[The "e" antigen (HBeAg) in leprosy].

Sixty-six sera from leprosy patients, 42 with lepromatous leprosy (l.l.) and 24 with tuberculoid leprosy (t.l.) were examinated for the presence of e-antigen and e-antibody by immunodiffusion. The test for e-antigen was negative in all leprosy patients; e-antibodies were found in only three patients (4.5%) with l.l., all carriers of hepatitis B antigen. The HBsAg test was positive in 23.8% of the l.l. cases and in 12.5% of the t.l. cases; the anti-HBs was present in 38.1% of the l.l. cases and in 41.6% of the t.l. cases. The results suggest the possibility that the leprosy patients positive for the surface B antigen represent a very low risk and can be considered as chronic asymptomatic carriers of HBsAg.

[Study of skin reactivity to tuberculin and sensitins of atypical mycobacteria (with special reference to M. africanum in Somalia)].

Skin testing with antigens from M. tuberculosis (PPD-S) and various atypical mycobacteria was carried out in Somalia among 170 patients with active tuberculosis. Percentage of positive reactons (5 mm or greater) was 70,58% with PPD-S, 60,0% with the antigen from M. africanum and M. scrofulaceum, 56,47% with the antigen from M. xenopei, 10,58% with the antigen from N. otididis and 4,50% with the antigen from N. farcinica. The percentage of subjects in whom the reaction with the antigen from M. africanum was greater than with PPD-S was 10,58%; more precisely in a percentage of 16,0% the skin testing from M. africanum was positive against a negative reaction to PPD-S, meanwhile in a percentage of 8,33% both skin tests were positive, with a greater reaction for a M. africanum test. As far as other antigens are concerned at least one of these tests was greater on the 28,23% the PPD-S one.

Mammary fetal gland: identification of new oncofetal antigens by monoclonal antibodies B72.3, MM1.80 and 4.36.

AIMS AND BACKGROUND: The B72.3, MM1.80 and 4.63 monoclonal antibodies directed against tumor-associated antigens react in human breast also with metaplastic, preneoplastic and metastatic cells, whereas they do not react with normal adult mammary tissue. The aim of our study was to point out the expression of these antigens during mammary gland development, since tumor-associated antigens are known to represent antigens of differentiation. STUDY DESIGN: Fifty female fetal breasts between 20 and 40 weeks of gestational age were studied. RESULTS: Mammary tissue was identified only in 15 cases. B72.3, MM1.80 and 4.36 monoclonal antibodies reacted with epithelial antigens and maintained the same location and intensity in the various gestational ages. Immunoreactivity was weak, cytoplasmic and widespread for the 4.36 monoclonal antibody, intense and cytoplasmatic in a large number of cells for the B72.3 monoclonal antibody, and intense and luminal for the MM1.80 monoclonal antibody. CONCLUSIONS: Such data further support the hypothesis that the normal process of development and differentiation can occur during tumor progression processes. Identification of these new oncofetal markers could offer a new perspective able to recognize the different phases of neoplastic progression and could be useful for prevention.

Quantitative determination of mammary tumor virus in individual samples of mouse milk.

The possibility to determine quantitatively the intact mammary tumor virus (MTV) in milk of mice carrying milk-transmitted MTV has been assayed by a method that allows direct comparison between individual milk samples. The method is based on (a) the measure of light scattering of partially purified MTV preparations, (b) the use of milk from genetically identical MTV free mice as blank and (c) the quantitative reference to the total protein content of whole milk. The sensitivity, specificity and reproducibility of the procedure, as well as the requirement of appropriate quantitative references, are illustrated and discussed. BALB/c (MTV free), BALB/cfC3H, and BALB/cfRIII mice have been used.

Quantitative comparison of milk-released C3H and RIII mammary tumor viruses in infected BALB/c hosts.

The C3H and RIII mammary tumor viruses (MTV) carried by BALB/cfC3H and BALB/cfRIII breeding females have been quantified and compared in milk samples, after partial purification with a sucrose density gradient. The samples were collected at identical times during the first 3 lactation periods from individual mice (6 per strain), standardized for age at delivery and size of litter. Milk samples from 6 MTV negative BALB/c controls have also been analyzed. Data for comparison are expressed in optical density units (ODU) and refer to the protein content of the whole milk using MTV-negative Balb/c milk as blank. The results have shown 1) an increase of MTV released through milk in each MTV-carrying female from the first (average ODU, 0.542) to the second (1,351) and third (2.105) lactation, 2) individual variations, and 3) a significant difference in release between C3H and RIII MTV, the latter being more than double (average ODU, 1.801) in respect to the former (0.847). The apparent discrepancy between these results and the bioactivity in BALB/c mice of C3H and RIII MTV, significantly lower for the latter, is discussed.

Molecular profile, tissue distribution and prognostic evaluation of a human melanoma-carcinoma antigen recognized by the murine monoclonal antibody B1.1.

Using the murine monoclonal antibody (MoAb) B1.1 we have analyzed the immunochemical profile and the tissue distribution of a human melanoma associated antigen (MAA) carrying an epitope shared by the 180 kd CEA. Results of this study have demonstrated that the epitope expressed by the MAA is carried by a distinct set of molecules of 110-140 kd. Similarly to the 180 kd CEA molecules synthesized by carcinomas, the expression of the melanoma associated CEA like components (MA-CEA) is upregulated by IFN-alpha. The tissue distribution of MA-CEA is not restricted to malignant primary and metastatic melanocytic lesions but is found also at low levels in 64% of benign nevi. No circulating CEA was found in patients bearing widespread metastatic disease of MA-CEA positive lesions. Preliminary clinical evaluation of stage I melanoma patients bearing MoAb B1.1 positive lesions has not shown a significative prognostic association of this phenotypic marker with clinical course of the disease.

Immunological properties of a radioiodinated monoclonal antibody and its F(ab')2mu fragments directed against the polymorphic epithelial mucin of human breast cancer.

The purification of the IgM monoclonal antibody 436 against a breast tumor antigen from mouse ascitic fluid is reported. The purified immunoglobulin was radioiodinated and the resulting product assessed for its binding capacity and binding specificity. Purified IgM-436 served for F(ab')2 mu preparation which was tested for its antigen binding capacity. Radioiodinated IgM-436 and its F(ab')2 mu retained their immunological activity which was never lower than those of the corresponding cold products.

Ehrlichia infection in Italy.

Immunoglobulin M seroconversion to Ehrlichia chaffeensis was documented in U.S. citizens bitten by ticks in Sardinia. Seven cases of suspected ehrlichiosis in local residents were not confirmed by laboratory tests. In Alpine areas antibodies to E. phagocytophila were detected in persons at high risk, i.e., foresters (8.6%) and hunters (5.5%), and in controls (1.5%). Of 153 persons bitten by ticks, only one was Ehrlichia antibody-positive after 6 months.
