Identification of seroreactive regions of the human papillomavirus type 16 protein E4, E6, E7 and L1.

Small fragments of the DNA of human papillomavirus type 16 (HPV-16) were randomly cloned into the bacteriophage fd which expresses the resulting peptides as part of its capsid. Antisera raised against different HPV-16 fusion proteins were used for screening of the phage clones and the reacting peptides were determined by sequencing the inserted HPV-16 DNA fragments of the positive recombinants. Seroreactive regions of the proteins derived from the E4, E6, E7 (two regions) and L1 (three regions) open reading frames could be found by this approach. Of these seven regions, four were defined by at least two overlapping inserts, thus limiting the domains to between 10 and 15 amino acids. In the case of the E4 open reading frame, the same region identified by immunoscreening was also found when synthetic overlapping octapeptides were tested by ELISA with the anti-E4 antiserum. Using an approach to predict 'receptor-like' regions within the respective proteins, five of the seven regions were also identified. From the data on these regions, synthetic peptides were produced and used for the detection of antibodies against HPV-16 proteins in human sera by ELISA.

Metabolic alterations in end-stage and less severe heart failure--myocardial carnitine decrease.

Severe tissue carnitine deficiency impairs fatty acid oxidation. In explanted hearts from patients with end stage heart failure a 57% carnitine decrease was found in comparison with healthy donor hearts (p less than 0.05). The reduction of myocardial carnitine levels affected all areas of the explanted hearts to a comparable extent. Carnitine decreases in patients with dilated cardiomyopathy or coronary artery disease were similar. Endomyocardial biopsies from patients with less severe heart failure due to cardiomyopathy (n = 28) or other myocardial diseases (n = 8) showed a 42% decrease of total myocardial carnitine (in nmol/mg non-collagen protein) in comparison with biopsies from patients with normal cardiac function (controls) (heart failure: 5.7, confidence interval 4.2-7.0; controls 9.3, confidence interval 7.6-12.0, p less than 0.005). Free myocardial carnitine in heart failure was also different from controls (heart failure: 4.2, confidence interval 3.7-5.3; controls 10.3, confidence interval 7.5-12.2, p less than 0.001). The decrease of free and total myocardial carnitine was comparable in dilated cardiomyopathy and heart failure due to other diseases. Alterations in myocardial carnitine content represent therefore non-specific biochemical markers in heart failure with yet unknown consequences for myocardial function.

Tissue concentration, mRNA expression and stimulation of IGF-I in luteal tissue during the oestrous cycle and pregnancy of cows.

The expression of IGF-I in bovine luteal tissue was demonstrated by parallel measurement of IGF-I tissue concentration and its mRNA; highest synthesis was observed during Days 12-17 of the cycle and the first months of pregnancy. Tissue levels of IGF-I increased from Days 1-5 to Days 12-17 of the cycle followed by a rapid decrease at luteolysis; there was a continuous decline from early pregnancy until Months 6-9. Microdialysis perfusion experiments with corpora lutea in vitro at Days 8-11 of the cycle revealed a major effect: release of progesterone and oxytocin were highly stimulated in a dose-dependent manner. We suggest that IGF-I could be important in regulating the function of the bovine corpus luteum and may act in an autocrine/paracrine way.

[The Dornier lithotripter in a comparison. Measuring shockwave fields and fragmentation effects].

For all Dornier lithotripters, comparative shock wave field measurements performed by using a modern piezoelectric pvdf-shock wave measuring technique. The high temporal and spatial resolution of the pressure probe provides many details of the focusing field right up to the focal point. The maximum amplitudes, the axial and lateral pressure distribution around the focus, and their changes as a function of primary energy are presented. Focusing is influenced in particular by the geometry of the reflector, diffraction- and non-linear propagation effects. A comparison between incident shock energies on model calculi in vitro and the resulting fragmentation shows the relationship between focus energy and disintegration effect. For a given lithotripter with fixed electrical and geometrical parameters, the number of shocks needed for complete fragmentation of the model calculus is inversely proportional to the square of the capacitor voltage, and proportional to the volume of the model calculus.

