Establishment of a cell culture line from a transplantable rat stomach cancer induced by N-methyl-N'-nitro-N-nitrosoguanidine.

A cell line designated as BV9 was established in culture from a transplantable rat stomach cancer, which was originally induced in the glandular stomach of a Wistar rat by p.o. administration of N-methyl-N'-nitro-N-nitrosoguanidine. The monolayer cells, which have been subcultured for more than 35 passages, revealed pleomorphic features. Chromosomal analysis showed hypertetraploidy (mode, 95), and marker chromosomes were present. Cultured cells were injected s.c. into cyclophosphamide-conditioned syngeneic rats and produced tubular adenocarcinomas resembling the original tumor.

In vitro citrate synthesis by the dog kidney. Investigations with cortex, red and white medulla slices.

In vitro utilization or production of citrate by the cortex, outer medulla or inner medulla of dog kidney was measured. Our data show: 1. An in vitro citrate synthesis or utilization capacity of the cortex greater than that of the red medulla. 2. An effect of pH on citrate synthesis or utilization capacity of the cortex, an effect not seen with medullary slices. 3. An absence of citrate synthesis or utilization by white medulla slices. It would seem that the citrate found in the white medulla and the papilla of the dog kidney in vivo was not produced in situ.

[Fibronolytic treatment with activator: clinical results, angiographic findings and laboratory results (author's transl)].

Two-day treatment with activator (equimolar streptokinase-human plasminogen complex) was given to 26 patients. Indications were chronic arterial occlusion and stenosis of the lower-limb arteries (25 patients), and thrombosis of the subclavian veins. The onset of claudication had on average been 7.5 months previously, average walking distance 288 metres. Five occlusions of iliac arteries and three of femoral arteries were removed. In addition, stenosis of the aorta (one case), the iliac artery (ten cases) and femoral artery (one case) was partly removed. Occlusion of the axillary vein was also removed. Average duration of the successfully treated occlusions was 4.3 months. Those stenoses that responded to activator had the well-known morphological criteria for fibrinolysis. The walking distance of the patients increased from an average of 288 to 401 metres. Activator treatment was well tolerated. No serious, especially cerebral, complications occurred. The most striking laboratory finding was of circulating plasmin during the entire duration of the infusion.

Relation between time-dose and local control of operable breast cancer treated by tumorectomy and radiotherapy or by radical radiotherapy alone.

67 patients with tumors 3 cm or less and with negative axilla, who had lumpectomy and radiotherapy, and 122 patients with larger tumors or clinically positive axilla, who had radical radiotherapy without prior lumpectomy, were evaluated at five years to determine the optimum dose for local control. 7000 rad in seven to eight weeks controlled 85% of lumpectomy cases, whereas 8000 rad in 81/2 weeks were required to control two-thirds of cases treated by radical irradiation alone. Subclinical (N0, N1a) and clinical disease (N1b) in the axilla was controlled, in a very high percentage, when 6000 and 7000 rad were delivered, respectively. Dose response curves were obtained for clinical disease in the breast and axilla. No such response was elicited for subclinical disease. Undifferentiated cancers had higher recurrence rates than well differentiated tumors. Radiation fibrosis in lumpectomy cases was insignificant, whereas 10% of radically irradiated patients had fibrosis of the breast.

Rat macrophage-mediated toxicity to cancer cells; effect of endotoxins and endotoxin inhibitors contained in culture media.

The cytotoxic effect of normal or BCG-activated rat macrophages on a syngeneic line of cancer cells was compared in media containing rat or bovine sera. Normal macrophages were usually cytotoxic to cancer cells in fetal or newborn bovine serum; however, they enhanced cancer cell growth in normal rat serum. BCG- activated macrophages were toxic to cancer cells regardless of the serum used in the assay. Many cell culture media or sera obtained commercially were found to be contaminated by bacterial endotoxins. When endotoxin-free reagents were used in the toxicity assay, different results were observed: normal macrophages were not cytotoxic, but rather, they often enhanced cell growth even in fetal bovine serum; toxicity of activated macrophages was significantly reduced in normal rat serum. These results suggest that endotoxin and endotoxin inhibitors play a role in the modulation of macrophage-mediated cytotoxicity. These results emphasize the importance of monitoring endotoxin contamination in cell culture reagents used in assays involving macrophages.

Partial characterization of a cubic Bacillus phage.

Partial characterization of a cubic Bacillus phage. Can. J. Microbiol. 24: 986--993. Phage Bam35 is an icosahedron of about 63 nm in diameter. It has a double capsid with spikes at the vertices, and a tail which seems to appear upon nucleic acid ejection. The phage contains DNA and, probably, lipids which seem to be located in the inner coat. The phage is Bacillus-specific, UV- and lipase-resistant, and sensitive to heat, chloroform, and ether. The latent period is 50 min and the burst size is 39. Page Bam35 belongs to a new virus group which includes a phage of B. anthracis and four phages of gram-negative bacteria harboring drug-resistance plasmids.

The use of Pro-Banthine to induce gastrointestinal hypotonia.

Immediate and satisfactory gastrointestinal hypotonia was induced in 48 patients with 5--10 mg of intravenous Pro-Banthine during radiographic examination; side-effects were minimal. If given intravenously in small doses, Pro-Banthine is a satisfactory alternative when glucagon is contraindicated or not available.

[Autism Asperger of early schizophrenia (author's transl)].

Sometimes there exist difficulties to differentiate between adolescents with autistic personalities (autistic psychopathy, Asperger) and schizophrenia. The authors describe a 16-years old autistic boy demonstrating that a clear cut separation of these two different nosologic entities is only possible by means of a longitudinal observation. By this method psychopathological disturbances will be interpreted correct and within the context of the underlying syndrome.

[Isolated cystic lymphangioma in the mediastinum].

Cystic lymphangioma localized only in the mediastinum is a rare type of primitive tumour (less than 1% of the mediastinum primitive tumours). These three new observations concern lymphangiomas with an anterior position ; therir anatomical connections with the lymphatic system are difficult to show. Treated by surgery these benign tumours have a good prognosis and only a few cases of local recurrences have been reported.
