Effects of recombinant human granulocyte colony stimulating factor (rG-CSF) on murine myeloid leukemia: stimulation of proliferation of leukemic cells in vitro and inhibition of development of leukemia in vivo.

We have established an experimental murine myeloid leukemia model and investigated the effects of recombinant granulocyte colony stimulating factor (rG-CSF) on myeloid leukemia in vitro and in vivo. rG-CSF stimulated colony formation by the leukemic cells in semisolid agar medium, and exponential growth of the clonogenic cells in suspension medium. Thus, rG-CSF was able to stimulate both the differentiation and self-renewal processes of the leukemic stem cells in vitro. However, 14 consecutive daily injections of rG-CSF prolonged the mean survival time of the mice implanted with the leukemic cells. This effect of rG-CSF was accompanied by a delay in the emergence of the blast cells in peripheral blood and by a decreased blast population in the spleen, suggesting that development of leukemia was suppressed in the rG-CSF-treated-mice. The prolongation of the survival time by rG-CSF was more evident when rG-CSF was administered in therapeutic combination with cyclophosphamide. These results indicate that the effect of rG-CSF on the development of leukemia is not exactly predicted from in vitro experiments.

Pharmacological studies of lappaconitine. Occurrence of analgesic effect without opioid receptor.

The contribution of opioid receptor to lappaconitine (LA) induced analgesia was investigated by in vivo and in vitro methods. The antagonistic effects of naloxone (NLX) on the analgesic actions of LA (s.c.) and morphine (MOR, s.c.) were determined by the tail pressure method, the hot plate method and the acetic acid-induced writhing method in mice. The ED50 values of MOR in the absence of NLX were 3.82, 0.50 and 0.33 mg/kg, respectively, but in the presence of NLX, the analgesic effect of MOR was decreased and the ED50 values were changed to 41.6, 4.40 and 5.97 mg/kg, respectively. However, NLX did not affect the analgesic activity of LA. The ED50 values of LA in the three methods were 9.12, 3.34 and 1.70 mg/kg in the absence of NLX and 12.1, 4.19 and 2.88 mg/kg in the presence of NLX, respectively. The inhibitory effect of the LA on the electrically evoked contractions of isolated guinea-pig ileum was weaker than that of MOR, and differently from MOR, its effect was not antagonized by NLX. Additionally, LA did not abolish the electrically evoked contractions of isolated rabbit and mouse vas deferens. In conclusion, the results suggest that the opioid receptor was not involved in the analgesic activity of LA.

[Liver transplantation for biliary atresia with special reference to the Japanese children who received liver transplantation in foreign countries].

Seven cases out of 8 Japanese children who received orthotopic liver transplantation (OLT) in foreign countries because of biliary atresia as of Oct 1988 are reported. Their clinical courses after OLT have been good. Despite being satisfied with the results, their parents and family feel anxiety such as long term postoperative care and side effects of immunosuppressive therapy. Cost and the handling of rejection or retransplantation were pointed out as the greatest problems related to receiving OLT in foreign countries.

[Chest X-ray findings in infantile Chlamydia trachomatis pneumonia; report of two cases].

Chest X-ray findings in two infants with Chlamydia trachomatis pneumonia were reported. Both cases showed bilateral interstitial infiltrates and hyperinflation. These findings are not specific to this pneumonia. Chlamydia trachomatis pneumonia should be considered as one possibility when we find such findings on chest X-ray in infants.

[Investigation of intracavernous injection of papaverine for erectile impotence].

Forty milligrams papaverine HCl was intracavernously injected to 28 patients with erectile dysfunction for diagnosis and treatment in 14 cases in which it was injected a few times. We designed a brief manual method for measurement of penile hardness by artificial erection. Twenty of the patients corresponding to 71.4%, reacted, but only 16 (57.1%) had efficient erection for possibility of coitus. It was useful for the diagnosis of vascular dysfunction with dorsal penile arterial pulse sound examination in impotence. It was useful for the cases of small vascular impediment with preservation of nerve supply after the operation of intrapelvic malignancies, and self injection might be possible, but on the other hand, it had no effect for the patients of advanced age with vascular impediment and diabetes mellitus neuropathy, and with arteriosclerosis.

Multiple rheumatoid bursal cysts.

We have encountered a patient with rheumatoid arthritis (RA) who developed multiple subcutaneous swellings containing sterile milky fluid. The resected material showed chronic granulomatous changes compatible with rheumatoid inflammation. We suggest that multiple chronic rheumatoid inflammation may occur in the bursae as well as joint synovium as one of the extraarticular manifestations of RA.

[A digital filtering system for extracting crackles from lung sounds].

