to evaluate the effect of catecholamine on the myocardium reperfused after hypothermic global ischemia, by changes of hemodynamics, biochemistry and ultrastructure. Under cardiopulmonary bypass (CPB) at flow rate 80 ml/kg/min., the aorta was clamped for 60 min. at 28 degrees C of myocardial temperature and reperfused for 60 min. in 26 mongrel dogs. They were divided into 4 groups by infusion of physical saline solution (control), epinephrine 1 microgram/kg/min. (group 1), epinephrine 0.1 microgram/kg/min. (group 2) and dobutamine 5 micrograms/kg/min. (group 3) during reperfusion. The hemodynamic parameters and myocardial isoenzyme (m-AST, MB-CPK) of coronary sinus venous blood were measured before CPB, 30 and 60 min. after declamp. The myocardial adenosine triphosphate (ATP), creatine phosphate (CP), water content, tissue Ca content and fine structure with score of mitochondrial membrane and cristae were examined in epicardium and endocardium at the end of experiment. Hemodynamic parameters after declamp were higher in group 1, 2 and 3 than control (p less than 0.05). The water content and tissue Ca content in group 1 were higher than control. The ATP of endocardium was lowest but CP was no significant difference among four groups. The mitochondrial score in group 1 was lower than control. These data suggest that epinephrine and dobutamine increase hemodynamics and tissue Ca content on the reperfused myocardium following 60 min. of hypothermic global ischemia, but they do not improve depletion of ATP and disruption of myocardial ultrastructure. High dose of epinephrine accentuates ischemic damage of reperfused myocardium after hypothermic global ischemia.

A new class of nitrosoureas. 4. Synthesis and antitumor activity of disaccharide derivatives of 3,3-disubstituted 1-(2-chloroethyl)-1-nitrosoureas.

A series of 33 N-(2-chloroethyl)-N-nitrosocarbamoyl derivatives of N-substituted glycosylamines has been prepared and tested for antitumor activities. The compounds were obtained by reaction of glycosylamines with isocyanate, followed by nitrosation with N2O4. Structure-activity relationships of these trisubstituted nitrosoureas were investigated by varying the N-substituents and disaccharide groups and by comparing them with the corresponding disubstituted analogues. A large number of the nitrosoureas bearing a maltosyl group exhibited strong antitumor activities against leukemia L1210 and Ehrlich ascites carcinoma, and 60-day survivors against leukemia L1210 were found at the optimal dose for these derivatives. In contrast, the lactosyl and the melibiosyl derivatives were almost inactive. The most interesting compound in this series, the 3-isobutyl-3-maltosyl derivative (37), was tested against leukemia L1210 by single and multiple treatment. Its therapeutic ratio (96.3) obtained by multiple treatment is 3 times larger than that (31.5) obtained by single treatment, suggesting a possible clinical utility of 37 by multiple treatment. The favorable effect of a maltosyl moiety in this class of compounds is discussed.
