A highly-sensitive sandwich enzyme immunoassay for serum dipeptidyl aminopeptidase IV (DAP IV) was developed; its level in sera of patients with oral cancer [1.50 +/- 0.35(SD)mg/l serum] was significantly lower than in normal subjects [2.27 +/- 0.77(SD)mg/l serum].
Complex protein mixtures of unstimulated human sublingual and submandibular saliva were fractionated by two-dimensional (2-D) gel electrophoresis, visualized by silver staining and then analysed by immunostaining. Specific proteins were identified by incubation with specific antibody and peroxidase-conjugated second antibody (Western blot). Electrophoresis and silver staining revealed over 50 protein components in 2 microliter of unconcentrated mixture. The Western-blot technique allowed detection of protein spots of plasma origin when an antibody against whole serum was used, but only the albumin spot could be found. Albumin, secretory IgA, acid phosphatase and alpha-amylase were identified with specific antibodies.
Using this method, without denaturing agents, over 60 protein components in 2 microliter (about 4 micrograms of protein) of unconcentrated parotid saliva could be detected. After two-dimensional separation, the spots of albumin, secretory IgA, IgG, acid phosphatase, amylase and plasma-originating protein were identified by antibodies with the Western-blot technique.
A new Japanese nickel-titanium (NiTi) alloy wire was developed by the Furukawa Electric Co., Ltd. of Japan. This wire was subjected to uniaxial tensile testing and a specially designed three-point bending test to determine the wire stiffness, and to evaluate spring-back, shape memory, and super-elasticity. The Japanese NiTi wire exhibited an unusual property termed "super-elasticity," which no other orthodontic wire has shown. This phenomenon was researched thoroughly. The wire delivered a constant force over an extended portion of the deactivation range. Among all the wires compared, Japanese NiTi alloy wire was the least likely to undergo permanent deformation during activation. The new alloy exhibited a specific stress-strain curve unlike those of the other tested materials. Stress remained nearly constant despite the strain change within a specific range. This unique feature is the manifestation of so-called super-elasticity. Heat treatment enabled the load magnitude at which super-elasticity is reflected to be influenced and controlled by both temperature and time. A unique and useful process was also developed so that an arch wire delivering various magnitudes of force for a given activation could be fabricated from the wire of the same diameter. The clinical application of wires of this new alloy should be more likely to generate a physiologic tooth movement because of the relatively constant force delivered for a long period of time during the deactivation of the wire. Japanese NiTi alloy should be considered an important material addition to clinical orthodontic metallurgy.
At physiological pH, the hydrolytic activity of purified bovine pulp alkaline phosphatase toward phosphorus compounds was observed to be in the order of inorganic pyrophosphate greater than beta-glycerophosphate greater than phosphorylcholine greater than p-nitrophenylphosphate greater than glucose-6-phosphate. Optimum pH of the enzyme toward inorganic pyrophosphate was shown to be 8.5, with around 60% of the activity at pH 7.5. The activity was increased by the addition of Mg2+, but a different pattern of activation was observed between pH 7.5 and 8.5.
Ampullarius canaliculatus is a large fresh-water snail of which the original habitat is said to be Argentina. Recently, the snail which grew wild has propagated in various parts of Japan, and is causing damage to aquatic plants by feeding on them. The present study was performed to find out whether the snail can be a suitable intermediate host for Angiostrongylus cantonensis. Natural infection with A. cantonensis was confirmed in the snails collected from 5 different places in 4 hamlets in Okinawa and in those from Ishigaki Island. All snails experimentally infected with first-stage larvae of the parasite showed second-stage and third-stage larvae developing at the time of examination later on. Thus, A. canaliculatus is a suitable intermediate host for A. cantonensis, and, if eaten raw, it can be a potential source of human infection with eosinophilic meningoencephalitis in the endemic areas.
Streptococcus mitis contains two arginine aminopeptidases (I and II) as an arginine-supplying system and the arginine deiminase pathway as an arginine-utilizing system. The levels of arginine aminopeptidase I and three enzymes of the arginine deiminase pathway were suppressed by glucose in an apparently coordinate manner. Enzyme II appeared to be constitutive.
Kinetic studies of pig kidney dipeptidyl peptidase IV (dipeptidyl-peptide hydrolase, EC 3.4.14.5) were carried out using substrates possessing a side-chain of different length at the P2 position (or amino-terminal position in this case) such as Lys-, Arg-, Phe-, Met-, Ser-, His-, Glu- and Gly-Pro-pNA. The hydrolytic coefficient (Kcat/Km) has determined in the order Met- greater than Glu- greater than Ser- greater than His- greater than Phe- greater than Lys- greater than Gly- greater than Arg-, indicating a gradual increase with elongation of the side-chain from 0.03 to 0.60 nm followed by a decline when side-chain length approached 0.70 nm. Thus, the most probable depth of the side-chain pocket at the S2 subsite of the enzyme is proposed to be 0.50-0.60nm.
UNLABELLED: We tried an in vitro sensitivity test for the purpose of examining the effect of anti-neoplastic agents and their enhancement by Biscolaurine Alkaloid (Cepharanthin). Methods, HeLa cells were synchronized by excess of thymidine and hydroxyurea, and resuspended in Eagle's MEM with 10% FCS. The cells were mixed with Mitomycin C or Bleomycin and added to the wells of a microtiter plate. When required, Cepharanthin (the effect of this drug is considered to change the plasma membrane liquidity) was added to the medium, and then cultured for 24 hours in a CO2 incubator. After culture, each well received 3H-thymidine, and was incubated for 8 hours. The cells were harvested for the assay of 3H-thymidine. RESULTS: MMC or BLM suppressed the uptake of 3H-thymidine in relation to its concentration. MMC or BLM with various concentrations of Cepharanthin showed strong suppression compared with the same concentrations of MMC or BLM alone. From the above results, this experimental method is regarded as an useful model for selecting sensitive anti-neoplastic agents. In addition, Cepharanthin showed enhancing effects on anti-neoplastic agents.