A nonlinear digital filter system is proposed for automatic extraction of crackles which are discontinuous adventitious sounds in lung sounds. This system is composed of two nonlinear digital filters; one is a stationary nonstationary separating filter, and the other is a width-discriminating filter. The former separates nonstationary signals from stationary ones in the lung sounds, using the prediction error to the input lung sound signal. If the prediction error is small enough, the lung sound is considered to be stationary, but if the error is large, a nonstationary signal is considered to occur and the nonstationary part is separated. The latter, the width-discriminating filter, performs signal extraction by considering the signal wave form of the crackles, simply realized by logical algebra. This filter extracts an impulsive signal, which is a small-width wave, and its succeeding waves; such wave form is typical of that of crackles. First, crackles are roughly separated by the stationary-nonstationary separating filter as nonstationary signals, and then are more precisely extracted from the nonstationary output by the width-discriminating filter. Some examples of processing actual lung sound data by this system show its high performance.

Rotational stability of diaphyseal fractures of the radius and ulna fixed with Rush pins and/or fracture bracing.

The rotational stability of fractures of the radius and ulna treated with Rush pins and/or fracture bracing was studied in six fresh cadaver forearms. Forearm rotation and fracture site motion (rotation) were measured as functions of applied forearm torque and rotation (pronation-supination). Values were obtained from the forearm: (1) intact and with both bones fractured; (2) without fixation; (3) with a fracture brace; (4) with Rush pins; and (5) with a combination of Rush pins and a fracture brace. A brace was ineffectual in reducing fracture site motion when the wrist was rotated to specified angles compared to fracture site motion for the forearm with no fixation treatment rotated to the same angles. When loading to specific torque levels, however, the brace reduced fracture site motion to one-half the motion with no fixation treatment. Under both loading conditions, Rush pin fixation significantly and markedly reduced the fracture site motion (to approximately one-eighth of the motion with no fixation treatment), whereas a brace in conjunction with Rush pins did not significantly further reduce the fracture site motion. The radius showed more motion at fracture site than the ulna.

A new class mutation of low density lipoprotein receptor with altered carbohydrate chains.

In a monensin-resistant mutant (Monr-31) of Chinese hamster ovary cells, the O-linked sugar chains of the low density lipoprotein (LDL) receptor are altered, suggesting a mutation at a Golgi apparatus gene. In a compactin-resistant mutant (MF-2) of Chinese hamster V79 cells, the mature LDL receptor is apparently 5000 daltons smaller; the difference is due to altered glycosylation of O-linked sugar chains. Hybrids between MF-2 and Monr-31 still produced LDL receptor molecules with aberrant sugar chains; thus both mutants are in the same complementation group. Krieger and his colleagues (Krieger, M., Kingsley, D., Sege, R., Hobbie, L., and Kozarsky, K. (1985) Trends. Biochem. Sci. 10, 447-452) have classified Chinese hamster ovary cell mutants with altered LDL receptor structure into four groups: ldlA, ldlB, ldlC, and ldlD. Cell-cell hybrids between their ldl mutants and Monr-31 produced wild type mature LDL receptors with normal molecular sizes, suggesting that these compactin- and monensin-resistant mutants define a new class of LDL receptor mutant. Since both of our mutants are defective in internalization of LDL, we assign them as int mutants. This may imply a further etiology for hypercholesterolemia, and cases can now be examined for such a class.

Structures of two kinds of mRNA encoding the chum salmon melanin-concentrating hormone.

The structures of two kinds of melanin-concentrating hormone (MCH) cDNA clones isolated from a chum salmon hypothalamus cDNA library were described. The MCH heptadecapeptide was present at the C terminus of a putative MCH precursor consisting of 132 amino acid residues. The two clones were 80% homologous with each other at the amino acid sequence level. Two genes, each directing one of the mRNAs was noted at about a single copy per haploid salmon genome. MCH genes were efficiently expressed as 0.9-kb poly(A)+RNA in salmon hypothalamus, and sequences hybridizable with salmon MCH cDNA were found in rat hypothalamus.

Phosphorylation of beta-glucuronidases from human normal liver and hepatoma by cAMP-dependent protein kinase.

beta-Glucuronidases purified from human hepatoma and from normal liver could serve as a substrate for a cAMP-dependent protein kinase. The rate of phosphorylation reaction of the hepatoma beta-glucuronidase was rapid, whereas that of the normal liver beta-glucuronidase was slow and much lower. Stoichiometry of phosphorylation was 4.3 and 0.46 mol of phosphate/mol of the beta-glucuronidase from the hepatoma and normal liver, respectively. Tryptic peptide mapping of 32P-labeled beta-glucuronidase from hepatoma identified two distinct phosphopeptides (X and Y). The peptide from hepatoma hydrolase was phosphorylated predominantly at the X, while the peptide Y was the major phosphopeptide in the hydrolase of normal liver. Analysis of phosphoamino acids revealed two sites, phosphoserine and phosphothreonine. beta-Glucuronidase from hepatoma consisted of a major subunit with molecular mass of 64,000 (64 kDa) and a minor subunit with 76 kDa, whereas the hydrolase from normal liver had almost exclusively 64 kDa subunit. 32P-labeled beta-glucuronidase indicated that the 64 kDa subunit was phosphorylated both in hepatoma and normal liver beta-glucuronidases.

