Identification of fetal blood stains by radioimmunoassay of alpha 1-fetoprotein.

Radioimmunoassay of alpha 1-fetoprotein(AFP) for medico-legal identification of fetal blood stains using a commercial kit is described. The AFP content in fetal blood stains on filter paper ranged from 21--320 ng/9 mm2. The protein was detected in stains of adult blood and retroplacental blood in only negligible amounts. Aging of the blood stains did not influence the values up to 1 month. The method is simple and sensitive enough for application to medico-legal-practice.

Potentiating mechanism of bradykinin action on smooth muscle by sulfhydryl compounds.

(4R)-3-[(2S)-3-Mercapto-2-methylpropanoyl]-4-thiazolidinecarboxylic acid (YS-980), a potent angiotensin I (AI) converting enzyme inhibitor, inhibited the contractile response of isolated guinea-pig ileum to AI but not to AII, while it potentiated the response to bradykinin. Other sulfhydryl compuonds also produced inhibition of AI action, and the effects were closely correlated with their potentiating activities against bradykinin action. These results suggest that it is mainly inhibition of the enzyme in the tissue which participates in the mechanism of potentiation of kinin action by YS-980.

Multiplicity of monoamine oxidase in chick brain.

The substrate- and inhibitor-related characteristics of monoamine oxidase (MAO) were studied on chick brain mitochondria. It was found that neither 5-hydroxytryptamine nor beta-phenylethylamine is the specific substrate for type A and type B MAO in chick brain.

A comparison of angiotensinase and placental leucine aminopeptidase during normal pregnancy.

1. Serum leucine aminopeptidase activity measured by its action on hydrolysis of L-leucyl-beta-naphthylamide was increased progressively as pregnancy advanced. Angiotensinase activity determined by bioasaay showed parallel increase during normal pregnancy. 2. The effects of heat treatment and L-methionine on the activity of angiotensinase were compared with those on leucine aminopeptidase activity in pregnancy serum.

The origin of human seminal leucine aminopeptidase (LAP).

Examination of the distribution of leucine aminopeptidase (LAP) activity in the male reproductive system showed the existence of the largest amounts of this enzyme in the prostate. Precipitin electrophoresis revealed that seminal LAP is immunologically identical with the prostatic enzyme and distinct from the enzymes in the other male reproductive organs. It was concluded that the origin of seminal LAP is the prostate, by which the enzyme is secreted into the seminal plasma.

Multiple forms of monoamine oxidase in developing chick brain.

The developmental change in the multiplicity of monoamine oxidase (MAO) forms of chick brain was studied with regard to substrate specificity and inhibitor sensitivity. The developmental pattern of MAO activity towards beta-phenylethylamine was generally similar to that of MAO activity towards 5-hydroxytryptamine. The inhibition study showed that the ratio of type A MAO to type B MAO in chick brain increased during development. This is quite different from results obtained in mice, rats, guinea pigs, and humans, and is not consistent with the idea that in immature brain type A MAO predominates.

Placental and non-placental leucine aminopeptidases during normal pregnancy.

1. A simple and rapid method discriminating between placental and non-placental leucine aminopeptidases (LAP) existing in pregnancy serum is described, which includes technics of heat inactivation at 60 degrees for 30 minutes and inhibition by 0.02 M L-methionine. 2. Serial determinations of serum total, heat-stable and L-methionine-insensitive LAP activities during normal pregnancy were carried out. 3. The results were ascertained by disc electrophoresis and it was found that the heat-stable enzyme corresponds to normal serum LAP; the L-methionine-insensitive enzyme to pregnancy serum LAP of placental origin; the sum of these two to total serum LAP.
