Why Congo red binding is specific for amyloid proteins - model studies and a computer analysis approach.

BACKGROUND: The complexing of Congo red in two different ligand forms - unimolecular and supramolecular (seven molecules in a micelle) - with eight deca-peptides organized in a b-sheet was tested by computational analysis to identify its dye-binding preferences. Polyphenylananine and polylysine peptides were selected to represent the specific side chain interactions expected to ensure particularly the stabilization of the dye-protein complex. Polyalanine was used to verify the participation of non-specific backbone-derived interactions. MATERIAL AND METHODS: The initial complexes for calculation were constructed by intercalating the dye between the peptides in the middle of the beta-sheet. The long axis of the dye molecule (in the case of unimolecular systems) or the long axis of the ribbon-like micelle (in the case of the supramolecular dye form) was oriented parallel to the peptide backbone. This positioning maximally reduced the exposure of the hydrophobic diphenyl (central dye fragment) to water. In general the complexes of supramolecular Congo red ligands appeared more stable than those formed by individual dye molecules. Specific interactions (electrostatic and/or ring stacking) dominated as binding forces in the case of the single molecule, while non-specific surface adsorption seemed decisive in complexing with the supramolecular ligand. RESULTS: Both the unimolecular and supramolecular versions of the dye ligand were found to be likely to form complexes of sufficient stability with peptides. The low stability of the protein and the gap accessible to penetration in the peptide sheet seem sufficient for supramolecular ligand binding, but the presence of positively charged or hydrophobic amino acids may strengthen binding significantly. CONCLUSIONS: The need for specific interaction makes single-molecule Congo red binding rather unusual as a general amyloid protein ligand. The structural feature of Congo red, which enables specific and common interaction with amyloid proteins, probably derives from the ribbon-like self-assembled form of the dye.

Neurotensin stimulates the growth and secretion of rat adrenal zona glomerulosa.

Within 2 days neurotensin (NT) and ACTH administrations markedly enhanced the average volume of zona glomerulosa (ZG) cells and plasma aldosterone concentration (PAC) in intact rats. In dexamethasone-treated rats, both NT and ACTH evoked a clearcut ZG-cell hypertrophy, but only NT was able to raise PAC. In conclusion, our findings indicate that NT is a potent stimulator of the growth and secretion of rat ZG in vivo, and suggest that the mechanism underlying this action of NT does not involve the well-known NT-induced stimulation of ACTH release.

Homocysteine, vitamin B12, and folic acid in age-related macular degeneration.

PURPOSE: Hyperhomocysteinemia is considered an independent risk factor for atherosclerosis and thrombosis. The purpose of this study was to evaluate plasma homocysteine, red blood cell folate, plasma folate, and plasma vitamin B12 concentration in patients with exudative age-related macular degeneration (ARMD). METHODS: The participants of this study included 30 patients aged 60 to 71 years (mean age 66.2+/-3.6) with exudative ARMD. Plasma homocysteine levels were determined by high performance liquid chromatography (HPLC). Red blood cell folate, plasma folate, and plasma vitamin B12 concentration were determined using a standard kit (Dualcount Solid Phase No Boill radioassay kit for B12/folic acid, DPC Diagnostic, USA) by radioassay method. RESULTS: The plasma concentration of Hcy (14.88+/-6.23 micromol/L) in ARMD patients was significantly increased (p<0.0001) compared with the control group (8.72+/-3.34 micromol/L). We found not a significant decrease of the plasma vitamin B12 concentration in the ARMD group (476.88+/-220.91 pg/mL) compared with the control group (527.08+/-208.97 pg/mL). Red blood cell folate (158.44+/-56.30 ng/mL) and plasma folate (6.5+/-3.4 ng/mL) in ARMD patients were also not significantly decreased when compared with the control group (183.86+/-59.33 ng/mL and 7.93+/-5.05 ng/mL). CONCLUSIONS: Hyperhomocysteinemia might be one of the risk factors for the exudative form of ARMD.

[Incidental granulocyte locomotion in hay fever].

Accidental locomotion of granulocytes in 10 patients with pollinosis has been analysed. Marked impairment of this function was noted during grass pollen season. This phenomenon may partially explain increased incidence of the purulent dermatitis and mucosal involvement in this group of patients.

The effect of 4-aminopyrazolo (3,4-d) pyrimidine (4-APP) on some parameters of carbohydrate metabolism in the rat.

The study was designed to evaluate some parameters of carbohydrate metabolism in the rat as influenced by 4-APP, an adenine analogue. Adult female rats were injected with 1 mg 4-APP/100 g body weight/day for 3 days. 4-APP evoked a marked enlargement of the liver with lipid droplets accumulation in hepatocytes. This was accompanied by a marked lowering of the liver glycogen content. Within 3 days 4-APP did not change serum glucose, insulin and free fatty acid concentration. Serum glycogenolytic activity studied in an in vitro system showed 7 times as high glucose releasing ability in 4-APP treated rats as that of the serum of control animals. 4-APP resulted also in a marked enlargement of the adrenal medulla and lowered adrenaline and noradrenaline concentrations in the gland. The possibility of an activation of glycogenolysis in the liver of 4-APP treated rats has been discussed.

Gestational changes in hamster adrenal cortex. II. Proliferative activity of adrenocortical cells in the course of gestation.

Proliferative activity of adrenocortical cells was determined in the course of hamster gestation. The highest adrenal gland weight was noted at day 5 of pregnancy, followed by a gradual decrease towards the end of gestation. As revealed by stereology, total number of adrenocortical cells increased notably at days 5 and 10 of pregnancy and was followed by a prompt drop to the value observed in the control group. In vitro 3H-thymidine incorporation per pair of adrenals increased slightly at days 5 and 10 of pregnancy and decreased at day 15. The highest DNA content was found in adrenals taken at day 10 of gestation. The number of mitoses per adrenal section also increased at days 5 and 10 and reached the control value at day 15 of pregnancy. After vincristine administration, mitoses were predominantly seen in glomerulosa and outer fasciculata zones. Thus in the course of hamster pregnancy, a marked increase in proliferative activity of adrenocortical cells occurs up to day 10 of gestation and is followed by a significant drop in proliferative activity in the final days of pregnancy.
