Brugia pahangi: identification and characterization of an aminopeptidase associated with larval molting.

The third-stage larvae (L3) of the filarid parasite Brugia pahangi were surveyed for protease activity beginning with release from the mosquito vector through molting to the fourth-stage larvae (L4). A metalloaminopeptidase with a substrate preference for phenylalanine was released during the molting process. A screen of aminopeptidase inhibitors identified H-boroPhenylalanine-(pinacol) as highly effective, with a Ki of 1.3 x 10(-11) M. This peptidase inhibitor also arrested molting of L3 larvae in vitro at a concentration of 100 microM.

Learning styles of LD and NLD ADHD children.

Attention Deficit Hyperactive Disorder (ADHD) children (N = 26) with a learning disability (LD) and without (NLD) were compared on measures of information processing using the Learning Efficiency Test (LET). Method of presentation (visual vs. auditory), order of recall (ordered vs. non-ordered), and memory processes (immediate, short-term, and long-term) were assessed. While both groups showed more difficulty with short-term and long-term memory processes than with immediate memory processes, the ADHD-LD subjects demonstrated more difficulty processing information effectively. Ordered recall was significantly more difficult for both short-term and long-term memory processes than was unordered recall. While both groups demonstrated difficulty with auditory ordered recall under short-term and long-term conditions, the ADHD-LD subjects lost the most information under auditory conditions.

Protection against fludarabine neurotoxicity in leukemic mice by the nucleoside transport inhibitor nitrobenzylthioinosine.

Fludarabine phosphate (F-ara-AMP, Fludara) is rapidly converted in the circulation to fludarabine (F-ara-A) and is among the most effective single agents in the treatment of chronic lymphocytic leukemia. Although current treatment protocols are well tolerated, severe neurotoxicity was a consequence of high-dose F-ara-AMP regimens used in early phase I trials against adult acute leukemia. The present study showed that in mice implanted with leukemia L1210, fatal neurotoxicity, which initially manifested as hind-limb paralysis, was a consequence of high-dose F-ara-AMP treatment. However, the incidence of neurotoxicity was reduced by the coadministration of NBMPR-P, the 5'-phosphate of nitrobenzylthioinosine, a potent inhibitor of the es equilibrative nucleoside transport (NT) system. NBTGR-P, the 5'-phosphate of nitrobenzylthioguanosine (also a potent NT inhibitor) similarly prevented F-ara-AMP neurotoxicity in this experimental system. Treatment with F-ara-AMP/NBMPR-P combinations was more effective with respect to the fractional yield of "cured" mice than were the same treatment regimens without NBMPR-P.

Immunoglobulin E recognition of Dirofilaria immitis antigens is more specific than immunoglobulin G.

The chronological development of the serum IgE and IgG response to microfilaria, third and fourth stage larvae, and male and female adult Dirofilaria immitis was examined by enzyme-linked immunosorbent assay (ELISA) and enzyme-linked immunoelectrotransfer blot (EITB). Dirofilaria immitis-specific IgE and IgG levels peaked 16-18 weeks post-infection after increasing in response to the fourth larval molt. Specific IgG levels plateaued after patency, while IgE continued to decline. The use of ammonium sulfate cut sera showed there was no quenching or blocking of IgE binding by IgG in the ELISA and EITB methods used in this study. IgE-specific EITB showed 30-49 bands for the five respective extracts that were identified by M(r) or relative mobility. Eighty-five to 100 bands were visualized by IgG-specific EITB for the same five extracts. The isotype-specific ELISA and EITB were shown to be closely related by significant correlations (P < 0.0001) between S/N ratios and the number of bands found on blots. The isotype-specific EITB bands non-specifically recognized were greater in size than 21 kDa for IgG and 45 kDa for IgE. Recognition of bands changed over time with some bands being recognized only by prepatent sera. Ten antigen bands of seven M(r) were consistently and specifically recognized by IgE in the five-stage extracts by sera from prepatent and patent infections; only one such M(r) at 13.9 kDa, was described for IgG. A potentially diagnostic 31.9 kDa antigen band was identified on the IgE-specific EITB of D. immitis female extract and was shown to be recognized by IgE in sera from all infected dogs at all time points examined from 2 weeks until 1 year post-inoculation. Overall, IgE reactivity was more specific for D. immitis infections than IgG reactivity.

Oscillations of cytosolic sodium during calcium oscillations in exocrine acinar cells.

In acinar cells from rat salivary glands, cholinergic agonists cause oscillations in cytoplasmic free calcium concentration, which then drive oscillations of cell volume that reflect oscillating cell solute content and fluid secretion. By quantitative fluorescence ratio microscopy of an intracellular indicator dye for sodium, it has now been shown that large amplitude oscillations of sodium concentration were associated with the calcium and cell volume oscillations. Both calcium and sodium oscillations were dependent on the continued presence of calcium in the extracellular medium and were abolished by the specific sodium-potassium adenosine triphosphatase inhibitor ouabain. Thus, calcium oscillations in salivary acinar cells, by modulating the activities of ion transport pathways in the plasma membrane, can cause significant oscillations of monovalent ions that may in turn feed back to regulate calcium oscillations and fluid secretion.

A cell-free culture system for development of large numbers of Brugia pahangi larvae.

