Platelet tocopherol levels in mucocutaneous lymph node syndrome (MCLS: Kawasaki disease).

Studies of changes in platelet tocopherol in 16 children with mucocutaneous lymph node syndrome (MCLS) were undertaken during the disease course. Plasma tocopherol was lowest at the beginning of the disease (during the first 1 to 2 weeks), while it increased with improvement in symptoms at 4 to 5 weeks. Platelet tocopherol however was lowest in the first 2 to 3 weeks, and at about the 4th week or later increased. When compared with normal levels, determined in 24 children without MCLS (5 to 14 years old), the lowest level seen throughout the disease course coincided with it. Thus, platelet tocopherol may be generally higher in MCLS. There was little correlation between plasma and platelet tocopherol levels which were simultaneously assayed, (r = 0.26, p less than 0.1, n = 44), while, the correlation between platelet tocopherol and plasma tocopherol/total lipid ratio was increased significantly to 0.55 (p less than 0.001, n = 38). Individual patients in whom platelet tocopherol and plasma tocopherol/total lipid ratios were repeatedly assayed, were divided into two groups based on the Asai scoring, as a high risk group and a low risk group. No correlation was observed between the risk grades and either platelet tocopherol or tocopherol/total lipid ratios in plasma.

[Clinical efficacy of T-1982 (cefbuperazone) for infections of children].

T-1982 (cefbuperazone) was given at a daily dose of 42-80 mg/kg to 7 children with bacterial infections; 3 with bacterial intestinitis, 1 with bronchitis, 1 with tonsillitis and 2 with urinary tract infections. Clinical effectiveness was obtained in 6 cases, and the rate was 85.7%. Bacteriological responses were "disappeared" in 4 cases, "decreased" in 1 case and "unknown" in 2 cases. No noticeable side effects were observed except elevation of eosinophil in 1 case.

Effect of body lipid mass on red blood cell-and plasma-tocopherol levels.

A study on the relation of plasma- and RBC-tocopherol to plasma lipids and adiposity led to the following findings. Plasma tocopherol increased in parallel with increased plasma total lipid and beta-lipoprotein levels, while RBC tocopherol levels had little correlation with those of plasma lipids or beta-lipoproteins. The RBC-to-plasma tocopherol ratio decreased with increased plasma lipids and beta-lipoproteins. Though the relations of HDL-cholesterol, total lipids and beta-lipoproteins to RBC tocopherol levels resembled each other, those of HDL-cholesterol to plasma tocopherol levels and RBC-to-plasma tocopherol ratios differed greatly from the relationship in total lipids and beta-lipoproteins, especially the RBC-to-plasma ratios which had no definite pattern. Plasma tocopherol was unrelated to obesity grade, while the RBC tocopherol and RBC-to-plasma ratios decreased with increased obesity.

Effects of intralesional interferon on neuroblastoma: changes in histology and DNA content distribution of tumor masses.

Local injections of interferon, 30 X 10(4) IU, ten times, were found to reduce masses of neuroblastoma. Histologic findings and nuclear DNA measurements of cells from tumors in which interferon has been injected compared with cells from tumors that did not receive injections demonstrated that the reduction of neuroblastoma masses by interferon did not depend on increased cell differentiation or on interruption of tumor cell production. Although the mechanism of interferon's action on reducing neuroblastoma is unknown, the disappearance of tumor cells may be caused by high levels of interferon in tumor tissues.

Changes of alpha-tocopherol levels in red blood cells and plasma with respect to hemolysis induced by dialuric acid in vitamin E-deficient rats.

The plasma and red blood cell (RBC) levels of alpha-tocopherol in rats were determined in association with hemolysis induced by dialuric acid for a 7-week period of tocopherol deprivation following a 2-week period on tocopherol-containing diet. The patterns of decreases in plasma and RBC levels during the 7-week period were similar, differing in one respect. The RBC levels were characterized by a prompt decrease to non-detectable levels in the 4th week, while at that time a plasma level of 100 microgram/dl, albeit low, was maintained during the subsequent two weeks. Consequently, tocopherol ratios of RBC to plasma lowered throughout the experimental course. As for the relationship between these levels and hemolysis values, non-hemolysis was recorded in animals whose minimum RBC and plasma levels were 40 microgram/dl packed cell and 180 microgram/dl, respectively. This experiment showed that the plasma tocopherol level which has traditionally been accepted as the only index of hemolysis values does not always reflect them, and that in animals with hemolysis, only RBC tocopherol levels were found to reflect hemolysis values.

