A testis antigen related to the brain D2 adhesion protein.

The localization of an antigen immunochemically crossreactive with the rat brain interneuronal adhesion molecule, D2-protein, has in testis been demonstrated by immunoelectrophoresis and immunocytochemistry. This D2-like antigen is localized to spermatids and residual bodies in testis, but it is absent both from spermatogonia and mature spermatozoa and from Sertoli cells. By immunoperoxidase electronmicroscopy D2-like antigen was observed in the head region posterior to the acrosomal membrane of late spermatids. In the nervous system D2-protein is involved in interneuronal adhesion. It is suggested that D2-like antigen in testis may be involved in a similar adhesion between late spermatids and Sertoli cells during spermiation.

Influence of conjugation reactions on the mutagenicity of aromatic amines.

2-Acetylaminofluorene (AAF) and 2-aminofluorene (AF), as well as their N-hydroxylated metabolites, N-OH-AAF and N-OH-AF, were studied for mutagenic effects in Salmonella typhimurium with rat- and mouse-liver S9 and microsomal subfractions in the presence of cofactors for glucuronidation and glutathione (GSH) transfer. Addition of UDPGA did not affect the mutagenicity of AAF, AF or N-OH-AAF under any experimental condition. Addition of GSH, on the other hand, markedly inhibited AAF, AF and N-OH-AAF. This seemed to be due to the direct effect of GSH, and not through an enzyme-catalyzed conjugation. Further, GSH inhibited the direct mutagenicity of N-OH-AF.

Mutagenicity of air samples from various combustion sources.

The emission of mutagens from various combustion sources was compared. Flue gas samples from power plants and boilers burning coal, oil and wood were studied. Little or no mutagenic activity was observed in samples from big boilers operated under optimal conditions. The mutagenic activity of emission samples from different boiler systems burning the same fuel varied considerably. This variation was larger than the difference obtained from boilers of comparable size utilizing different fuels. The highest mutagenic activity was observed in samples from a small coal combustion unit, utilizing the fluidized-bed technique. In this case the activity was highest without metabolic activation. Extracts from all samples contained toxic compounds that, in high doses, inhibited mutagenicity.

Mutagenicity of extracts from typewriter ribbons and related items.

Extracts of typewriter ribbons and carbon papers were found to be mutagenic in the Salmonella/microsome assay with strain TA98. Fractionation of ribbon extracts indicates that at least 2-3 different classes of mutagenic component are present in these extracts. Nitro-containing compounds may be responsible for the high mutagenicity observed for some of the ribbon extracts in the absence of S9. The results indicate that impurities in the products may be causing part of the mutagenic effect.

The effect of coronary artery bypass surgery on ventricular arrhythmias.

The effect of coronary artery bypass surgery (CAB) on ventricular arrhythmias (VA) was studied in a prospective investigation involving 32 patients (mean age 54 years) who underwent CAB because of severe stable angina pectoris. Prior to CAB as well as 12 months later each patient was subjected to the following investigational programme: resting ECG, exercise ECG, 24-h ECG, selective coronary arteriography, ventriculography and cardiac catheterization. Exercise ECG showed VA in only three patients. The prevalence of VA during 24-h ECG was 56 and 66% on the two occasions (NS), while complicated VA (multiform, repetitive, R on T) was seen in 18 and 28%, respectively (NS). The persistence (number of 6-h periods showing VA) was 33 and 47% with regard to any VA (P less than 0.05), while complicated VA occurred in 13 and 15% of the 6-h periods (NS). Except for an increase in dp/dtmax/P at the postoperative measurement (P less than 0.05), no significant change in the performance of the left ventricle was seen after CAB though the graft patency was 77%. It is concluded that in patients with 'medically intractable' stable angina pectoris, CAB does not effect the occurrence of VA to any great extent--probably because left ventricular function is unchanged one year after as compared with that prior to CAB.

Mutagenicity in emissions from coal- and oil-fired boilers.

The mutagenicity of emission samples from three oil-fired and four coal-fired boilers have been compared by using the Salmonella/microsome assay. Very little or no mutagenic activity was observed in samples from five of these boilers. The sample from one oil-fired boiler showed mutagenic activity of about 500 revertants/MJ, and the sample from a coal-fired fluidized bed combustor had an activity of 58,000 revertants/MJ measured with strain TA 98 in the absence of metabolic activation. All samples contained substances that were cytotoxic to the test bacteria, thus making it difficult to obtain linear dose-response curves. Mutagenic activity at low levels may remain undetected due to this toxicity of the samples. Samples with mutagenic activity below the detection limit in the Salmonella test have also been tested for forward mutations at the HGPRT locus in V79 hamster cells. Weak mutagenic effects were detected in two of the samples, whereas the sample from one oil-fired boiler remained negative. In this test, as well as in the Salmonella test, a strong cytotoxic effect could be observed with all samples.

