Classification and Structural Comparison of Full-Length cDNAs for Pathogenesis-Related Proteins.

Fourteen cDNA clones of pathogenesis-related (PR) proteins, PR1a and PR1b of tobacco were obtained and classified into six groups based on restriction enzyme maps. To assign the groups to different classes of PR1 proteins, all the clones were partially sequenced and compared with amino acid sequences of PR1a and PR1b. Two groups of these corresponded to PR1a and four to PR1b. The results indicate that there are at least two kinds of PR1a mRNAs and four kinds of PR1b mRNAs. In fact, one cDNA insert hybridized to at least six to seven DNA fragments in restriction enzyme fragments of Samsun NN genomic DNA, indicating that the PR1 protein genes exist as a multigene family in the tobacco genome. Two sequences of essentially full-length cDNAs for PR1a and PR1b were determined and compared. The coding sequences of two cDNAs share 93% homology and the deduced amino acid sequences of PR1a and PR1b precursors, which are synthesized as larger precursors containing signal peptides, are 91% homologous. The homology of mature PR1a and PR1b regions is higher than that of larger precursors, 94% in the nucleotide sequence and 93% in the amino acid sequence, whereas that of the signal peptide regions is 80 and 90%, respectively. The hydropathy patterns and the secondary structures predicted by Chou-Fasman rules are similar to tomato PR protein in the half-side of the C terminus, which suggests that the half-C terminus side is important for the function of PR1 proteins.

Nephritogenic glycoprotein. XII. Focal glomerulonephritis with myeloid bodies produced in rats by synthetic analogues of the glycopeptide, nephritogenoside.

Glomerular lesions were successfully induced in rats by a single footpad injection of synthetic analogues of nephritogenoside [alpha-Glc-(1----6)-alpha-Glc-(1-NHCO-Asn, and -Gln)]. Characteristic morphological changes of these glomerular lesions are (1) focal glomerulonephritis (FGN) and (2) the appearance of myeloid bodies in the epithelial cells of the podocytes. FGN produced by a single injection of the synthetic analogues of nephritogenoside may provide an experimental animal model of this potentially diffuse disease, since morphological changes in the early stage of sclerosing glomerulonephritis (end-stage kidney) induced by a single footpad injection of natural nephritogenoside are often identical to those of FGN.

Unsaturated fatty acids in mammalian cell strains cultured for more than 10 years in serum-free synthetic media.

In cultured cells, essential lipids are usually supplied by the serum added to the culture medium. However, in a serum-free medium, essential lipids would need to be synthesized by the cells. Cell strains derived from kidney, spleen, uterine carcinoma and fibroblasts were cultured in serum-free synthetic media for more than 10 years and their fatty acids were analyzed capillary column gas-liquid-chromatography. The following conclusions were drawn: 1. Polyunsaturated fatty acids were not detected. Thus, prostaglandins are not appear to be essential for initiation of DNA synthesis in these cells. 2. A compensatory increase of the content of monounsaturated fatty acids was detected. 3. A large difference in vaccenic/oleic acid ratio was found among several strains of cells, but the ratio does not reflect that of the original tissues.

Biochemical mechanisms of aminoglycoside cell toxicity. I. The uptake of gentamicin by cultured skin fibroblasts and the alteration of lysosomal enzyme activities.

In order to study the toxicity of aminoglycoside, human skin fibroblasts were used as a model for basic studies, since they are known to have a specific aminoglycoside-binding site and to translocate the drug into the cells. Following the exposure of fibroblasts to gentamicin for 3 days, the cells formed many osmiophilic lamellar materials (myeloid bodies) in the lysosomes, while the other cellular structures appeared to remain normal. Although gentamicin was intensively accumulated within the lysosomes, intralysosomal pH, determined by the fluorescence intensity ratio method using fluorescein-isothiocyanate-labeled dextran, did not alter. Among the lysosomal enzymes, the activities of six different glycosidases were unchanged. On the other hand, sphingomyelinase and acid lipase activities were greatly decreased, while phospholipase A activity was increased. These results indicate that the lipid metabolism of fibroblasts is altered by gentamicin treatment, and that perturbation of intralysosomal pH can not be the cause of the changes observed in cell lysosomal enzyme activities.

Erythrocyte membrane abnormalities in patients with amyotrophic chorea with acanthocytosis. Part 1. Spin labeling studies and lipid analyses.

Low fluidity was observed in the erythrocyte membranes of familial chorea-acanthocytosis by spin labeling with 12NS. The high content of the saturated fatty acids support the finding of low fluidity. Other lipid contents (cholesterol and phospholipids) and the cholesterol/phospholipid ratio were within normal ranges. The sharp decrease in the parameter, (h0/h-1)1/2, suggests phase separation deep inside the patients' erythrocyte membrane matrix. This distortion of the membrane matrix may be caused by membrane components other than the lipid components.

Erythrocyte membrane abnormalities in patients with amyotrophic chorea with acanthocythosis. Part 2. Abnormal degradation of membrane proteins.

Bands 2.1, 3 and 4.2 of the erythrocyte membrane proteins in chorea-acanthocytosis are self-digested faster than those in normal controls. The junction between spectrin and band 3 protein at the cytoplasmic side of the patients' erythrocyte membranes may have some conformational defects. The conformational defects of the proteins may be responsible for the low fluidity in the interior of the patients' erythrocyte membranes. These basic molecular defects may be widespread in other extraneural cell membranes as well as in neuronal cell membranes.

Assessment of single vessel coronary artery disease: results of exercise electrocardiography, thallium-201 myocardial perfusion imaging and radionuclide angiography.

The sensitivity of the commonly used stress tests for the diagnosis of coronary artery disease was analyzed in 46 patients with significant occlusion (greater than or equal to 70% luminal diameter obstruction) of only one major coronary artery and no prior myocardial infarction. In all patients, thallium-201 perfusion imaging (both planar and seven-pinhole tomographic) and 12 lead electrocardiography were performed during the same graded treadmill exercise test and radionuclide angiography was performed during upright bicycle exercise. Exercise rate-pressure (double) product was 22,307 +/- 6,750 on the treadmill compared with 22,995 +/- 5,622 on the bicycle (p = NS). Exercise electrocardiograms were unequivocally abnormal in 24 patients (52%). Qualitative planar thallium images were abnormal in 42 patients (91%). Quantitative analysis of the tomographic thallium images were abnormal in 41 patients (89%). An exercise ejection fraction of less than 0.56 or a new wall motion abnormality was seen in 30 patients (65%). Results were similar for the right (n = 11) and left anterior descending (n = 28) coronary arteries while all tests but the planar thallium imaging showed a lower sensitivity for isolated circumflex artery disease (n = 7). The specificity of the tests was 72, 83, 89 and 72% for electrocardiography, planar thallium imaging, tomographic thallium imaging and radionuclide angiography, respectively. The results suggest that exercise thallium-201 perfusion imaging is the most sensitive noninvasive stress test for the diagnosis of single vessel coronary artery disease.