[Absorption profile and absolute bioavailability of a theophylline--retard preparation].

Absorption Profile and Absolute Bioavailability of a Theophylline Retard Preparation. In a single-dose cross-over study with 16 healthy male nonsmoking volunteers the absorption profile and absolute bioavailability of the retard pellets contained in all dosage strengths of Bronchoretard (100, 200, 350, 500 mg anhydrous theophylline) were investigated. One capsule (500 mg theophylline) was given after an evening meal. The reference treatment consisted of an i.v. infusion over 8 h in a total dosage of 350 mg theophylline. Plasma concentration-time profiles were evaluated for 72 h after dosing, respectively start of infusion. Theophylline was measured by high-performance liquid chromatography (HPLC). Absorption profiles (Wagner-Nelson) were derived and the absolute bioavailability was calculated. For the retard pellets a liberation/absorption of zero order for 12 h could be noted; the dose-corrected mean absolute bioavailability was 88%. The assumptions of an investigation on absolute bioavailability are discussed and the limitations on generalizability resulting therefrom.

[Experimental studies of extracellular matrix constituents for coating artificial prostheses before endothelialization].

Endothelialization seems to be a possibility to reduce thrombogenicity of artificial vascular prostheses. We have been occupied to find an ideal coating for the cells. Substances of extracellular matrix (ECM) were used: human fibronectin (HFN), laminin, collagen type I + III + IV. We have compared the influence of these materials with regard to adherence, sheer stress resistance and growth of cultivated human vena saphena endothelial cells (SEC). There was not any statistically significant difference according to cell proliferation. During a cultivation period of 7 days SEC increased by population doubling number 5 corresponding to a population doubling time of 76 hours. PTFE prostheses were coated with the materials, seeded with SEC and exposed to sheer stress in a physiological perfusion unit. Adherent SEC were counted after 24 hours of perfusion. Counts were correlated to the number of seeded cells. After perfusion only 23 +/- 4%, resp. 25 +/- 6% of cells were counted on collagen I + IV + HFN and on collagen I + IV + laminin. We found better results on collagen I + III and on collagen I + III + HFN; there were adherent 55 +/- 5%, resp. 53 +/- 7% after perfusion. These results document that there is no difference for SEC cultivation on different native components of basal membrane, under sheer stress conditions there seems to be differences in cell adherence.

[Influence of local anesthetics and narcotics on the energy metabolism of Ehrlich ascites tumor cells].

The suitability of Ehrlich ascites tumour cells as a test object for potential drugs affecting ATP-producing or consuming processes directly or indirectly is made likely by actions of local anesthetics and narcotics on the energy metabolism of these cells. The influence of local anesthetics on cation transport and Ca+(+)-dependent processes as well as the inhibition of NADH-ubiquinone reductase by narcotics is demonstrated for Ehrlich ascites tumour cells, too.

[The therapy of hypovolemic shock in cows with right-sided abomasal displacement].

In this trial, the effect of supplementary administration of the opioid antagonist Naloxone was tested, in comparison to infusion of Ringer's-solution only, in the treatment of hypovolemic shock in cows suffering from right-sided abomasal displacement. Twenty cows were selected by several criteria (pulse rate greater than 95/min; plasma chloride content less than 90 mmol/l; body temperature less than 39.5 degrees C) and alternatively assigned to the Naloxone group and the control group. All animals were treated surgically by the usual clinical method. After opening the abdominal cavity paralumbally from the right, intravenous administration of 13 l of Ringer's-solution was started and continued for about 5 hours. In addition, the cows of the experimental group received 0.75 g Naloxone as an intravenous bolus before, and 1 g Naloxone dissolved in 7 l fluid during the first hour of the permanent drip infusion. Further treatments (additional fluid therapy or administration of purgatives and ruminomotorics on the following days) were given according to the condition of the animal. The evaluation of efficacy was done in regard to the clinical (course of disease), and clinicochemical (e.g. hemogram, blood gas analysis, plasma chloride, plasma lactate) parameters as well as to blood pressure. No statistically significant differences were found between both groups. Only nine of the twenty cows were sent home cured, one patient of each group died one day after surgery, and the remaining 9 animals had to be sent to the slaughterhouse within 3 to 22 days after surgery after only transient improvement. These results suggest, that under the conditions described, conventional fluid therapy is equivalent to fluid therapy with addition of Naloxone. Hypoxic damage of the abomasum caused by its displacement and the subsequent hypovolemic shock are discussed as main causes for the poor postsurgical prognosis of right-sided abomasal displacement.

