Ontogeny of sulphated glycoconjugate-producing cells in the rat fundic gland.

The ontogeny of sulphated glycoconjugate-producing cells in the rat fundic gland has been studied using high iron diamine (HID), Alcian Blue (AB) at pH 1.0, high iron diamine in combination with Alcian Blue at pH 2.5 (HID-AB), cationic colloidal gold (CCG) at pH 1.0 under light microscopy and CCG (1.0), HID-thiocarbohydrazide (TCH)-silver proteinate (SP)-physical development (PD) under electron microscopy. From day 19.5 of gestation, sulphated glycoconjugate-producing cells were discernible under both light and electron microscopy. The development of such cells can be classified into four stages: (1) a prenatal period from day 19.5 of gestation extending to 0.5 days after birth; (2) 1 day to 2 weeks after birth; (3) 2 to 4 weeks after birth; and (4) the final period from 4 to 8 weeks after birth. Glycoconjugate-producing cells reached maturity by 4 weeks after birth. Our results indicated that glycoconjugate-producing cells were cells along the wall of foveolar lumen, but not those covering the gastric mucosa surface. Our results also suggested that the trans to transmost Golgi apparatus lamellae were the sites of sulphation in the developing rat stomach.

Immunohistochemical demonstration of prostaglandins E2, F2 alpha, and 6-keto-prostaglandin F1 alpha in rat dental pulp with experimentally induced inflammation.

Using formalin-fixed and EDTA-decalcified cryostat sections, the immunohistochemical localization of prostaglandin (PG) E2, PGF2 alpha, and 6-keto-PGF1 alpha (a stable metabolite of PGI2) was examined in normal rat and inflamed dental pulp. Inflammation was induced by opening the pulp chamber. There was no immunoreactivity for prostaglandins in normal dental pulp, whereas positivities for PGE2, PGF2 alpha, and 6-keto-PGF1 alpha were demonstrated in the cytoplasm of macrophages and endothelial cells in the inflamed dental pulp. In addition to these cells, numerous pulp cells and odontoblasts existing in the inflamed pulp and its apical noninflamed area also were intensely stained for PGF2 alpha. Such an area with positive cells gradually extended in an apical direction with the progression of inflammation. These findings suggested that PG production from these host cells is involved in development of inflammation of rat dental pulp.

Immunohistochemical detection of prostaglandins E2, F2 alpha, and 6-keto-prostaglandin F1 alpha in experimentally induced periapical inflammatory lesions in rats.

The immunohistochemical localization of prostaglandin (PG) E2, PGF2 alpha, and 6-keto-PGF1 alpha (a stable metabolite of PGI2) was demonstrated in rat periapical inflammatory lesions induced by opening the pulp chamber. Two wk postoperatively, suppurative periapical lesions were formed, and active bone resorption was seen surrounding these lesions. Immunohistochemical examination showed that macrophages infiltrating in inflammatory tissue were positively stained for the examined PGs. In some lesions, wherein acute inflammatory changes subsided and proliferation of fibroblasts started, the fibroblasts were positively stained for 6-keto-PGF1 alpha. Osteocytes and osteoblasts were also positive for 6-keto-PGF1 alpha not only in experimental animals, but also in untreated animals. However the staining intensity of the PG in these cells was higher in periapical lesions than in normal condition. These findings suggested that the cellular sources of the PGs in the periapical lesions are mainly macrophages and fibroblasts, and that the PGs produced by these cells, and possibly osteoblast and osteocytes, may contribute to the osteolytic resorption of periapical lesions.

Immunohistochemical observations on a possible ameloblastic fibro-odontoma.

An ameloblastic fibro-odontoma which occurred in the mandible of a 3-year-old Japanese boy is reported together with immunohistochemical findings. Histologically, the tumor consisted of an ameloblastic fibroma-like area and some typical complex odontoma-like areas. The epithelial islands in the ameloblastic fibroma-like area showed different developmental stages of the epithelial-connective tissue interface. Immunohistochemical examination revealed that all epithelial components in the ameloblastic fibroma-like area showed expression of CK 8, CKs 13, 16, CK 14, CK 18 and CK 19, and coexpression of these cytokeratins and vimentin. These findings suggest that even the epithelial component without obvious epithelial-mesenchymal induction showed the final cell differentiation of the enamel organ with the potential for epithelial-mesenchymal induction.

