Es-6, a further polymorphic esterase in the rat.

A further polymorphic rat esterase with broad tissue expression and restricted substrate specificity is described and tentatively called Es-6. Inbred rat strains have either fixed allele Es-6F or fixed allele Es-6S. Es-6 is not linked to the established esterase cluster consisting of the eight esterase loci Es-1, Es-2, Es-3M, Es-4M, Es-4W, Es-5 (= Es-3W), Es-7, and Es-8 in LG V of the rat or to RT1, Gc, c, a, and h. Esterases with apparently identical biochemical and genetical characteristics are Es-17 of the mouse and Es-A4 of humans.

[Crohn's disease in the stomach and duodenum, a diagnostic problem (author's transl)].

Six cases reports are given of patients with Crohn's disease localized in the stomach or the duodenum, and problems of diagnosis are discussed. In many cases the disease cannot be differentiated from other inflammatory conditions or from gastric carcinoma by means of radiological or endoscopic examination. Careful search for other localisations of the disease, and multiple, accurate biopsies as well as histological examination will allow to establish the diagnosis. If this cannot be done by the before mentioned means, laparotomy has to be performed in order to allow removal of enough tissue for extended histological examination needed to rule out malignancy.

A sensitive method for determination of conjugated catecholamines in blood plasma.

Conjugated catecholamines in blood plasma of blood donors were determined from the difference between the total and the free catecholamines. Total catecholamines were measured after complete hydrolysis of the conjugated amines (heating of acidified samples for 40 min at 95 degrees C). Free catecholamines were determined in untreated plasma. Determinations were performed radioenzymatically. We combined and modified the procedures of Peuler & Johnson (1977) Life Sci 21, 625-636) and Da Prada & Zürcher (1976) Life Sci 19, 1161-1174). The resulting method was less laborious and cheaper.

[The antistreptolysin reaction with and without dextran sulfate addition in various diseases].

It is reported on the results of determinations of the antistreptolysin titre in patients with infectious hepatitis, renal and essential hypertension, mitral stenosis, acute tonsillitis, scarlet fever, rheumatic fever as well as rheumatoid arthritis according to the usual and the dextran sulphate absorption method in altogether 739 patients. Here as in a former publication in 1,700 normal persons partly considerable significant differences between the antistreptolysin titre with and without dextran sulphate were found. The two techniques are discussed, also the influence of the beta-lipoproteins with their proportion of phosphatide on the result of the antistreptolysin titre. It is possible to absorb beta-lipoproteins and perhaps also phosphatides with the help of the addition of dextran sulphate. The present investigations show unequivocally that the absorption the dextran sulphate is a necessary demand which saves the clinician from diagnostic and therapeutic errors. The antistreptolysin reaction is to be regarded only as one constituent for the clinical diagnosis and must not be overvalued in its importance.

Mechanism of inhibition of dopamine beta-hydroxylase evoked by FLA-63. An in vitro study.

Bis-(4-methyl-1-homopiperazinylthiocarbonyl)-disulphide (FLA-63) inhibits in vitro purified bovine adrenal medullary dopamine beta-hydroxylase (BDH) concentration-dependently; 50% inhibition is produced by 2 x 10(-6)M FLA-63. Spectrophotometric experiments show that FLA-63 is reduced by ascorbate very likely to the corresponding dithiocarbamate-derivative, which acts as a Cu++-chelator. Cu++-ions are able to completely abolish the inhibition of the DBH caused by FLA-63 in concentrations up to 2.5 x 10(-6)M. It is concluded that FLA-63 inhibits the DBH mainly in its reduced form via the formation of a chelate-complex with copper ions derived from the DBH.
