Noncultivated and short-term cultivated blood leukocytes of BLV-infected cattle both with and without tumorous leukosis have been investigated for BLV-p24 antigen expression and for their capacity to bind BLV-p24 antigen. In animals with persistent lymphocytosis using cell extracts of 3-6 X 10(8) non-short-term cultivated blood leukocytes, BLV-p24 antigen could be identified by means of competitive RIA. In cattle with tumorous leukosis, the antigen detection in non-short-term cultivated leukocytes may be masked by the presence of antigen-binding receptors. The capacity of p24 antigen binding is likely to be a phenotypical marker for the cells occurring in the tumour phase only.
The anatomical-pathological findings of 15 woolly monkeys were comparable with those described in man with essential benign or malignant hypertension. Kidneys revealed arterio- and arteriolosclerosis or an onion peel-like proliferation, in some cases necrotizing endarteritis. Obesity due to restricted physical activity, unnatural feeding, and psychical and physical stress could have contributed to the development of this condition. Further, high vulnerability of the genus woolly monkey is postulated. Thus, essential hypertension of woolly monkeys appears to be a multifactorial disease.
The present study deals with the induction of teratomas and teratocarcinomas in two strains of mice (C3H/Bln and 129/terSv). 6 to 7 days old egg cylinders were transplanted beneath the kidney capsule of adult syngeneic male and female recipients. Out of 115 grafted embryos 32 gave rise to teratoid tumors. Both the overall tumor incidence (teratomas and teratocarcinomas) and the overall percentage of teratocarcinomas were approximately the same in the strains used. In strain C3H/Bln the gender of the recipient seemed to influence the outgrowth of malignant tumors. Two transplantable C3H-teratocarcinomas could be established (DTC-4, DTC-8). Up to date both have retained their pluripotent differentiation pattern which makes them useful for intended further investigations.
Papillary carcinomas of the thyroid gland induced in rats by combined methylthiouracil (MTU)/nitrosomethylurea treatment were studied electron microscopically and histochemically for their acid phosphatase activity, radioactive iodine uptake and for their thyroglobulin synthesis. The papillary carcinomas accumulated radioactive iodine, synthesized thyroglobulin and showed a perinuclear localization of acid phosphatase activity in the cytoplasm. After omitting the reactive thyrotropic effect (cessation of MTU administration) a decrease of both iodine organification and acid phosphatase activity was observed. The synthesis of thyroglobulin was also reduced. The changes in the ultrastructure of tumour cells corresponded to the respective functional activities. The results suggest that the papillary carcinomas may have a functional dependence on thyroid-stimulating hormone.
The skeletal muscle has the capacity to respond adaptively to increased use. This observation could open up the feasibility of constructing pumping chambers to support or even replace cardiac work. We investigated the changes in enzyme activity due to chronic stimulation in an animal skeletal muscle. In 5 adult sheep the psoas muscle of one side was electrically stimulated through the muscle nerves, with an implantable stimulation unit for 5 weeks. The activity of the hexokinase (E.C.2.7.1.1.), lactate dehydrogenase (E.C.1.1.1.27), malate dehydrogenase (E.C.1.1.1.37), creatine kinase (E.C.2.7.3.2.) choline acetyltransferase and the contents of adenosine triphosphate and adenosine diphosphate were determined in bioptic specimen. The use of only 15 Hertz as a stimulation frequency led to a transformation of an originally fast-twitch muscle into a slow-twitch muscle with reduced susceptibility to fatigue. These results indicate a potential role of the skeletal muscle as an ideal myocardial substitute with the ability to perform hemodynamic work.

A report is given on a young Charolais bull showing stiff gait and muscle atrophy of the rear limbs. The sign of Ortolani and a distinct subluxation of the hip on the radiography lead to the diagnosis of a hip dysplasia. These findings were confirmed by pathomorphological investigations.
Transgenic mice harbouring growth hormone gene constructs have been produced by DNA microinjection into pronuclei of fertilized oocytes. We examined transgenic mice carrying a mouse metallothionein I-human growth hormone (mMT I-hGH) fusion gene. Here, we present our results concerning gene integration, gene expression, and phenotypical, clinical and pathomorphological alterations found in mice expressing the hGH transgene. Body and organ growth was significantly increased in transgenic mice, whereas fertility was found to be reduced. The life-span was markedly shortened indicating detrimental side-effects of the high levels of circulating hGH. Lesions of kidneys, liver and heart were the predominant pathological findings. Our own results are compared with those obtained by other authors who have investigated mice carrying rat, bovine or ovine growth hormone fusion genes. GH-transgenic mice may serve as a model system to investigate ectopic expression of hormone genes thus circumventing endogenous feedback control mechanisms in complex hormonal cascades.