F1-ATPase alpha-subunit made up from two fragments (1-395, 396-503) is stabilized by ATP and complexes containing it obey altered kinetics.

Inferred from the crystal structure of mitochondrial F1-ATPase (Abrahams, J.P. et al. (1994) Nature 370, 621-628), the proteinase-sensitive region around Phe-395 of thermophilic F1-ATPase alpha-subunit corresponds to the loop which connects main part of the carboxyl-terminal helical bundle domain with the ATP binding domain. This loop is in contact with the gamma- and adjacent beta-subunits. Two polypeptides corresponding to the sequence 1-395 and 396-503 of the alpha-subunit were expressed in Escherichia coli cells and they were copurified as an apparently functional alpha-subunit (alpha(395/396)) made up of two polypeptides. The isolated alpha(395/396) was stabilized by ATP-Mg, but not by ADP-Mg, although it bound both ATP-Mg and ADP-Mg with similar affinities (Kd, 11 microM and 14 microM, respectively). The alpha(395/396) was reconstitutable into alpha(395/396)3 beta 3 and alpha(395/396)3 beta 3 gamma complexes. Different from the intact the ATP-Mg-induced dissociation into alpha 1 beta 1 heterodimers. ATP hydrolysis by the alpha(395/396)3 beta 3 gamma complex underwent a slow initial phase, whereas the intact alpha 3 beta 3 gamma complex exhibited an accelerated initial phase. Steady-state ATPase activity at various ATP concentrations showed negative cooperativity for the intact alpha 3 beta 3 gamma complex but apparently positive cooperativity for the alpha(395/396)3 beta 3 gamma complex. The ATPase activities at a saturating ATP concentration of the complexes containing the alpha(395/396) were 180% of those containing intact alpha-subunits. These results indicate that a loop around Phe-395 is involved in intersubunit interaction in F1-ATPase.

Changing pattern of mental health utilization in Hachijojima.

We have reviewed retrospectively 229 patients interviewed by the mental health team in the Hachijojima since the establishment of our services in 1987. The 6 year observation period was classified into three chronological stages for the purposes of identifying any change in the utilization pattern of the mental health services of the island. The rate of schizophrenia was observed to have decreased from 48% in the first 2 years of the study period to 28% in the last 2 years; while that of other mental disorders increased yearly. Interestingly, an increasing number of patients were referred to our services by professionals in areas other than mental health (from 42 to 77%), and patients without any history of psychiatric treatment were more frequently seen (from 36 to 75%) in the later part of the study period. These changes suggest the importance of access to mental health services.

Correlation between argyrophilic nucleolar organizer region (AgNOR) counts and histologic grades with respect to biologic behavior of salivary adenoid cystic carcinoma.

The quantification of argyrophilic nucleolar organizer regions (AgNOR) was performed in 34 cases of adenoid cystic carcinoma (ACC) to determine (1) whether AgNOR count correlates with its different histologic grades pertinent to prognosis, and (2) whether AgNOR counts can offer any additional prognostic advantage over histologic grading. According to Szanto et al.'s histologic grading criteria (4), 12 cases were Grade 1, 7 cases Grade 2, and 15 were Grade 3. Patients were divided into 20 favorable cases (without metastases) and 14 unfavorable cases (with metastases). Although most Grade 3 tumors had high AgNOR counts (> or = 4) and Grade 1 tumors with low (< 4) AgNOR counts outnumbered those with high AgNOR counts, considerable overlap of AgNOR values in different grades was observed. However, all unfavorable cases had high AgNOR counts regardless of their histologic grades, suggesting that the metabolic alterations associated with the malignancy level of ACC may partly be portrayed by the AgNOR count, irrespective of the histologic appearance. Cumulative survival rates of Grade 1 tumors and of tumors with low AgNOR counts were better than those of Grade 3 tumors and those with high AgNOR counts. Within the limited number of cases in this series the AgNOR count exhibits a potential for identifying some aggressive ACCs that cannot be detected by histology alone.

Florid cemento-osseous dysplasia with concomitant simple bone cysts: a case in a Japanese woman.

