Microinjection of neurotensin into the CNS induces hyperdipsia in the rat.

Neurotensin (NT) is a neuropeptide and putative neurotransmitter that has been shown to exert a variety of effects on digestive and ingestive processes. In order to address the possibility that NT might play a role in the regulation of water intake as well, the peptide was infused into the lateral cerebral ventricle, amygdala, ventral tegmental area, lateral hypothalamus, and preoptic area of the anterior hypothalamus of rats deprived of water for 16 hours. Neurotensin produced a significant and dose-dependent increase in water intake when injected into the ventricular system but had no effect when it was applied to the other brain sites. It was concluded that this peptide may play a physiological role in the control of water ingestion and that central sites of action remain to be determined.

Anticoagulant effect of sulphated poly/vinyl alcohol-acrylic acid/copolymers.

Copolymers of poly/vinyl alcohol-acrylic acid/ with various content of sulphate and carboxyl groups have been synthetized and tested for their in vitro effect on blood coagulation. The results indicate that the sulphated copolymers display an inhibitory effect but there is a requirement in the charged groups of about 20% in the molecule to possess effective anticoagulation. The biochemical mechanism of their actions is complex, i.e. the inhibition of blood clotting is a consequence of both the accelerated inactivation rate of thrombin by antithrombin-III and a direct inhibitory effect on the thrombin-fibrinogen reaction. Moreover, additional effects may occur on other blood coagulation enzymes than thrombin, depending on the chemical composition of the copolymers.

A deletion that includes the segment coding for the signal peptidase cleavage site delays release of Saccharomyces cerevisiae acid phosphatase from the endoplasmic reticulum.

We studied ultrastructural localization of acid phosphatase in derepressed Saccharomyces cerevisiae cells transformed with a multicopy plasmid carrying either the wild-type PHO5 gene or a PHO5 gene deleted in the region overlapping the signal peptidase cleavage site. Wild-type enzyme was located in the cell wall, as was 50% of the modified protein, which carried high-mannose-sugar chains. The remaining 50% of the protein was active and core glycosylated, and it accumulated in the endoplasmic reticulum cisternae. The signal peptide remained uncleaved in both forms. Cells expressing the modified protein exhibited an exaggerated endoplasmic reticulum with dilated lumen.

[Isolation and characterization of dipeptidylpeptidase IV from human seminal plasma].

Two forms of dipeptidylpeptidase IV with molecular weights (gel distribution chromatography) of 270 kD and about 1450 kD were isolated from human seminal plasma. The enzymes migrating differently in electrophoresis possess identical catalytic properties in regard to pH optimum, kinetic constants and the influence of effectors. The high-molecular form loses at 65 degrees C its enzymatic activity significantly more slowly. After 60 min of incubation at 68 degrees C, human seminal plasma has still about 12-32% (N = 50) dipeptidylpeptidase activity. Through the action of Triton X-100 or chymotrypsin the high-molecular form can be transferred into the low-molecular one without loss of activity. In 80 healthy human sperm donors the average proportion of the low-molecular form of peptidase was determined to be 9-22%.

The relationship between the growth retardative effect of CCC and ethylene production.

The ethylene production of the hypocotyls of CCC-treated bean plants was studied, and it was concluded that the treatment induced changes in the quantity of ethylene produced by the apical and basal hypocotyl parts. The ethylene production of the basal hypocotyl parts showed considerable increase on the effect of the treatment, in comparison with the control. The obtained results suggest a possible relationship between the longitudinal-growth inhibiting, stem-thickness inducing, root-formation stimulating effects of CCC and the effect exerted on ethylene production.

Aetiological monitor of congenital abnormalities: a case-control surveillance system.

An Aetiological Monitor of Congenital Abnormalities was launched in Hungary on 1 January 1980 with the purpose to obtain aetiological information parallel with the reporting of congenital abnormalities. A questionnaire is sent to all mothers having babies with well defined major congenital abnormalities within one month after being reported to the Register. Diseases during pregnancy, drugs taken during pregnancy as well as employment and type of work made by the pregnant are studied. According to the data of the first year, progestogens were taken in 33.4% and oestrogen in 10.8% of all pregnancies. Other data indicate that the drugs studied mean either no teratogenic risk or, if there is any risk, it is very small.

Nuclear localization of aspartate transcabamoylase in Saccharomyces cerevisiae.

The cytochemical technique using the in situ precipitation of orthophosphate ions liberated specifically by the aspartate carbamoyltransferase (ATCase) (EC 2.1.3.2) reaction indicated that in Saccharomyces cerevisiae this enzyme is confined to the nucleus. This observation is in accordance with the result reported by Bernhardt and Davis (1972), Proc. Natl. Acad. Sci. U. S. A. 69:1868-1872) on Neurospora crassa. The nuclear compartmentation was also observed in a mutant strain lacking proteinase B activity. This finding indicates that this proteinase is not involved in the nuclear accumulation of ATCase, and that the activity observed in the nucleus corresponds to the multifunctional form associated with the uracil path-specific carbamoylphosphate synthetase and sensitive to feedback inhibition by UTP. In a ura2 strain transformed by nonintegrated pFL1 plasmids bearing the URA2-ATCase activity encoding gene, the lead phosphate precipitate was observed predominantly in the cytoplasm. This finding enhances the reliability of the technique used by eliminating the possibility of an artifactual displacement of an originally cytoplasmic reaction product during the preparation of the material for electron microscopy. On the other hand, nuclei isolated under hypoosmotic conditions do not exhibit the ATCase activity that is recovered in the cytosolic fractions after differential centrifugation of the lysate in Percoll gradient. A release of the protein from the nuclei during the lysis step, consistent with its nucleoplasmic localization, is postulated.

Studies on purine transport and on purine content in vacuoles isolated from Saccharomyces cerevisiae.

The transport of purine derivatives into vacuoles isolated from Saccharomyces cerevisiae was studied. Vacuoles which conserved their ability to take up purine compounds were prepared by a modification of the method of polybase-induced lysis of spheroplasts. Guanosine greater than inosine = hypoxanthine greater than adenosine were taken up with decreasing initial velocities, respectively; adenine was not transported. Guanosine and adenosine transporting systems were saturable, with apparent Km values 0.63 mM and 0.15 mM respectively, while uptake rates of inosine and of hypoxanthine were linear functions of their concentrations. Adenosine transport in vacuoles appeared strongly dependent on the growth phase of the cell culture. The system transporting adenosine was further characterized by its pH dependency optimum of 7.1 and its sensitivity to inhibition by S-adenosyl-L-methionine. In the absence of adenosine in the external medium, [14C]adenosine did not flow out from preloaded vacuoles. However, in the presence of external adenosine, a very rapid efflux of radioactivity was observed, indicating an exchange mechanism for the observed adenosine transport in the vacuoles. In isolated vacuoles the only purine derivative accumulated was found to be S-adenosyl-L-homocysteine.
