The Candida albicans plasma membrane and H(+)-ATPase during yeast growth and germ tube formation.

PMA1 expression, plasma membrane H(+)-ATPase enzyme kinetics, and the distribution of the ATPase have been studied in carbon-starved Candida albicans induced with glucose for yeast growth at pH 4.5 and for germ tube formation at pH 6.7. PMA1 expression parallels expression of the constitutive ADE2 gene, increasing up to sixfold during yeast growth and twofold during germ tube formation. Starved cells contain about half the concentration of plasma membrane ATPase of growing cells. The amount of plasma membrane ATPase is normalized prior to either budding or germ tube emergence by the insertion of additional ATPase molecules, while ATPase antigen appears uniformly distributed over the entire plasma membrane surface during both growth phases. Glucose addition rapidly activates the ATPase twofold regardless of the pH of induction. The turnover of substrate molecules per second by the enzyme in membranes from budding cells quickly declines, but the enzyme from germ tube-forming cells maintains its turnover of substrate molecules per second and a higher affinity for Mg-ATP. The plasma membrane ATPase of C. albicans is therefore regulated at several levels; by glucose metabolism/starvation-related factors acting on gene expression, by signals generated through glucose metabolism/starvation which are thought to covalently modify the carboxyl-terminal domain of the enzyme, and possibly by additional signals which may be specific to germ tube formation. The extended period of intracellular alkalinization associated with germ tube formation may result from regulation of proton-pumping ATPase activity coupled with higher ratios of cell surface to effective cytosolic volume.

Identification of galanin-immunoreactive cells in the anterior pituitary of male monkeys (Macaca fascicularis).

The localization of galanin (GAL) was studied in the anterior pituitary of adult male macaque monkeys immunohistochemically. GAL-immunoreactive fiber bundles were noted in the posterior lobe of the pituitary. Cells immunoreactive for GAL were observed in the anterior lobe, and colocalization studies revealed that GAL-like immunoreactivity was present in gonadotrophs and thyrotrophs. The differences among the monkey, human and rat in GAL-immunoreactive localization may indicate that the regulation of GAL in the three species differs.

Increase of histidine decarboxylase activity in mice hypothalamus after intracerebroventricular administration of lipopolysaccharide.

The effect of intracerebroventricular (icv) administration of lipopolysaccharide on histidine decarboxylase activity and histamine content in the hypothalamus were investigated in male mice of ddY strain in vivo. Two-fold increase in histidine decarboxylase activity (HDC) was observed 4 h after administration of 50 mcg lipopolysaccharide, and HDC activity returned to the basal level within 12 h after injection. Furthermore, histamine contents showed a slight decrease at 1 and 2 h and a mild increase at 12 h after administration. However, changes in histamine content were not statistically significant. These results suggest that the increase of HDC activity in the hypothalamus by lipopolysaccharide may be involved in the central neuroimmune responses.

[Development of an enzymatic fluorescence method for detecting of cholesterols in circulating monocyte].

Since the infiltration of macrophage/monocyte into the arterial intima would be the first and most important step to developing the atherosclerosis, it is important to analyze the lipid composition not only in the circulating blood but also in the cell. We developed the high sensitive enzymatic assay method of total and free cholesterol and employed this assay to the measurement of monocyte cholesterol ester. Diabetic patients had high total cholesterol and cholesterol ester of monocyte. The elevated monocyte cholesterol ester was related to the LDL-cholesterol and Lp(a) concentrations in the plasma.

Stimulatory effects of quinpirole hydrochloride, D2-dopamine receptor agonist, at low concentrations on prolactin release in female rats in vitro.

Dopamine (DA) has dual actions (inhibitory and stimulatory) in the regulation of prolactin (PRL) release, depending on its concentration. To investigate the stimulatory effects of DA, perifused rat anterior pituitary cells were exposed to the highly-specific DA D2 receptor agonist, quinpirole hydrochloride (LY). Very low concentrations of LY (10(-12)-10(-10) M) stimulated PRL release and potentiated thyrotropin-releasing hormone (TRH)-induced PRL release. Higher concentrations of LY did not stimulate. Pretreatment with pertussis toxin (30 ng/ml, 24 h) completely abolished these effects of LY. The D2 receptor antagonist, metoclopramide, also blocked the potentiation by LY of TRH-induced PRL release. These data indicate that very low concentrations of dopamine stimulate PRL release via an interaction with a D2 receptor connected to a pertussis toxin-sensitive G protein.

Corticotropin releasing factor and galanin-containing neurons projecting to the median eminence of the rat.