Developmentally regulated expression of the calcitonin gene related peptide (CGRP) in rat lung endocrine cells.

Calcitonin (CT) and the calcitonin gene-related peptide (CGRP) are generated by alternative RNA processing from a single CT/CGRP gene. Recently, we reported the existence of CGRP-immunoreactivity and CGRP mRNA in endocrine cells or Kulchitsky (K) cells of human and rat lung [Wada et al. 1987b]. In this report, an examination was made of developmental changes in the expression of the CGRP gene in rat lungs by immunohistochemistry, radioimmunoassay (RIA) and Northern hybridization. CGRP-positive K-cells in lung tissue appeared on the 18th day of gestation. Their number was greatest on the 20th day of gestation and then decreased postnatally. The level of CGRP in rat lung was found to be highest in a 1-day-old neonate by RIA. In the Northern hybridization of rat lung using the CGRP 3' non-coding region (exon 6) of the first human CT/CGRP gene as the probe, 1.0 kilobase (kb) CGRP mRNA was found to be abundant on the 20th day of gestation and in a 1 day-old neonate. It thus appears that CGRP in rat lung is essential for pulmonary adaptation at birth and/or from the last intrauterine stage to the early neonatal period.

In situ freezing of the brain for metabolic studies: evaluation of the "box" method for large experimental animals.

The box method of freezing the brain in situ was assessed in baboons. The cooling rate of the tissue was monitored in several regions located at various depths from the skull surface. These measurements allowed us to examine the time required for the tissue to reach 0 degree C, in relation to its depth measured from the top of the skull. To define brain regions with proven ischaemia, frozen tissue sections were surveyed for areas of decreased pH. In addition, concentrations of ATP, phosphocreatine, and lactate were determined in gray matter located at various depths from the top of the brain surface. Normal tissue pH and low lactate concentration, without any significant decrease in high-energy phosphate levels, were found in regions at a depth less than approximately 10 mm from the brain surface. Deep structures including the inferiomedial aspect of the temporal lobe, the lateral geniculate body, and the limbic system (hippocampus) consistently showed mild tissue acidosis, indicating that these regions were subjected to some degree of ischaemia before they were reached by the freezing front. In some cases, acidosis was also detectable in the thalamus, basal ganglia, and in the deeper part of some sulci. We conclude that, with baboons, in situ freezing using the box method is valid for metabolic studies of the cerebral cortex and structures located at a depth less than approximately 10 mm from the top of the brain surface.

Brain tissue concentrations of ATP, phosphocreatine, lactate, and tissue pH in relation to reduced cerebral blood flow following experimental acute middle cerebral artery occlusion.

Local CBF (LCBF) was compared with the corresponding local tissue concentration of ATP, phosphocreatine (PCr), and lactate in anaesthetized baboons subjected to focal ischaemia produced by middle cerebral artery occlusion (MCAO). LCBF hydrogen electrodes were implanted in cortical regions where MCAO had been previously shown to produce severe and penumbral ischaemia and in posterior regions where blood flow is not altered. Metabolites were assayed in small tissue samples collected either by cryoprobe biopsy in the regions where LCBFs were measured (series 1) or by sampling appropriate regions of the rapidly frozen brain (series 2). Subsequent topographical study of brain tissue pH with umbelliferone was performed in this latter series. The results from these two series are compared and discussed in terms of the more appropriate way to perform simultaneous electrode measurements and analysis of tissue samples for studying focal ischaemia in the primate brain. They confirm that the concentrations of ATP and PCr decrease, and that lactate level increases, with decreasing blood flow. These metabolites tended to change more rapidly below a blood flow threshold, rather than showing a steady decrease as the blood flow was reduced, although the variability of the data precluded us from establishing this with confidence. Topographical study of tissue pH often showed sharp boundaries between zones of very low pH and regions with normal pH.

Functional and biochemical effects on rat lung following instillation of crocidolite and chrysotile asbestos.

Groups of rats received a single intratracheal instillation of saline (control), 1.6 mg crocidolite, and 1.6 mg chrysotile. At 1, 3, 6, 9, and 12 mo after instillation, respiratory functional tests were performed and the lungs were biochemically analyzed. Throughout 12 mo, obstructive small-airway abnormalities with air trapping were observed in the asbestos-treated rats. The degree of airway obstruction and air trapping was higher in the chrysotile-treated group than in the crocidolite-treated group. Lung weight increased significantly in both asbestos-treated groups. Collagen and elastin content of unit lung weight significantly increased in the chrysotile-treated group compared with the control and crocidolite-treated group throughout the experimental period up to 12 mo. However, the collagen and elastin content of unit lung weight in the crocidolite-treated group was not as different from the control group. These results indicate that chrysotile is more fibrogenic than crocidolite, as has been suggested in histopathological studies.