Stimulation of rat red blood cell glycolysis by phenylhydrazine hydrochloride.

The effects of phenylhydrazine hydrochloride (5, 10 and 20 mM) on the energy metabolism of red cells of rats were studied. There were found an 1.3-2-fold increase of lactate formation, an up to seven-fold accumulation of pyruvate and of the flux via the oxidative pentose pathway. The stimulation of the glycolysis was caused by 1) a large decline of the ATP concentration and a corresponding increase of ADP and AMP, and 2) by the stimulation of the actual activities of the Na+,K(+)-ATPase and Ca+(+)-ATPase owing to a greater permeability of the plasma membrane. A significant increase of the Ca+(+)-influx was demonstrated. A decline of the sum of adenine nucleotides after 60 and 90 min of incubation with phenylhydrazine was not accompanied by accumulation of hypoxanthine.

[Endemic poxvirus infection in white storks (Ciconia ciconia) and black storks (Ciconia nigra) in Switzerland].

Thirty young white storks (Ciconia ciconia) and black storks (Ciconia nigra) from a stork restocking station in Switzerland suffered from cutaneous, verrucous pox-efflorescences on legs, feet, beak and eyelids. The poxvirus was demonstrated ultrastructurally. As the endemic was in autumn, mosquitos may be a possible vector. It is uncertain, if the disease is connected with the previously imported storks from the Federal Republic of Germany.

[Cost-benefit analysis of the control and cure of enzootic bovine leukosis, described in the example of a selected territory].

Severe economic damage had been inflicted upon numerous industrialised dairy farms by enzootic bovine leucosis in a selected region with predominance of stepwise production. Initial contamination at the beginning of control action and sanitation had been as high as 98 percent. The annual damage, in monetary terms, had been 3,205,041. - Marks, according to calculations. Now, after eleven years of control and sanitation, eradication of enzootic bovine leucosis has come into sight. Even during these eleven years, control action yielded a benefit of 19,005,451. - Marks, that is 1,727,768.20 Marks per annum or 69. - Marks per cow and annum. It will be possible in forthcoming years to bring the cost factor down and, consequently, to further increase the benefit.

The action of N,N-diethyldithiocarbamate(DDC) on red blood cells.

Glutathione exerts a pro-oxidative effect by increasing H2O2-formation in the presence of methemoglobin and a radical-generating substance (e.g. DDC). This effect might contribute to the loss of glutathione in cells in which formation of methemoglobin is provoked by radicals.

[The normal kidney size in children. An ultrasound study].

Our clinical observation that the kidney size and volume of pediatric patients, determined by ultrasound, differ significantly from the standard value schedules (Weitzel) in general use led us to reevaluate the size and volume of normal kidneys in infancy and childhood sonographically. In a retrospective study, 205 out of 3500 children investigated sonographically in the years 1984-1987 at the University Hospital for Children in Ulm were selected and the ultrasound findings evaluated. Children with any disease that might possibly influence kidney growth were excluded. Ultrasound volume and size were analyzed statistically and showed a close correlation with body size, body weight and body surface. Our results show that the values in the Weitzel schedules are wrong.

[Injuries of the occipital condyles in fatal traffic accidents].

Among 600 deaths of traffic accidents we found injuries of the occipital condyles in 10 cyclists, 12 car drivers and 3 pedestrians. In most cases the collision speed ranged from 70 to 100 km/h. In 18 cases of direct head impact the more severe injury was on the opposite side, in 4 cases of indirect side impact (whiplash injury) on the same side. Most types of fractures were 12 horizontal disconnections and 4 fractures of the inner edges. In 9 cases the condyle fractures were on one side, in 16 cases on both sides.