To identify corticotropin releasing factor (CRF)- and galanin (GAL)-containing neurons projecting to the median eminence, a retrograde tracing method with True blue was combined with a double-staining method. It was demonstrated that some True blue-labeled CRF neurons in the paraventricular nucleus were also immunoreactive for GAL. These findings support the hypothesis that GAL and CRF are simultaneously involved in regulation of ACTH secretion from the anterior pituitary.

Identification of dopamine and growth hormone-releasing factor-containing neurons projecting to the median eminence of the rat by combined retrograde tracing and immunohistochemistry.

The topographical location of tyrosine hydroxylase (TH) neurons, a marker for dopamine neurons in the hypothalamus, that project to the median eminence was identified with immunofluorescence staining in combination with retrograde transblue, indicating that they projected to the median eminence. Only a few retrogradely labeled TH cells were observed in the periventricular nucleus and the lateral basal hypothalamus. The elution restaining procedure revealed that an average of 32% of the labeled TH cells in the ventrolateral portion of the ARC contained GRF. These findings support the hypothesis of co-expression of dopamine with GRF from the ARC.

[Countermeasures for urinary incontinence in patients with senile dementia: correlation between urinary incontinence severity, senile dementia severity, and activity of daily living].

The actual conditions of urinary incontinence in 101 patients with senile dementia (23 men and 78 women, average age 80 years) and their urinary control was studied. The three categories of 1) dementia severity, 2) activity of daily living and 3) degree of urinary incontinence, were each divided into five grades and the patients were evaluated accordingly. The correlation among the grades was studied. Of the 101 patients, 87 had urinary incontinence. These 87 were further subdivided into two groups: an ambulatory patient group (25) in which the patients could manage their own daily activity, and the bedridden group (62). The countermeasures for the first group of patients were, usage of a portable chamber pot for 6 patients and induced urination in 5. An anticholinergic agent was administered to 47 of the bedridden patients. The condition of urinary incontinence was reevaluated in both groups 4 weeks later. A high degree of correlation was observed among the grades of dementia severity, activity of daily living and urinary incontinence severity. After the countermeasures taken, improvement in urinary incontinence was observed in the 6 patients who used a portable chamber pot, and in the 5 patients who were subjected to induced urination. No improvement was seen in urinary incontinence in the bedridden patients who were treated with an anticholinergic agent. In conclusion, countermeasures for urinary incontinence in senile dementia should be indicated primarily for patients who can manage their own daily activity.

Neurotensin and growth hormone-releasing factor-containing neurons projecting to the median eminence of the rat: a combined retrograde tracing and immunohistochemical study.

The origins of the neurotensin (NT)-containing nerve terminals in the median eminence were determined in rats by a combination of retrograde labeling with True blue and immunofluorescence staining. Moreover, the distribution of NT and growth hormone-releasing factor (GRF) was analyzed in the arcuate nucleus (ARC) with an elution-restaining procedure. The retrogradely labeled NT neurons were found mainly in the ARC. Only a few labeled NT neurons were seen in the periventricular and the paraventricular nuclei. The elution restaining procedure demonstrated that an average of 43% of the labeled NT cells in the ventral part of the ARC restained for GRF. These findings support the hypothesis of co-expression of NT with GRF from the ARC.

Characterization of the stimulatory effect of galanin on growth hormone release from the rat anterior pituitary.

The present study was undertaken to investigate the direct actions of rat galanin (R-GAL) on growth hormone (GH) release from the rat anterior pituitary in vitro. R-GAL modestly but significantly stimulated GH release without an increase in intra- and extracellular cyclic AMP levels in monolayer cultures of rat anterior pituitary cells. This stimulatory effect of R-GAL was dose-dependent but not additive with that of GH-releasing factor (GRF). R-GAL-stimulated GH release was less sensitive to the inhibitory effect of somatostatin than was GRF-stimulated GH release. In perfusions of rat anterior pituitary fragments, R-GAL induced a gradual and sustained increase of GH release. Incremental GH release derived in part from preformed stored GH. These data confirm that R-GAL acts at the pituitary level to stimulate GH release by a mechanism distinct from that of GRF.

Initial attachment of Candida albicans cells to buccal epithelial cells. Demonstration of ultrastructure with the rapid-freezing technique.