Serum free amino acid depletion in ketotic children.

Changes in serum free amino acids as a response to ketogenic provocative test were studied in children with a history of ketosis but without hypoglycemia. 1. Of the 17 children who underwent the provocative test, only three showed clinical manifestations. They were among five children who responded to the test and developed hypoglycemia. 2. Hypoglycemic and non-hypoglycemic children differed greatly in the decrement of serum total gluconeogenic amino acids after completion of the test due to the relatively lower levels before provocation in the hypoglycemic group and the inversely higher levels in the non-hypoglycemic group. 3. Gluconeogenic amino acids that decreased sharply after the test were glycine, alanine, glutamine and glutamate.

Hydranencephaly in twins.

The sixth case of hydranencephaly in a twin is reported. The patient is an 11-year-old girl, and her twin was stillborn and macerated. Intrauterine disseminated intravascular coagulation may be the cause of hydranencephaly in this patient.

Muscular changes in Engelmann's disease.

In a case of Engelmann's disease in an 11-year-old Japanese boy the muscular changes were studied in detail. Muscle weakness was maximal about the pelvic girdle. Muscle biopsy showed the selective atrophy of type II fibres, and no degenerative change could be seen histologically, histochemically, or electron-microscopically. Although the distribution of muscular weakness in Engelmann's disease is similar to that of a progressive muscular dystrophy, the disease does not seem to be a myopathy.

Multicystic encephalomalacia in liveborn twin with a stillborn macerated co-twin.

Computed tomography showed multiple cystic lesions in the brains of three infants with cerebral palsy. Each patient was the product of a twin pregnancy with a stillborn, macerated co-twin. In two patients angiography was performed and suggested that the cystic lesions were multicystic encephalomalacia due to perinatal arterial occlusion.

An autoradiographic study of new fat cell formation in adipose tissue in adult mice during malnutrition and refeeding.

The renewal of adipose cells in adult mice has been autoradiographically studied. The number of adipose cells was diminished by eighty percent during malnutrition and the same number of adipose cells proliferated during the refeeding stage. The results of our study showed that adipose tissue, which had previously been believed to be stable in cell number, has the capacity for cell proliferation according to changes in nutritional status.

Tocopherol level in human blood cells.

The study was undertaken on alpha-tocopherol levels, separated from other analogs, in the red blood cells (RBC) of cord blood, premature infants, healthy children and adults as well as in pregnant women, and compared with the plasma levels. 1. The majority of tocopherol found in the RBC was localized in the membranes. 2. Only alpha-tocopherol was found in the RBC, while alpha- and gamma-tocopherol were found in the plasma. 3. Alpha-tocopherol level in the RBC changes during development in parallel with that in the plasma. However, changes in the level in RBC were smaller than those of the plasma. 4. The ratio of alpha-tocopherol level in the RBC to the plasma was higher in the cord blood and premature infants than in the children and adults. 5. With regard to the cases with high plasma tocopherol levels, a different finding was obtained on the RBC level between pregnant women and adults with a large amount of tocopherol. In the latter, high RBC levels were observed as the plasma levels were elevated, while in spite of a high plasma tocopherol level, the lowest RBC levels were found in the pregnant women.

Studies on the factors influencing the hydrogen peroxide hemolysis test.

There seems to be a greater variation between laboratories in hydrogen peroxide hemolysis techniques than in other clinical laboratory procedures. In this report, factors influencing hemolytic values were examined and the effect of the combination of these factors on the results was analysed statistically by using the orthogonal array. Factors influencing hemolysis induced by hydrogen peroxide were as follows; the concentration of hydrogen peroxide, temperature in the peroxide reagent when added to the red cell suspension, the red cell concentration in the cell suspension and the addition of charcoal to the reaction mixture. However, addition of charcoal may not be essential to stabilize the hemolytic values and other factors such as keeping blood for 4 hours at room temperature before testing, the difference between investigators, a reaction time 2 or 3 hours and the technique of adding peroxide reagent to the cell suspension, had little effect on hemolysis. The most important factor was the temperature in hydrogen peroxide solution. The estimated hemolytic values by the orthogonal array linearly correlated with the plasma tocopherol levels.