Florid cemento-osseous dysplasia (FCOD) with concomitant simple bone cysts is not common. We report a case of FCOD involving three quadrants of the jaws and associated with two large cystic lesions of the mandible in a 40-year-old Japanese woman. Microscopic examination revealed unencapsulated fibroblastic proliferation with formation of bone and cementum, showing different developmental stages, and cystic lesions resembling a simple bone cyst.

Periodontal tissue regeneration on the surface of synthetic hydroxyapatite implanted into root surface.

Several studies have demonstrated that periodontal ligament (PDL)-derived cells can form a new connective tissue attachment (NCTA) not only on denuded dentin but also on nondental inorganic materials. Although the cellular response to these materials may be different than that to denuded dentin, the process of NCTA formation on the surface of non-dental inorganic materials is still undetermined. In the present study, we compared the process of NCTA formation on the surface of an inorganic material with that on a denuded root surface by implanting a block of synthetic hydroxyapatite (HA) into a root cavity prepared on the first molar of a rat. We then observed tissue regeneration under conditions where the PDL-derived cells could populate the HA surface using an occlusive membrane. After fibrinous exudation at 3 days postoperatively, granulation tissue from the PDL filled the space between the HA block and the membrane at 1 week. Four weeks after the operation, although the granulation tissue increased in density at the interface to the HA block, no obvious deposition of cementum-like tissue was seen on the HA block. Six weeks after the operation, the deposition of cementum was observed on the surface of the HA block. The deposition of cementum became more extensive and insertion of collagen fibers into the cementum layer became more obvious 8 weeks postoperatively. Twelve weeks after the operation, the cementum covered almost the entire surface of the HA block. On the surface of the denuded dentin in control teeth, cementum deposition was seen at 4 weeks after the operation and increased in thickness from 6 weeks on.(ABSTRACT TRUNCATED AT 250 WORDS)

V-onc mutation associated with host cell growth in retroviral tumors.

In sarcomagenesis in rats infected neonatally with feline sarcoma virus (ST-FeSV), v-fes product (P85) was previously shown by us to be a predictive and preventive determinant. In order to explore the part played by P85 in tumor suppression, DNA was extracted from precancerous granulomas and from slow or rapid growing sarcomas induced by neonatal injection of the virus. The v-fes signal from extracted DNA was analyzed by PCR-SSCP. The prototype v-fes gene signal was detected in most lesions and found to be generally amplified in rapid growing sarcomas and in some granulomas. Several v-fes homologs showing varying mobilities in gel were seen in most sarcomas and some granulomas with or without the prototype v-fes signal. In slow growing sarcomas and granulomas induced in hosts that were immunized with ST-FeSV induced syngeneic sarcoma and proved to carry IgG antibody to P85, the prototype v-fes gene was found to be down-regulated and v-fes homologs were found to be reduced in number or eliminated. These results suggest that the development of v-fes mutations is associated with the growth potential of cells carrying the v-fes gene, and that host immunity to v-onc product influences the development of virogene rearrangements and results in slow and suppressed growth of tumors caused by neonatal infection with retrovirus.

Temperature-dependent change of serological specificity of Candida albicans NIH A-207 cells cultured in yeast extract-added Sabouraud liquid medium: disappearance of surface antigenic factors 4, 5, and 6 at high temperature.

The cells of Candida albicans NIH A-207 strain (A-strain) cultivated in YSLM at high temperatures (37 and 40 degrees C) did not undergo agglutination with the factor sera 4, 5, and 6 in a commercially available factor serum kit, 'Candida Check', and formed a grape-like shape. The mannans isolated from the cells had lost their reactivity against the factor sera in ELISA. It was also revealed by 1H NMR analysis that the mannans contained neither a phosphate group nor a beta-1,2-linked mannopyranose unit, although these mannans increased the non-reducing terminal alpha-1,3-linked mannopyranose unit. The cells and the mannans prepared by cultivation at such high temperatures followed by 27 degrees C in the same medium entirely recovered the reactivity with the factor sera.

Antigen expression associated with lymph node metastasis in gastric adenocarcinomas.

