[Radiotherapy of nasopharyngeal carcinoma].

The results of irradiation on fifty seven patients with nasopharyngeal carcinoma, treated from 1964 to 1985 at the Shinshu University Hospital, were studied. Forty-two percent of the patients had T3-4 primaries and 68% had N1-3 regional nodes; 86% had Stages III-IV of the disease. Of the 57 patients, 32 had squamous cell carcinomas, and 25 had lymphoepitheliomas. The overall 5-year survival rate was found to be 57%. Primary size (T) and histology influenced the survival of patients; the 5-year survival rate was 70% for T1-2 primary and 39% for T3-4 primary, and 39% for squamous cell carcinoma and 80% for lymphoepithelioma.

Distribution of collagen type IV in soft tissue tumors. An immunohistochemical study.

The distribution of collagen type IV, one of the major constituents of basement membrane, was studied immunohistologically in a series of 103 soft tissue tumors including those of peripheral nerve origin, smooth muscle origin, striated muscle origin, fibrous tissue origin, fibrohistiocytic origin, adipose tissue origin, synovial tissue origin, and blood vessel origin, paragangliomas, alveolar soft part sarcomas, granular cell tumors, and epithelioid sarcomas. Intensely positive staining for collagen type IV was observed in neurilemomas, neurofibromas, malignant schwannomas, and blood vessel tumors. Weakly to moderately positive staining was seen in leiomyomas, angiomyomas, and leiomyosarcomas. In contrast, synovial, fibroblastic and fibrohistiocytic tumors, benign or malignant, were negative. In paragangliomas, granular cell tumors, and alveolar soft part sarcomas, positive staining was evident surrounding nests or clusters of tumor cells. In all tumors, staining for collagen type IV clearly illustrated the vascular pattern.

Secretion of type IV collagen by keratinocytes of human adult.

Secretion of type IV collagen by keratinocytes was studied by anti-type IV collagen immunoglobulin in pure keratinocyte culture of the human adult. The cultivated keratinocytes secreted type IV collagen into the space between the cell growth and the underlying fixed film of human skin collagen type I. The secreted collagen was accumulated on the cell surface to form junction structures.

Atypical pityriasis rosea.

Six cases of pityriasis rosea with atypical morphology and distribution of the eruption are reported. The eruption did not show a typical 'Christmas-tree' arrangement, confined to the trunk and proximal parts of the extremities. However, the histology of the eruption revealed dyskeratotic cells in the epidermis and extravasated erythrocytes in the dermis, which were recently reported as rather characteristic findings of this disease. Prodromal symptoms, course and response to therapy were compatible with pityriasis rosea. Histological examination is important and helpful for the diagnosis of atypical cases.

Dermal changes in Ehlers-Danlos syndrome.

Skin biopsies from thirteen patients suffering from Ehlers-Danlos syndrome, including 6 of the mitis type, 4 of the benign hypermobile type, one of the X-linked type, one of the ocular type and one of the periodontitis type, were studied by electron microscopy after routine preparation. Collagen fibrils showed a distorted arrangement of bent, curled or twisted fibrils and thread-like material. Similar changes may be seen in the skin of other hereditary disorders of connective tissue. However, abnormal collagen fibrils in normal skin suggests one of eight types of Ehlers-Danlos syndrome. Clinical variants cannot be differentiated on the basis of ultrastructural findings. Elastic fibres were normal without degenerative changes. Perineurium was lacking in dermal nerves of most patients. Fibroblast-like cells showed no cystic cisterna of endoplasmic reticula.

Separation of osteoblast-like cells from bone marrow by fluorescence-activated cell sorting.

The purification of the osteoblast-like cells (2-3%) among the bone marrow cells (BMC) of C57BL/6 mice using a specific anti-osteoblast serum and a fluorescence-activated cell sorter is described. The antiserum was raised against osteoblast cells isolated from calvaria from neonatal mice. The majority of the cells of the osteoblast-enriched fraction from bone marrow showed a parathormone-induced increase in cyclic adenine monophosphate but no response to calcitonin. This is similar to the response of osteoblast cells obtained from the calvaria. Electron microscopic studies of the extracellular matrix of cultured osteoblast-like cells purified from bone marrow showed the deposition of apatite crystals within and in close apposition to the vesicles. These findings suggest that the isolated cell population was enriched in osteoblasts. Such a cell system from bone marrow might provide an experimental system for investigating the mechanism of bone formation.

Effect of PSK on interferon production in tumor-bearing mice.