The rapid-freezing technique was applied in association with scanning and transmission electron microscopy to observe the initial attachment (or contact) of Candida albicans cells to exfoliated human buccal epithelial cells. Low temperature scanning electron microscopy provided detailed three-dimensional morphological features of the yeast-epithelial cell association; adhesion of C. albicans cells to host cells was primarily owing to an interaction between fibrillar layer of the yeast cell wall and the membrane interdigitations of the epithelial cells. Such a particular interconnection between the two cells was confirmed by the freeze-substitution fixation for transmission electron microscopy. These results clearly demonstrate the outermost fibrillar cell wall layer of C. albicans responsible for adhesion to host cells.

Immunohistochemical identification of galanin and growth hormone-releasing factor-containing neurons projecting to the median eminence of the rat.

Through the combined demonstration of the retrograde transport of True blue and the immunohistochemical staining of galanin (GAL), the GAL neurons that project to the median eminence were identified. Moreover, the distribution of GAL and growth hormone-releasing factor (GRF) was analyzed in the arcuate nucleus (ARC) with an elution-restaining procedure. Following the injection of True blue into the median eminence, GAL-positive cells labeled with True blue were found mainly in the ARC. In addition, a few GAL neurons in the periventricular, the paraventricular and the supraoptic nuclei were labeled with True blue. The elution-restaining results revealed that many retrogradely labeled GAL neurons in the ARC contained GRF. These findings suggest that a subpopulation of GAL neurons in the ARC sending axons to the median eminence produces, stores and releases a GRF-like peptide.

DNA homology between Candida albicans strains: evidence to justify the synonymous status of C. stellatoidea.

Genetic relatedness between strains of C. albicans and C. stellatoidea was studied by measuring G + C content and overall sequence homology. G + C contents determined by high-performance liquid chromatography were 32.6 to 34.2% for 26 strains of C. albicans and 33.0 to 33.9% for eight strains of C. stellatoidea. DNA-DNA hybridization with two C. albicans and two C. stellatoidea probes revealed that all 34 test strains formed a single cluster in which the extents of hybridization with the heterologous probes ranged between 77.9 and 105.6% of those with the homologous probes. These results give support to the unification of C. albicans and C. stellatoidea into a single species.

Reduction and fixation of jaw fractures using acrylic splints.

This paper proposes the criteria for application of acrylic splints in the treatment of various types of maxillofacial fractures. These were based mainly on the number of remaining teeth that are important for the restoration of occlusion. The acrylic splints studied consisted of intermaxillary, lingual and labiolingual types that hold a dental arch and a cap type that covers a dental arch. It was found that the intermaxillary type was best indicated for the loss of multiple teeth, the lingual type for the predicted intraversion of bone fragments and the labiolingual as well as cap types for the deciduous or mixed dentition. Clinical application based on the above standard, which was attempted in our 60 patients, produced successful results. In addition, the usefulness of these acrylic splints as an intraoral apparatus was discussed.

Acute and late effects of A-bomb radiation studied in a group of young girls with a defined condition at the time of bombing.

Ninety girl students have been identified, who were 14-15 years old when exposed to the atomic bomb while at the Central Telephone Office in Hiroshima located at a distance of 550 meters from the hypocenter. The mortality rate of the students exposed on the second floor of the building was estimated to be 50.9% and those exposed on the first floor (ground level) 33.3%. Doses to the students exposed on the second floor were estimated from cytogenetic evidence to be around 6 Gy in the T65 Dose system or appear to be 4 Gy in the DS86 system. These data indicate that LD50 is around 4 Gy in these young females. Among 28 students who were confirmed to be alive in 1965 and followed to the end of 1988, six students had breast cancer, mostly of invasive ductal type carcinoma. The incidence of breast cancer in the adolescent group was very high, the relative risk being 23.1 with 95% confidence limits of 12.9 to 42.2.

Adansonian study of Candida albicans: intraspecific homogeneity excepting C. stellatoidea strains.

Fifty-six strains of Candida albicans (40 fresh human isolates, 10 laboratory strains, and 6 Candida stellatoidea strains), seven strains of other Candida species, and one strain of Saccharomyces cerevisiae were examined for a total of 182 biochemical and physiological characters. As 121 characters proved positive or negative in all of the strains, analysis of similarity values (simple matching coefficients) derived from the remaining 61 characters revealed that C. albicans strains could be discriminated from other species with similarity values of 70% or lower, and that the cluster of C. albicans strains with internal similarity values above 70% could be divided into two subclusters representing classical C. albicans and C. stellatoidea, with intergroup similarity of 70 to 85% and intragroup similarities of 85% or higher. No discernible difference was noted between isolates from pathological specimens and those from healthy individuals.
