Localization of nascent NADPH-cytochrome c reductase in rat liver microsomes.

Rat liver microsomes incubated with [3H] puromycin in high salt buffer were digested with a mixture of protease, trypsin and chymotrypsin, in both the presence and absence of 1 % deoxycholate. Our observations revealed that the proteolysis of peptidyl puromycin labeled with [3H] puromycin was at least partially protected by the presence of microsomal membrane. Immuno-chemical analyses have further shown that most of the nascent NADPH-cytochrome c reductase in the microsomes was digested with the proteases while serum albumin was effectively protected from the digestion. It is thus proposed that NADPH-cytochrome c reductase synthesized on the membrane bound ribosomes is not transported to the vesicular cavity but directly to the outer surface of the microsomal membrane in a form which is accessible to the proteases.

Avoidance coping behaviors and low social support are related to depressive symptoms in HIV-positive patients in Japan.

The authors examined the influences of several psychosocial factors (i.e., coping behavior responses, social support, etc.) on mood states in 47 human immunodeficiency virus (HIV)-positive patients without the acquired immunodeficiency syndrome (AIDS). No patients fulfilled the DSM-III-R diagnostic criteria for mood disorders, including major depression. However, the HIV group indicate significantly stronger depressive symptoms and lower social support than the healthy control group. The strength of depressive symptoms and poor social support were significantly correlated with one another. Although the HIV group indicated significantly stronger active coping behaviors than the healthy control group, depressive symptoms were significantly and positively correlated with avoidance coping behaviors. When existence of social support was controlled for, this significant correlation was not noted, indicating that avoidance coping behaviors are independently and significantly related to depressive symptoms. The results suggest that, although depressive symptoms are not strong enough to warrant a psychiatric diagnosis of mood disorders, including major depression, avoidance coping behaviors and poor existence of social support may be a high-risk combination for the manifestation of depressive symptoms in HIV-positive patients without AIDS in Japan.

Nuclear receptors CAR and PXR in the regulation of hepatic metabolism.

The nuclear receptors CAR and PXR were first characterized as xenosensing transcription factors regulating the induction of phase I and II xenobiotic-metabolizing enzymes as well as transporters in response to exogenous stimuli. It has now become clear, however, that these receptors cross-talk with endogenous stimuli as well, which extends their regulation to various physiological processes such as energy metabolism and cell growth. As recognition of the function of these receptors has widened, the molecular mechanism of their regulation has evolved from simple protein-DNA binding to regulation by complex protein-protein interactions. Novel mechanisms as to how xenobiotic exposure alters hepatic metabolic pathways such as gluconeogenesis and beta-oxidation have emerged. At the same time, the molecular mechanism of how endogenous stimuli, such as insulin, regulate xenobiotc metabolism via CAR and PXR have also become evident.

The effects of recombinant interleukin 2 on HBe antigen positive chronic hepatitis B.

Recombinant interleukin (IL-2) was administered to 16 patients with HBe antigen-positive chronic active hepatitis in which the diagnosis was ascertained histologically. In 7 of the 16 patients, a decrement of the serum HBe antigen value was observed (Group A). In group A, the findings showed an increment of peripheral Leu 11-positive cells and NK and LAK cell activity, an acute exacerbation during and after IL-2 administration, disappearance of HBc antigen observed in liver biopsy histology, and decrement of serum DNA-p activity. However, seroconversion of HBs antigen was not observed and no case showed the elimination status of continuous HB virus infection. On the other hand, in the other 9 patients (Group B), these changes were not observed and the existence of a HLA type difference between Group A and B was shown by HLA analysis. These results indicated that the immune responses mediated by IL-2 may play an important role in the development of chronic hepatitis B, and these results may be regulated genetically.