Tripteroides aranoides was colonized in the laboratory. Total duration of the immature stages was ca. 3 weeks at 28 degrees C, L:D = 15.5:8.5 with an ample food supply. Retardation of 4th instar development was observed in larvae fed on insufficient food. Females were autogenous for the first clutch of eggs and required a blood meal for maturation of the second clutch. Mating was initiated in flight and copulation occurred on the cage wall. Gravid females hovered in small oblique loops above water in bamboo cups, whereupon a white egg appeared at the abdominal tip, which was propelled by the swing of the abdomen towards water surface. The females propelled eggs in the same manner into small apertures (11 x 4 mm) bored in bamboo.

A sensitive sandwich enzyme immunoassay for tyrosine hydroxylase (TH) from bovine and human adrenals has been developed. Anti-TH antibody was prepared from bovine adrenal TH. The assay system consisted of an antibody F(ab')2 immobilized on polystyrene beads as a solid phase and of beta-D-galactosidase-conjugated antibody. This method was highly sensitive and specific for the assay of TH. Human adrenal TH level was determined by similar sensitivity as bovine adrenal TH, suggesting the presence of common antigenic sites between human and bovine adrenal enzymes. The presence of inactive or less active forms of TH in human adrenals was revealed by purification of the enzyme and monitoring with this enzyme immunoassay as well as with enzyme activity assay.
The fluorescence activated cell sorter (FACS) was used with an indirect membrane immunofluorescence technique to detect antibody against the Forssman antigen, a glycosphingolipid. Sheep erythrocytes, which contain Forssman antigen as a major membrane glycosphingolipid, were used as the target antigen. Detection of the anti-Forssman antibody on the sheep erythrocytes was done with specific fluorescein-conjugated second antibody and analyzed on a FACS. Compared to other available methods, analysis with the FACS was simple, sensitive, reproducible and quantitative. More than 250 pg of antibody could be detected. In addition, as little as 1 ng of Forssman antigen could be estimated by a binding inhibition experiment.
Rat erythrocytes contained ganglio-series gangliosides, GM1, fucosyl GM1, and GD1a, in a high concentration. The concentrations of GM1, fucosyl GM1, and GD1a in rat erythrocyte ghosts were 889.0 nmol, 470.6 nmol, and 462.0 nmol per g dry weight, respectively, and the molar ratio of lipid-bound sialic acid, cholesterol and lipid-bound phosphorus was 3.1:73.9:100.0. The reactions of fucosyl GM1 and GM1 on rat erythrocytes with rabbit anti-fucosyl GM1 and anti-GM1 antisera were measured by means of haemolysis in the presence of complement and a binding assay of antibodies with a FACS after staining erythrocytes by the indirect membrane immunofluorescence technique. When measured by ELISA, anti-fucosyl GM1 antiserum was found to react almost exclusively with fucosyl GM1 with a slight cross-reaction with GM1, but anti-GM1 antiserum cross-reacted to a significant extent with asialo GM1. Rat erythrocytes were haemolyzed specifically with anti-fucosyl GM1 antiserum, but not with antisera to GM1, asialo GM1, asialo GM2, Forssman and globoside, and the haemolysis was proved to be definitely caused by the specific recognition of fucosyl GM1 on rat erythrocytes by anti-fucosyl GM1 antibody according to the haemolysis inhibition reaction using various glycosphingolipid-containing liposomes as inhibitors. In addition, although the binding of anti-fucosyl GM1 antibody on rat erythrocytes was clearly demonstrated with a FACS, anti-GM1 antibody did not bind. The observations that the haemolysis of rat erythrocytes and the binding of antibody to rat erythrocytes were found only with anti-fucosyl GM1 antiserum, and not with anti-GM1 antiserum, but that nevertheless the titer of anti-GM1 antiserum was higher than that of anti-fucosyl GM1 antiserum and GM1 on rat erythrocytes was more abundant in concentration than fucosyl GM1, seem to be a matter of great importance in assessing the specificity of anti-ganglioside antibody and the surface distribution of gangliosides on the cell.
Analysis of epidemiological and meteorological data for Nagasaki Prefecture, southwest Japan, for the period 1950-1979, showed a correlation between size of epidemics of human Japanese encephalitis (JE) and weather factors. It was demonstrated that during the period 1950-1969 the epidemic size (ES) had two types of correlation: ES correlated inversely with total summer precipitation, and ES correlated directly with mean summer temperature. The fact that in the 1970s there were fewer cases of JE cannot be attributed to changes in the weather factors. Large epidemics occurred in summers with low precipitation and high temperatures, while the epidemic size was small when there was heavy precipitation and temperatures were low. The two weather factors jointly explain 0.53 of the observed variance in annual JE ES. Furthermore, they strongly correlate with each other, and total summer precipitation alone is sufficient to explain about one-half of the total variance. How these factors influence the number of JE cases is discussed in relation to the population dynamics of the vector mosquito, Culex tritaeniorhynchus.
