Preliminary observations on the effects of the Chinese medicinal herbs Astragalus membranaceus and Ligustrum lucidum on lymphocyte blastogenic responses.

The biological effects of extracts of two Chinese medicinal herbs, Astragalus membranaceus and Ligustrum lucidum, on in vitro lymphocyte blastogenesis were assessed. Aqueous extracts augmented the spontaneous [3H]thymidine incorporation in the mononuclear cells (MNC) of 14 normal subjects from 273.0 to 609.3 counts per minute (cpm) and 252.9 to 656.9 cpm for the two herbs, respectively. The stimulation indices were 2.4 and 3.1, respectively (p less than 0.001). They also augmented the proliferation of normal subjects' lymphocytes induced by suboptimal concentrations of phytohemagglutinin (PHA) from 5084.6 to 23,398.3 and 221.7 to 24,132.8 cpm, of concanavalin A (con A) from 4046.5 to 15,661.5 and 677.6 to 14,644.6 cpm, and of pokeweed mitogen (PWM) from 4377.9 to 24,405.6 and 322.7 to 11,730.0 cpm, respectively (p less than 0.00). Herb extracts augmented the PHA responses of the MNC from 14 cancer patients significantly (p less than 0.01 and p less than 0.05, respectively). Extracts of L. lucidum also augmented the con A response of patients (p less than 0.05). The augmenting effect of the herbs on the PHA, con A, and PWM responses was dose dependent, and proliferation was inhibited at higher concentrations. The optimal concentration for stimulating the MNC of cancer patients was 100 micrograms/ml, compared to 10 micrograms/ml for the MNC of normal donors. MNC of seven patients depressed the mitogen responses of normal cells in a co-culture system. This was partially abrogated in five by preincubating the patients' cells in herb extracts for 45 min or by irradiation of the patients' cells. These results suggest that the herb extracts contain immunomodulatory components which may be useful in the immunotherapy of disease.

Positive group efficacy in adolescent treatment.

The Positive Group as a treatment modality was evaluated in a pilot study with 24 adolescents over an eight-week period. This technique requires adolescents to make positive statements about themselves and others in the group. Each adolescent must also verbally acknowledge the compliment paid him. Proscribed are all negative comments. The results were evaluated primarily through clinical observation and anecdotal reports as given by counselors, nursing staff, program coordinators, and attending psychiatrists. The results support incorporating the Positive Group Therapy into both inpatient and outpatient treatment practices. Positive Group experience was found facilitative over a broad range of diagnostic categories, helping enhance self-image, social skills and leadership potential in the presence of and with the encouragement of peers. Illustrative clinical vignettes are offered. The authors conclude that the Positive Group is a probably beneficial therapy approach.

Monomethylformamide levels in human urine after repetitive exposure to dimethylformamide vapor.

Eight healthy male subjects were exposed to DMF vapor at a concentration of 8.79 +/- 0.33 ppm for six hours daily for five consecutive days. All urine voided by the subjects was collected from the beginning of the first exposure to 24 hours past the end of the last exposure and each sample was analyzed for monomethylformamide (MMF). MMF was rapidly eliminated from the body with urine values peaking within a few hours following the end of each exposure period. Very little was found in the 24-hour postexposure sample and none was found in a 48-hour postexposure sample. There was no increased excretion of MMF in the urine following repetitive exposure. The mean for the seven-hour (end of exposure) sample was 4.74 microgram/ml or 736.8 microgram. Lower and upper one-sided 95% tolerance limits for 95% of the population were 1.2 microgram/ml (367 microgram) and 13.9 microgram/ml (1625 microgram). The coefficient of variation (CV) for microgram MMF/ml was approximately 25 times more variable than the CV for total microgram.

Cation and hydrogen ion effect on canine acid and gastrin output.

Differential stimulating effects on the parietal cell of dogs by calcium, magnesium and sodium were noted only at an alkaline pH. Acid inhibited the parietal cell, and antral perfusion did not cause gastrin stimulation at any pH level. The pH of the milieu of the parietal cell appears to be a more critical regulator of acid secretion than physiologic antral gastrin variation.

Human gastrin response to secretin after vagotomy.

The gastrin response to a liquid meal with and without secretin infusion was studied in nine patients undergoing selective or truncal vagotomy with pyloroplasty for duodenal ulcer disease. Fasting gastrin levels were significantly increased in eight of nine patients after vagotomy, but secretin infusion did not consistently suppress these basal gastrin levels either pre- or postoperatively. Infusion of secretin did significantly lower the integrated gastrin response to feeding both pre- and postoperatively in eight of nine patients. Vagotomy alone did not significantly alter the integrated gastrin response to feeding. This data gives evidence that secretin infusion remains a helpful diagnostic test, differentiating those patients with recurrent ulcer and elevated gastrin levels postvagotomy from those patients with occult Zollinger-Ellison syndrome.

Effects of hypercapnia on uterine and umbilical circulations in conscious pregnant sheep.

Changes in the uterine and umbilical circulations during induced hypercapnia were studied in nine unanesthetized near-term pregnant sheep. Blood flows were measured with electromagnetic flow transducers and arterial pressures with vascular catheters implanted under anesthesia 2-16 days prior to experiments. Hypercapnia was induced in the fetus alone by giving acetazolamide iv to the fetus, 100-200 mg/kg. Mean fetal arterial Pco2 increased from 49.5 to 63.4 mmHg but no significant changes in umbilical blood flow occurred. Stepwise increases in both maternal and fetal arterial Pco2 were induced by increasing maternal inspired CO2 concentration to a maximum of 12%. No dignificant changes occurred in uterine or umbilical circulations until hypercapnia was severe (maternal arterial Pco2 greater than 60 mmHg, fetal arterial Pco2 greater than 70 mmHg). With severe hypercapnia uterine vascular resistance increased significantly and uterine blood flow decreased despite an increase in maternal arterial pressure; fetal arterial pressure and umbilical blood flow increased significantly, but umbilical vascular resistance did not. We conclude that hypercapnia in conscious pregnant sheep is associated with significant changes in uterine and umbilical circulations, but only when hypercapnia is severe. Carbon dioxide is unlikely to be a factor in normal physiological regulation of the uteroplacental circulation in this species.

Effect of amendments on the microbial utilization of oil applied to soil.

Replicate field plots comprising a control, plus oil, plus oil and bacteria, plus oil and fertilizer (urea-phosphate; 27:27:0), and plus oil, bacteria, and fertilizer were monitored over a 308-day period for changes in bacterial and mold numbers. Changes in the chemical composition of the oil applied to the plots was followed by using chromatographic techniques. Application of fertilizer resulted in a stimulation of bacterial numbers and in the rate of utilization of n-alkane components of the saturate fraction. The application of oil-utilizing bacteria, however, resulted in only a slightly accelerated rate of utilization of n-alkane components of chain lengths C20 to C25. The isoprenoids, phytane and pristane, were still present in gas-liquid chromatography profiles after digestion of the n-alkane components of the saturate fraction. Those plots which received fertilizer showed an accelerated rate of recovery of native vegetation.
