Angiolipoma in brothers.

Two brothers with angiolipoma are presented. The patients, 33- and 37-year-old men, have multiple subcutaneous nodules on the trunk and extremities. Skin biopsies have confirmed the diagnosis of angiolipoma.

A case of Schönlein-Henoch-purpura: an immunopathological study.

A case of Schonlein-Henoch-purpura is presented. The immunofluorescent study of skin lesions of the patient showed granular deposits of IgA and C3 in the blood vessel walls perivascular deposits of plasminogen and diffuse localization of fibrin and fibrinogen in the upper dermis. Complement activation via the alternative pathway through IgA, C3 and plasminogen deposits was suggested.

Corticocerebellothalamic degeneration with chromatolytic neuronal swelling: a light and electron microscopic case study.

A 50-year-old woman showing psychoorganic syndrome, cerebellar symptoms, choreatic involuntary movements, Romberg's sign and epileptic fits over a period of 14 years, was neuropathologically studied. The calcarine and postcentral cortex, the medio-dorsal nucleus of the thalamus and the cerebellar cortex were affected with neuronal loss and astrocytic gliosis. Neurons in various regions showed chromatolytic neuronal swelling. Ultrastructurally, these chromatolytic neurons contained a large hollow core with numerous mitochondria, dispersed rough endoplasmic reticulum, degenerated cisternal structures and a few neurofilaments. Some neuronal mitochondria showed membranous trans-somal bridges composed of about 60 A parallel filaments with a spacing of about 80 A.

Immunosupressive activity of the basic protein from tumor tissue.

The basic protein prepared from AH-130 ascites hepatoma cells was tested for its effect on the cell-mediated immune response of Donryu strain rats. The basic protein suppressed the response of peripheral and lymph node lymphocytes to phytohemagglutinin and lymphocyte trapping in the antigen-draining lymph node. These results indicated that the basic protein from tumor tissue possibly had an immunosuppressive activity and affected inadequately the cell-mediated immune response as usually observed in tumor-bearing hosts.

Carcinogenesis in tissue culture. 28. Comparison of various effects of a chemical carcinogen, 4-nitroquinoline I-oxide, on normal human cells and on normal mouse cells in culture.

A comparison of the effects on cultured cells of a chemical carcinogen, 4-nitroquinoline 1-oxide (4NQO), between normal human cells which were resistant to malignant transformation with 4NQO and normal mouse cells rather easily transformable was carried out. We studied the following effects of 4NQO on normal human and normal rodent cells; 1) cytotoxicity, 2) DNA, RNA and protein synthesis, 3) incorporation of 4NQO into cells and time course changes of the drug bound with macromolecular substances in cells, 4) DNA repair synthesis, and 5) chromosomal changes. Our results demonstrated that there were no differences in cytotoxicity and inhibition of cellular macromolecular syntheses between human and mouse cells. On the other hand, significant differences were noted in DNA repair synthesis and chromosomal aberrations between human and rodent cells. These differences suggest that mouse cells are easily transformed into neoplastic cells with chemical carcinogens as compared with human cells.

Chlorpromazine induces population increase of synaptic vesicles in the rat hypothalamic ventromedial nucleus.

The population of synaptic vesicles per synaptic area in the hypothalamic ventromedial nucleus of rats treated with chlorpromazine (CP) for 3 days was compared with that of controls. In type I synapse, which contained 400-800 A small spherical vesicles (s-vesicles), together with 800-1200 A large spherical dense-cored vesicles (lc-vesicles), the population of s-vesicles increased 57.8% in the CP rats, In type II synapse, which contained only s-vesicles, the rate of increase among CP animals was 15.4%. Findings suggested that CP blocks the release of the vesicles from the presynaptic site or inhibits the transport function for biogenic amines through the presynaptic membrane, and type I synapse may be aminergic.

Characteristics of hematopoietic cell line established from human myelomonocytic leukemia.

The peripheral blood of an acute myelomonocytic leukemia patient has been cultured for 16 months. The culture is at present at the 140th population doupling level. The cultured cells have the characteristics of so-called lymphoblastoid cells and proliferate actively as individual cells in small clusters, or in large clumps consisting of large mononuclear cells. Some of these cells appeare to be lymphoid, but the majority are immature mononuclear cells with a tendency to lobulate. They gave a weakly positive peroxidase reaction at the beginning of cultivation, and have given a strongly positive esterase reaction persistently. The cytoplasm shows ciliary or tail-like projections as the cell matures. Complement (C3) receptor and IgG receptors are found on the cell surface, and active phagocytosis is mannifest. Colloidal iron particles or viable red blood cells attached to the cell membrane suggesting possible differentiation to reticulum cells or macrophages. The cultured cells are mostly diploid but some cells show chromosome abnormality. Herpes type virus was foun in the nucleus, cytoplasma and on the cell membrane. The transplanatation of cultured cells to the cheek pouch of hamsters produced small tumors with histological findings resembling reticulum cell sarcoma.

A neuropathological case-study of myoclonus epilepsy.

A 28-year-old male of progressive myoclonus epilepsy was reported, who had showed a gradual progression in myoclonus, mental retardation and cerebellar symptoms, and had been treated with a large dosage of diphenylhydantoin. Neuropathologically, slight degenerative changes of the cerebrl cortex, especially in the IV layer, the external pallidum, and the dentate and olivary nuclei were observed. The most obvious change was diffuse reduction of Purkinje's and granular cells in the cerebellum. A congenital cyst was found with surrounding gliosis in the central tegmental tract of the pons. A significant relationship between myoclonus and the cyst was proposed, and furthermore, influences of diphenylhydantoin intoxication on the clinicopathological development of myoclonus epilepsy were emphasized.

Effect of oil-attached BCG cell wall on the kinetics of lymphocytes in the tumor-draining node.

The lymphocyte distribution into the tumor-draining node was studied with AH-130 hepatoma cells and Donryu strain rats in relation to the adjuvant activity of the oil-attached BCG whole cell wall. The mixed inoculation of the oil-attached BCG cell wall with tumor cells resulted initially in further augmentation of increased distribution of 51Cr-labeled lymphocytes into the draining-node induced by inoculation of the tumor cells alone, and secondarily in the systemic stimulation of response of the lymph node lymphocytes to phytohemagglutinin. Suppression of the inoculated tumor growth and lymph node metastasis was finally observed. These results were discussed in connection with the therapeutic effect of BCG and its cell wall fraction.

Establishment and some biological characteristics of human hepatoma cell lines.

Two cell lines of human hepatoma, HLE and HLF lines, were established in vitro from the hepatocellular carcinoma of a 68-year-old patient. One clone (HLEC) was obtained from a single HLE cell. The cells of HLE and HLEC were epithelial-like and both of these cells demonstrated glycogen granules in the cytoplasm when stained with periodic acid and Schiff reagent. Although HLF cells resembled fibroblasts in morphology, they appear to have originated from hepatoma cells, judging from epithelial characteristics in aggregates reconstituted by rotation culture and heterotransplantability. HLE cells produced alpha-fetoprotein until day 187 of culture, but HLF cells did not produce alpa-fetoprotein at any period examined. Chromosome number of both cell lines was distributed near the triploid range. HLF cells were transplantable into the cheek pouch of adult hamsters treated with cortisone acetate, but not HLE cells.