Bioassay of extracts of ambient particulate matter.

Organic extracts from airborne particles collected at various sites in Scandinavia have been tested for mutagenicity in the Ames Salmonella/microsome assay. Extracts from particles in the respirable size fraction (diameter less than 3 microns) were mutagenic with and without metabolic activation. The mutagenic activity varied from day to day, mainly due to variations in meteorological parameters, especially wind speed and atmospheric stability. A seasonal variation could also be observed, with the highest average values in winter time. Samples collected in urban areas were considerably more mutagenic than samples from background areas. The results suggest that exhaust from motor vehicles are the most important source of mutagenic particles in urban areas. Comparison of roof top samples with street level samples indicated that atmospheric reactions cause transformation of nonpolar compounds in the primary emission to more oxygenated mutagenic compounds. It is, however, not known to which degree this causes an overall increase of the mutagenic activity. The mutagenic activity of emissions from stationary combustion sources have also been studied, and residential heating by burning solid fuels in small combustion units have been shown to be a major contributor to mutagens in the environment.

Detection of mutagens in complex samples by the Salmonella assay applied directly on thin-layer chromatography plates.

A new technique is reported in which components in complex samples are separated on thin-layer chromatography plates and their mutagenic effect is registered directly on the plates by means of the Salmonella assay. The method is quick and simple and particularly useful for screening large numbers of environmental samples. Qualitative comparisons of mutagens in different samples can easily be made. REgistered mutagens can be identified by the chemical analysis of extracts from duplicate plates.

Overdosage of antidepressants: clinical and pharmacokinetic aspects.

Twenty-nine cases of self-poisoning with antidepressants (amitriptyline, imipramine, clomipramine, maprotiline, doxepine, nortriptyline, opipramol) were examined by frequent observation of CNS effects, heart rate, blood pressure and standard ECG, 24h-ECG-monitoring, measurement of systolic time intervals, EEG recordings and frequent measurement of serum levels of antidepressants and primary metabolites. None of the patients died. Maximum total serum antidepressant level (parent compound + desmethyl metabolite) ranged from 20 to 2200 micrograms/l, with concentrations above 500 micrograms/l in 11 cases. The serum amitriptyline concentration remained high for 3-4 days in some of the severely intoxicated patients and the decay curves were compatible with partly saturated elimination. A degree of unconsciousness and the occurrence of excitation and hallucinations were generally seen in cases with total serum antidepressant levels above 500 micrograms/l. Grand mal seizures occurred more frequently at high antidepressant levels, but could not be predicted from the EEG recordings. Increased heart rate and prolonged QRS- and QTc-intervals were significantly correlated with the total serum antidepressant level. 24 h-ECG-monitoring revealed no serious arrhythmias or instances of heart block. Hypotension was only seen initially in few patients. Systolic time interval measurements showed changes suggesting impaired myocardial performance (elevated PEP/LVET ratio) at intermediate (60-500 micrograms/l) but not high (greater than 500 micrograms/l) total serum antidepressant levels. Measurement of serum concentration in antidepressant intoxication is important for identification of patients with high serum levels and the corresponding risk of developing toxic reactions, and to exclude patients with a low concentration who do not require intensive observation.

Immunocytochemical localization of the D2-protein in rat retina.

The localization of the D2-protein in rat retina was investigated by quantitative immunoelectrophoresis and by immunoperoxidase staining on both the light microscopic and the electron microscopic level. The D2-protein was enriched 1.4 times in retina compared to whole brain and its was located to most presynaptic and some postsynaptic membranes in the inner an outer plexiform layers. Moreover, the D2-protein was located on the cell membrane confining the ciliary neck region connecting the inner and the outer segments of photoreceptor cells. Thus, on the electron microscopic level, the D2-protein is not restricted to the presynaptic membrane, but it is also found in other parts of the neuronal surface membrane.

Cardiovascular effect of imipramine and nortriptyline in elderly patients.

Cardiovascular effects in elderly depressed patients (age 62-78 years) treated with imipramine (N = 11) or nortriptyline (N = 10) were recorded by monitoring of heart rate, blood pressure, systolic time intervals, standard ECG and 24-h ECG. The two drugs exhibited distinctly different cardiovascular reactions. The use of imipramine was severely limited by orthostatic hypotension occurring at subtherapeutic plasma levels, which resulted in falls with fracture in two patients. In contrast, nortriptyline at therapeutic drug levels did not significantly influence orthostatic blood pressure regulation. Nortriptyline caused moderate changes in systolic time intervals, indicating impairment in myocardial contractility. This effect was not seen with imipramine, but a majority of the patients did not reach therapeutic plasma levels because of blood pressure reactions. Neither imipramine nor nortriptyline induced changes in cardiac conduction time measurements or arrhythmias. In addition to the blood pressure reactions, the use of imipramine was complicated by dose dependent kinetics.