A balance of energy budgeting of Ehrlich mouse ascites tumour cells including mitochondrial and glycolytic ATP production and about 80% of ATP consumption in a high phosphate medium is presented. In the share of glycolysis was about one-third of the total ATP production, more than twice that found in a low phosphate medium. The extent of a single energy reaction was assessed from the decrease of coupled oxygen consumption and lactate formation following the specific inhibition of this process. The inhibitory effects on coupled respiration and glycolysis were identical for the energy consuming processes measured: protein turnover, Na+/K(+)-ATPase, Ca2(+)-transport and RNA synthesis.
An antiserum against human thyroglobulin isolated by gelfiltration from the human thyroid gland was raised in rabbits. The specificity of the antiserum to detect human thyroglobulin was determined by an immunoperoxidase technique on formalin-fixed and paraffin-embedded tissue sections of 17 primary thyroid carcinomas and 80 carcinomas of other origins, such as pancreas, colon, breast, bladder, ovary, testis, and larynx. Positive reactions to immunological staining were recorded only from differentiated thyroid carcinomas, while no reactions were exhibited by the other malignant tumours tested in this study.
The radiological findings regarding eggs (variation in form, size, shell thickness, degree of calcification) combined with the clinical symptoms (Ca-Gluconate and Oxytocin application as well as unsuccessful labour contractions and constipation) set up the diagnosis of dystocia in the tortoise. Celiotomy is the remedy for cesarean section, ovariosalpingotomy and gastrointestinal foreign body. A large, topographically well-placed Plastron incision, separation and reposition of the m. pectoralis and m. obliquus internus abdominis using cross-stitch sutures, as well as the careful manipulation of tissues, are of great importance. Gastrotomy is even feasible on terrapins through a partial detachment of the stomach from the liver, enabling a sufficient presentation of the stomach.
High pressure freezing permits the successful cryoimmobilization of thick biological specimens (up to approx. 500 microns). A very high yield of adequately frozen specimens, in which no segregation patterns due to ice crystal formation is apparent after freeze-substitution or freeze-fracturing, is obtained with suspensions of microorganisms as well as plant and animal tissue. This very high yield is attributed to an optimized transfer of pressure and cold to the biological specimen. This is achieved by replacement of extraspecimen water or buffer by 1-hexadecene, a chemically inert, hydrophobic paraffin oil of low viscosity and low surface tension.
The importance of CT imaging in maximum inspiratory position for the identification of lung metastases is emphasised. In a woman patient suffering from respiratory insufficiency and hypernephroma and clinically suspected of lung metastases, it became necessary to produce two computed tomographs of the thorax at short intervals to clarify the number and size of the pulmonary foci. Since the first examination could not be conducted in a position of maximum inspiration because the patient was short of breath, the procedure was repeated six days later after intensive breathing exercises and subjective improvement of the patient's feeling tone. It was found that three of the four pulmonary metastases could not be seen on the technically unsatisfactory tomographs of the first examination, or could be detected only retrospectively.
The influence of the local anesthetics procaine and lidocaine on the anticonvulsant action of propranolol was investigated using the maximal electroshock seizure test in mice. Combined application markedly reduced ED50 values either of the local anesthetics or of propranolol. Selected anticonvulsants and antiarrhythmics were tested by comparison. Similar results could be obtained with phenytoin and isatin when administered together with propranolol, whereas procainamide and quinidine failed to exert synergism in a higher degree or in general. Na-Ca-exchange on the membrane level may be a link for the interpretation of the reported findings, since most of the drugs tested are involved in these processes.
Conservative treatment becomes an established procedure for small breast cancer worldwide. Randomized and non-randomized studies have shown equal prognostic results compared with the more radical operative treatment. This large-scale therapy only can be worthwhile, if the cosmetic results are better than breast reconstruction after mastectomy. Therefore a rigorous indication, a standardized technical procedure and a well-functioning team of interdisciplinary cooperation is required. Out of 500 conservatively treated patients we controlled a random sample of 134 relative to the cosmetic result. The results were related to our standard technique.

The homeodomain encoded by the Antennapedia (Antp) gene of Drosophila was overproduced in a T7 expression vector in Escherichia coli. The corresponding polypeptide of 68 amino acids was purified to homogeneity. The homeodomain was analysed by ultracentrifugation and assayed for DNA binding. The secondary structure of the isolated homeodomain was determined by nuclear magnetic resonance spectroscopy. DNA-binding studies indicate that the isolated homeodomain binds to DNA in vitro. It selectively binds to the same sites as a longer Antp polypeptide and a full-length fushi tarazu (ftz) protein. Therefore, the homeodomain represents the DNA-binding domain of the homeotic proteins.