The effect of PSK on the depressed interferon (IF) production in tumor-bearing mice was studied. In tumor-bearing mice, in vitro IF production by spleen cells treated with polyinosinic-polycytidylic acid (poly I:C) was remarkably inhibited. However, these inhibitions were prevented by the intraperitoneal (ip) administration of PSK. Mice were inoculated intravenously (iv) with poly I:C-treated spleen cells, administered with PSK ip at 3 days after the tumor inoculation. When PSK was not given, poly I:C-treated spleen cells did not show an inhibitory effect on tumor growth. In mice given PSK ip, poly I:C-treated spleen cells exerted slightly inhibiting effects on tumor growth. These results suggest that PSK prevented such a modulation in tumor-bearing mice.

Relationship between bactericidal and phagocytic activities of peritoneal macrophages induced by irritants.

Cellular accumulation to the peritoneal cavity and modification of various functions of peritoneal macrophages were observed in mice injected intraperitoneally (ip) with thioglycollate medium (TG), liquid paraffin, proteose peptone and Corynebacterium parvum. The cellular composition of peritoneal exudates at 4 days after injection of irritants was almost the same in all the groups and the proportion of macrophages was increased approximately 4 times more than nontreated controls. The ability to kill Listeria monocytogenes and to generate chemiluminescence (CL) were augmented strongly in C. parvum-induced macrophages, while depressed in TG-induced macrophages. The activities of liquid paraffin- or proteose peptone-induced macrophages were almost the same as those in nontreated controls. However, the ability to phagocytose native sheep erythrocytes was greatly augmented both in C. parvum- and TG-induced macrophages. There is thus a discrepancy between bactericidal activity and phagocytic activity among macrophages induced with various irritants.

Lipid droplets containing DNA-histones released in the culture medium of transformed rat fibroblasts (HY1).

Lipid droplets appeared during the growing phase in the culture medium of incompletely transformed rat fibroblasts (HY1) induced by AccI-H fragments of adenovirus 12 DNA. These droplets consisted of neutral lipids, DNA, histones and RNA. Electron microscopic observations showed that the droplets had no lipid-bilayers on their surfaces which accounted for the tendency of the droplets to readily fuse together and become larger, and that the inner structures of the droplets looked like networks of fibrous matter. Experiments with DNA hybridization showed that the droplet DNA was composed of both cellular and adenoviral DNA, and that the cellular DNA in the droplets seemed to be derived from various cell DNA sequences. These results suggest that the droplets were derived from parts of the nuclear components of HY1 cells. The mechanisms for droplet release are discussed.

Human Ia-like-antigen-expressed cutaneous T cell lymphoma associated with Epstein-Barr virus infection.

2 cases of cutaneous T cell lymphoma are reported. In both cases immunological studies revealed that the tumor cells from the cutaneous lesions had human Ia-like antigen on the cell surface, which was considered to be alloantigen primarily present on B lymphocytes. Serum examination showed an extremely high titer of two types of antibody against Epstein-Barr (EB) virus in the 1st case and a relatively high titer of one type of antibody in the 2nd case, suggesting that the patients had EB virus infection. Recently, some hematologists found the significant correlation of T lymphocytosis and the appearance of human Ia-like antigen in infectious mononucleosis. Based on these findings, it is assumed that EB virus has the ability of the immunological transformation of cutaneous neoplastic lymphocytes, and so we should not overlook the susceptibility of phenotypical changes of the original cell by EB virus when the immunological classification of cutaneous malignant lymphoma is done.

[Effect of PSK on tumor-specific immunity induced by MMC-treated syngeneic tumor cells].

Immunization with MMC-treated EL-4 tumor cells could raise cytotoxic activity of non-adherent PE cells and resistance against rechallenge with small or medium doses of viable tumor cells. Administration of PSK augmented the generation of cytotoxic lymphocytes and the induction of resistance against rechallenge in mice immunized with such MMC-treated tumor cells. Augmented generation of cytotoxic lymphocytes may be ascribed to systemic effects of PSK but not to local effects in the peritoneal cavity, since augmenting effects of PSK were observed not only after intraperitoneal administration but also after oral administration. Either after intraperitoneal administration or after oral administration cytotoxic activity was detected in PE cells but not in spleen cells. Cytotoxic activity was detected in PE cells but not in spleen cells after intraperitoneal injections of MMC-treated tumor cells. Cytotoxic lymphocytes appear to differentiate to their mature form capable of being detected by 51Cr-release test principally at the site of direct graft rejection. Intraperitoneal administration of PSK was more effective in the augmentation of cytotoxicity of PE cells than oral administration. PSK may be able to have contact with precursors of cytotoxic lymphocytes more efficiently after intraperitoneal administration. Immunity against syngeneic tumor cells appears to be effective in elimination of small doses of tumor cells but to be overcome by medium or large doses of tumor cells at the rechallenge. Administration of PSK increased the threshold number to be eliminated by immune hosts. This finding seems to be important in relation to augmentation of resistance against metastasis or local implantation with a limited number of tumor cells.
