The effects of 2-(alpha-Tenoyl) thiopropionylglycine (TTPG) on human lung mucus.

2-(alpha-Tenoyl)-thiopropionylglycine (TTPG) was tested for its mucolytic activity on mucus specimens from human lungs. Our data demonstrate that a significant amount of mucus is solubilized in the presence of TTPG; this aspect is more evident when rat liver microsomes are incubated together with lung mucus and TTPG. Furthermore our data demonstrate the presence of thioesterase activity associated with liver, lung, kidney and brain microsomes capable of hydrolysing TTPG and thus liberating free thiol groups. Further studies are in progress to characterize the microsomal thioester activity as well as its probable role in regulating the viscosity of mucus.

[Thioamide derivatives with gastro-protective activity].

A series of 3-aryl-prop-2-enthioamide derivatives has been synthesized. These compounds were evaluated for gastro-protective activity in the rat. It has been shown that the N,N-dimethylamino derivative was the most active compound in this series [Table I comp. (II)].

Effects of calcitonin on rat extrapyramidal motor system: behavioral and biochemical data.

The effects of i.v.c. injection of human and salmon calcitonin on biochemical and behavioral parameters related to the extrapyramidal motor system, were investigated in male rats. Calcitonin injection resulted in a potentiation of haloperidol-induced catalepsy and a partial prevention of apomorphine-induced hyperactivity. Moreover calcitonin induced a significant decrease in nigral GAD activity but no change in striatal DA and DOPAC concentration or GAD activity. The results are discussed in view of a primary action of calcitonin on the striatonigral GABAergic pathway mediating the DA-related behavioral messages of striatal origin.

Aromatization of testosterone by adipose tissue and sexual behavior of castrated male rats.

The influence of aromatization of testosterone occurring in adipose tissue has been studied in castrated obese male rats. The treatment for 30 days with a subliminal dose (115 micrograms/kg per day) of testosterone propionate improved significantly the sexual performance of castrated obese male rats as compared to the nonobese controls. Untreated castrated obese animals did not show any enhancement of sexual behavior. Furthermore, obesity failed to improve sexual performance in intact male rats. Measurements of plasma levels of testosterone and 17 beta-estradiol showed an increase in aromatization of testosterone in obese rats. Present data suggest that adipose tissue can be considered an important site for aromatization of testosterone to 17 beta-estradiol, and that aromatization is involved in the activation of sexual behavior in castrated male rats.

Effects of different doses of apomorphine on GAD activity in rat substantia nigra.

The effects of different doses of the dopamine (DA) receptor agonist apomorphine on the activity of the gamma-aminobutyric acid (GABA)-synthesizing enzyme glutamic acid decarboxylase (GAD, EC 4.1.1.15) were investigated in rat substantia nigra in comparison with haloperidol and sulpiride, two DA receptor blocking agents. Results obtained show that low doses (10,35 microgram/kg, s.c.) of apomorphine induce a decrease in nigral GAD activity whilst an opposite effect is observed with the highest dose (1000 microgram/kg, s.c.). No significant change is observed following injection of the intermediate doses (100 and 500 microgram/kg, s.c.). Moreover, sulpiride at the dose used (2 mg/kg, i.p.) induces an increase in GAD activity whilst no effect follows systemic injection of the same dose of haloperidol. The results are discussed in light of recent neurochemical and behavioral data.

[Control of the emesis induced by cisplatin in patients with head and neck neoplasms. Randomized study of metoclopramide alizapride and placebo].

A double blind study has been lead on 75 patients affected by head and neck cancer treated with CDDP, to verify the alizapride's antiemetic efficacy versus placebo and metoclopramide. On 76 patients examined, 28 has been treated with CDDP 50 mg/mq + alizapride; 28 with CDDP 50 mg/mq + metoclopramide and finally 19 with CDDP 50 mg/mq + placebo. Both alizapride and metoclopramide administered have resulted effective on the control of the emesis induced with CDDP without side effects.

[Treatment of acute leukemias in adults. IV. Treatment results on 375 patients at 9 hematology centers].

On the basis of standardized protocols of the therapeutic results of acute non-lymphoblastic leukaemias in adults sent to the Institute of Haematology in Warsaw from 8 haematological centres in Poland it was demonstrated that complete remission occurred in 34.4% of patients (129 out of 375 cases). The mean survival time of the patients treated intensively according to programmes I, II, III and IV 8.6 months, those of patients with complete remission - 13.5 months, patients without complete remission - 3.7 months. The most frequent cause of death (82.5%) were infections and/or haemarrhagic diathesis.
