Neonatal myosin heavy chains are not expressed in Ni-induced rat rhabdomyosarcoma.

Myosin heavy chain (MHC) composition of chemically-induced rhabdomyosarcoma (RMS) was analyzed by gel electrophoresis and Western blotting using a panel of monoclonal antimyosin antibodies specific for embryonic-, neonatal-, slow- and adult fast-type MHC isoforms. Myosin extracted from tumours and electrophoresed on 6%-sodium dodecyl sulfate (SDS)glycerol gels was found to migrate as three distinct MHC components. These polypeptides were present in different relative amounts in the five RMS studied. Western blotting experiments revealed that variable proportions of embryonic-, slow- and adult fast-, but not neonatal-type, MHC isoforms are consistently expressed in RMS. Indirect and double immunofluorescence procedures applied to cryosections of tumoral tissue showed that: (a) RMS cells were unreactive with antineonatal-type-MHC antibody, (b) the majority of neoplastic, desmin-positive, cells contained embryonic- as well as adult fast-type MHCs and (c) a minority of cells were labelled by anti-slow MHC antibody. The results of this study indicate that there is no obligatory sequence of MHC isoform expression in the molecular transition (emb----neo----adult) which occurs during rat skeletal myogenesis.

[Observations and significance of extrasystole in very young athletes].

80 very young football players (from 8 to 12) have been examined for three months by some clinical and instrumental cardiologic tests (starting E.C.G. and after graduated stresses on a football court). The starting E.C.G. showed variable extresystolic arrhythmias in 8 subjects, without any sure signs of a cardiopathy, to point out by deeper tests (such as polygraphic, echocardiographic test and rx heart teleradiography). The above-mentioned arrhythmias felt the effects of training variably, since they regressed in 6 cases, however two subjects needed a pharmacological intervention. They are still talking over the meaning to give to extrasystolic arrhythmias in very young people in evaluation of attitude to agonism and in programming training.

[Initial observations in a cardiovascular poly-instrumental study of a homogeneous sample of young football players].

A group of 20 football players (from 12 to 15 aged) has been taken for a boodless test of cardiology (such as polygraphic and echocardiographic test) at the beginning of a longitudinal study in order to give the exact meaning, as much as possible, to the fitness for an agonistic training. One of the subjects examined was not for that test because of his cardiomegaly, another one had an anomalous mitral valve, in both the subjects the clinic and instrumental side resulted within normal ranges only if searched by E.C.G. About the other athletes the starting parameters will be controlled again, after they will have been in that football team for 1-2 years more. The authors think the point that came out is very important and they hope everybody to execute those studies either to avoid some cardiopathies pass unnoticed to routine tests or to value the effects of training of heart sizes and performances.

[Effect of 2 mercaptopropionylglycine on the cytotoxic activity of lymphocytes from patients with chronic active hepatitis against rabbit hepatocytes isolated and cultured in vitro].

"In vivo" and "in vitro" action of 2 MPG on lymphocytotoxic activity was evaluated in 22 patients with CAH, 14 were HBsAg positive and 8 were negative. The test was performed with and without 2 MPG in the colture medium, and before and after treatment. HBsAg+ patients received 2 MPG treatment, while HBsAg- ones received immunosoppressive therapy and 4 of these were on 2 MPG treatment as well. Cytotoxic Index was reduced non-significantly by addition of 2 MPG in colture medium in both groups. 2 MPG treatment does not modify cytotoxic activity which is reduced by immunosoppressive therapy. These findings suggest a positive effect of 2 MPG on liver-cell metabolism with an increased resistence of the epatocyte to the "in vitro" lymphocytes aggression. Hence it may be suggested an association treatment with immunosoppressive agents and 2 MPG.

[Separation and characterization of human T G and T M lymphocyte populations].

During the last few years a number of experimental evidences have shown the presence of Fc receptors for IgG or IgM on the membrane of human T cells. These two different receptors are detectable and mutually exclusive on distinct cell populations named respectively TG, TM and T "null" (which lack detectable receptors). Studies on the functional activities of these cells have shown that TM and TG lymphocytes play an antitetical role in regulating B cell response, TM exerting an "helper" activity on the differentiation of B lymphocytes while TG having a "suppressor" one. The aim of this study has been to determine the values of these two subpopulations in a group of twenty control subjects. Our results have shown that TG constitute 10%, whereas TM represent 40% of the total T cells. After EA-G rosetting, the purification of this subpopulation on a density gradient has shown an enrichment of more than 90% in TG cells, while TM contaminate this fraction for less than 4%. The purity of the fraction containing TM has been evaluated using the localization of alpha-naphthyl acetate esterase activity, which has shown that more than 88% of the cells in this fraction are positive for this enzyme.

[Liver cell membrane autoantibody (LMA) in chronic liver disease].

The incidence of LMA, detected by indirect IF on isolated rabbit hepatocytes, was significantly higher in CAH-non B (58%) than in CAH-B (15%). Sera from AVH patients showed a smaller percentage of positivity whereas the CPH patients were negative. The organ-specificity, the high incidence in relation to other non-organ-specific autoantibodies and the inverse correlation with the markers of HB Virus, make LMA a specific and sensitive marker of "autoimmune" chronic liver disease.

[Infective endocarditis: a complication of idiopathic hypertrophic subaortic stenosis (author's transl)].

The idiopathic hypertrophic subaortic stenosis (IHSS) is thought to be infrequently complicated by infective endocarditis. Because IHSS is a disorder of the myocardium and not endocardium is not generally considered to have increased risk of infective endocarditis. Some Authors found, however, that approximately 5% of patients with IHSS develop bacterial endocarditis, because hemodynamically IHSS is like to valvular diseases. Therefore the incidence of endocarditis in IHSS is the same of valvular diseases. The vegetations can be found on anterior mitral valve leaflet, aortic cusps and subaortic endocardium. Some patients have dental surgery prior to the onset of endocarditis and others are infected with oral streptococci. For these reasons prophylactic antibiotics should be administered to patients with IHSS undergoing dental manipulation and in other settings where the risk of developing bacteremia is high. We describe one case of IHSS complicated with streptococcus viridans endocarditis. Vegetations, determined with echocardiography, were present on anterior mitral valve leaflet, aortic right coronary cusp and interventricular septum. In a review of literature we are been able to find twenty-seven cases of IHSS complicated with endocarditis.

Synthesis of (Glu-OMe)2-litorin.

The synthesis of the nonapeptide less than Glu-Glu(OMe)-Trp-Ala-Val-Gly-His-Phe-Met-NH2, corresponding to the formula of the (Glu-OMe)2-litorin, is described. The compound has the same chemical and biological properties of the second bombesin like peptide extracted from the skin of the Australian frog Litoria aurea.
