Application of sorbent extraction chromatography to the purification of diethylstilboestrol extracted from muscle tissue and determined by radioimmunoassay.

A chromatographic procedure is described for the purification of bovine muscle tissue extracts prior to the determination of diethylstilboestrol (DES) by radioimmunoassay. Sorbent extraction chromatography of tissue extracts on reversed-phase octadecyl (C18) columns gives adequate recovery of residue and a suitable sample for radioimmunoassay. This procedure, which is simple and rapid, provides an alternative to a more complex purification by high-performance liquid chromatography. Using this method, the limit of detection for DES in muscle samples is approximately 40 pg/g.

A controlled study of the ocular effects of tamoxifen in conventional dosage in the treatment of breast carcinoma.

The use of tamoxifen is expanding in its application in the treatment of breast carcinoma, and proposed prophylactic use. Tamoxifen in high dosage and in some reports at conventional dosage is associated with ocular toxicity. This study in a single blind fashion examined 79 patients receiving tamoxifen for varying treatment durations at conventional dosage and 115 patients not receiving tamoxifen. No ocular toxicity was found. Further follow up studies are required to ensure no toxicity develops in patients receiving high cumulative dosage on conventional therapy.

Human coffee drinking: manipulation of concentration and caffeine dose.

In a residential research ward coffee drinking was studied in 9 volunteer human subjects with histories of heavy coffee drinking. A series of five experiments was undertaken to characterize adlibitum coffee consumption and to investigate the effects of manipulating coffee concentration, caffeine dose per cup, and caffeine preloads prior to coffee drinking. Manipulations were double-blind and scheduled in randomized sequences across days. When cups of coffee were freely available, coffee drinking tended to be rather regularly spaced during the day with intercup intervals becoming progressively longer throughout the day; experimental manipulations showed that this lengthening of intercup intervals was not due to accumulating caffeine levels. Number of cups of coffee consumed was an inverted U-shaped function of both coffee concentration and caffeine dose per cup; however, coffee-concentration and dose-per-cup manipulations did not produce similar effects on other measures of coffee drinking (intercup interval, time to drink a cup, within-day distribution of cups). Caffeine preload produced dose-related decreases in number of cups consumed. As a whole, these experiments provide some limited evidence for both the suppressive and the reinforcing effects of caffeine on coffee consumption. Examination of total daily coffee and caffeine intake across experiments, however, provides no evidence for precise regulation (i.e., titration) of coffee or caffeine intake.

The use of mannitol in intraocular surgery.

100 ml of 20% Mannitol given between 30 and 60 minutes prior to surgery effectively lowers the intraocular pressure and increases the anterior chamber depth. Larger volumes give an almost identical drop in pressure and identical increase in depth of the anterior chamber although they act more rapidly and over a longer period. Rebound rise in intraocular pressure does not occur within the first two hours.

Comparison of matrix solid phase dispersion (MSPD) with a standard solvent extraction method for sulphamethazine in pork muscle using high performance liquid and thin layer chromatography.

A rapid and simple extraction/clean-up procedure (matrix solid phase dispersion, MSPD) for the determination of sulphamethazine (SMZ) in pork muscle tissue is compared with a solvent extraction method. Extracts of samples fortified with SMZ or of incurred samples were found to be free from interfering compounds when chromatographed using HPLC or TLC separation systems. Recovery of SMZ from fortified samples is greater than 80% and residue levels of incurred samples found using the MSPD procedure compare favourably with results obtained using the solvent extraction method. Use of aqueous back extraction of SMZ from dichloromethane is also reported as an alternative step to solvent evaporation for ease of use in both the laboratory and in industry (i.e. at slaughter plants).

Congenital absence of extraocular muscles: MR demonstration.

We describe a case of an infant with complete congenital absence of the extraocular muscles, noted at surgery and confirmed by multiplane MR, which found no evidence of extraocular muscle tissue. This almost certainly represents an extreme form of bilateral congenital fibrosis syndrome.