A total of 100 gastric adenocarcinomas, comprising 50 cases with lymph node metastasis and 50 cases without lymph node metastasis, were examined for immunohistochemical reactivity with the monoclonal antibody to urokinase-type plasminogen activator (u-PA), Lex related 4C9 antigen, Jun, or to nucleobindin (Nuc). In tumors with lymph node metastasis, 41 (82%) were positive for u-PA and 28 (56%) were positive for Nuc. In tumors without lymph node metastasis, 26 (52%) were positive for u-PA and five (10%) were positive for Nuc. The percentage of cases positive for u-PA or Nuc was significantly higher in tumors with lymph node metastasis than that in tumors without lymph node metastasis (P < 0.01). The expression of u-PA was found to be significantly correlated with that of Nuc (P < 0.001), mode of infiltrative growth (P < 0.05), depth of invasion (P < 0.01), and grade of lymphatic invasion (P < 0.01). However, the expression of Nuc was found to be significantly correlated with the expression of Jun (P < 0.05), depth of invasion (P < 0.01), and grade of lymphatic invasion (P < 0.001). These results suggest that immunohistochemical examination for the expression of u-PA and Nuc in tumor cells may help evaluate the potential of adenocarcinomas of the stomach for lymph node metastasis.

Prognostic value of argyrophilic nucleolar organizer regions (AgNOR) count in adenoid cystic carcinoma of salivary glands.

A retrospective study was performed on 34 patients with adenoid cystic carcinoma (ACC) to see if quantification of argyrophilic nucleolar organizer regions (AgNOR) was correlated with their prognosis. Mean follow-up period was 113 months. According to detailed clinical data that included tumor recurrences, metastases and patient survival conditions, two groups were classified. Group 1 consisted of 20 live patients without metastases, of whom 16 were free of disease, while group 2 comprised 14 patients with metastases among whom 12 died of tumor. The silver staining technique was applied to paraffin embedded specimens. In each case, 300 nuclei were randomly examined and the mean AgNOR per nucleus was calculated. All patients from group 2 had mean AgNOR counts greater than 4, whereas 65% of patients from group 1 had mean AgNOR counts less than 4. In addition, statistical analysis showed that the pooled mean AgNOR count in group 2 was significantly higher than in group 1 (P < 0.01). The present results suggest that silver staining technique promises to be a useful supplementary method for prognostic evaluation of salivary ACC, and may be performed before planning the treatment.

Structural modification of cell wall mannans of Candida albicans serotype A strains grown in yeast extract-Sabouraud liquid medium under acidic conditions.

The cell wall mannans of two Candida albicans serotype A strains, NIH A-207 and J-1012 (abbreviated as A and J strains, respectively), cultured in yeast extract-Sabouraud liquid medium at pH 2.0, contained neither a phosphate group nor a beta-1,2-linked mannopyranose unit (H. Kobayashi, P. Giummelly, S. Takahashi, M. Ishida, J. Sato, M. Takaku, Y. Nishidate, N. Shibata, Y. Okawa, and S. Suzuki, Biochem. Biophys. Res. Commun. 175:1003-1009, 1991). In this study, the mannans obtained from A and J strains grown in pH 2.0 medium (abbreviated as mannans A2 and J2, respectively) exhibited quite different reactivities against rabbit anti-C. albicans and anti-Saccharomyces cerevisiae sera compared with those of mannans from the corresponding strains cultured in conventional medium at pH 5.9 (abbreviated as mannans A and J, respectively). Namely, mannans A2 and J2 lost reactivity against the former serum but reacted with the latter serum to a higher extent than mannans A and J. In order to account for these difference in more detail, mannans A2 and J2 were subjected to acetolysis. Elution profiles of the acetolysates were completely different from those of acetolysates obtained from mannans A and J reported in our previous papers. The 1H nuclear magnetic resonance spectra of the oligosaccharides from mannans A2 and J2 obtained by this procedure indicate that the side chains are composed of alpha-linked mannopyranose units densely linked to the alpha-1,6-linked backbone. The long side chains containing one alpha-1,3-linked mannopyranose unit are markedly increased.

Contingent negative variations associated with jaw opening in humans.

We examined the relationship between contingent negative variation (CNV) amplitude and digastric muscle activity in jaw-opening motor task. Subjects were instructed to open their jaws in response to a sound stimulus following a visual warning stimulus. Electroencephalograms were averaged by using the visual stimulus pulse as a trigger. Bilateral CNVs were clearly recorded at several cortical locations, with a tendency for larger amplitudes at temporal placements.