Techniques used in the in vitro culture of massive numbers of Brugia pahangi third-stage larvae (L3) are described. Procedures for larval preparation and four culture conditions, with or without animal cell co-cultures, were studied, resulting in the adoption of a relatively simple cell-free culture system for routine harvesting of larval moulting excretory/secretory product.

Evaluation of a 'dipstick' immunoassay to detect cysticercosis in experimentally infected cattle.

A 'dipstick' immunoassay for bovine cysticercosis, using an antigen isolated from Taenia hydatigena cyst fluid, was evaluated in cattle experimentally infected with Taenia saginata. The assay correctly identified six out of seven infected cattle, including an animal in which only 12 living cysticerci were found. Cattle became seropositive as early as 3 weeks post-infection. A false-negative reaction was found for one very lightly infected animal, from which only four living cysticerci were recovered at necropsy. The assay was also used to detect circulating antibodies in experimentally infected cattle before and after therapeutic treatment with anthelminthics. The results suggest that praziquantel-treated animals gradually revert to being seronegative after the cysticerci are killed.

Motivation and academic achievement: the effects of personality traits and the quality of experience.

The present study examined the relationship of personality, experience while studying, and academic performance. One hundred and seventy talented high-school students (68 males, 102 females) completed the Personality Research Form (PRF) and recorded their experience via the Experience Sampling Method (ESM). The results showed that controlling for ability, work orientation, a PRF factor, was a better predictor of grade than experience. However, an experiential variable, intrinsic motivation while studying, was related to the difficulty level of courses students took over the 4 years of high school. The results supported the notion that there are two kinds of motivation in scholastic achievement, one directed toward long-term goals, the other directed toward ongoing experience.

Long-term effects of physiologic concentrations of dexamethasone on human bone-derived cells.

Bone cells derived from human trabecular explants display osteoblastic features. We examined the modulation of alkaline phosphatase activity and cAMP production as the result of exposing trabecular explants to physiologic concentrations of dexamethasone for 4 weeks during cellular outgrowth and subculture. Cells treated with dexamethasone were observed to grow generally more slowly than control cells. Cells appeared larger and more polygonal, and staining for alkaline phosphatase was more intense in the dexamethasone-exposed cultures. There was a progressive increase in cellular PTH responsiveness with increasing duration of exposure of cells to dexamethasone. Cells grown for 6 weeks in 3 x 10(-8) M dexamethasone had a 10-fold increase in PTH-stimulated cyclic AMP accumulation. Dexamethasone-treated cells also had a significantly increased alkaline phosphatase activity. 1,25-(OH)2D3-stimulated alkaline phosphatase activity was increased approximately 20-fold. cAMP responses were significantly increased to PTH (21.7-fold), PGE1 (2.67-fold), and forskolin (4.81-fold), but not to cholera toxin. Dexamethasone-treated cells also had a mean decrease in 1,25-(OH)2D3-stimulated osteocalcin production to 26.2% of control values (p less than 0.001). Hydrocortisone treatment gave rise to similar effects but of smaller magnitude than those of dexamethasone. Testosterone did not have a significant effect on alkaline phosphatase activity or cAMP production. Skin fibroblasts showed a significant enhancement of alkaline phosphatase activity in response to dexamethasone, but of a much smaller magnitude than in bone cells. The phenotypic changes induced by long-term culture in dexamethasone are consistent with the promotion of a more differentiated osteoblastic phenotype.

New antihepatotoxic naphtho-pyrone glycosides from the seeds of Cassia tora.

Two new naphtho-pyrone glycosides, 9-[(beta-D-glucopyranosyl-(1----6)-O-beta-D-glucopyranosyl)oxy]-10- hydroxy-7-methoxy-3-methyl-1H-naphtho[2,3-c]pyran-1 -one (5) and 6-[(alpha-apiofuranosyl-(1----6)-O-beta-D-glucopyranosyl)oxy]- rubrofusarin (6), together with cassiaside (3) and rubrofusarin-6-beta-gentiobioside (4) were isolated from the seeds of Cassia tora L. Their structures were elucidated on the basis of chemical and spectral data. The naphtho-gamma-pyrone glycosides (3, 4, and 6) were found to have significant hepato-protective effects against galactosamine damage, which were higher than that of silybin from Silybum marianum.

Enzyme immunoassay of Dirofilaria immitis-specific IgE in infected dogs.

An ELISA procedure was developed for monitoring the specific IgE response in dogs to Dirofilaria immitis infection. The results of this assay correlated well with, and appeared to be more sensitive than, the passive cutaneous anaphylaxis test. The IgE ELISA values of the positive reference serum and the passive cutaneous anaphylaxis test results showed that a serum to negative absorbance ration of 1.45 was statistically significant for discrimination and was used to evaluate the specific IgE response in the sera from 90 clinically diagnosed heartworm cases. This ELISA procedure was more sensitive, as it detected 78% of the 90 cases as compared to a detection rate of only 43-47% by IgG ELISA or IFA. Sera obtained from 23 experimentally infected dogs at 4-week intervals for 20 weeks post-infection, were assayed for D. immitis-specific IgE by ELISA. A group of the infected dogs was also treated with diethylcarbamazine during the course of infection. All the experimentally infected dogs developed a specific IgE response, with treated dogs generally responding earlier.

Experimental infection in a human subject by a possibly undescribed species of Taenia in Taiwan.

A cysticercus of a possibly undescribed species of Taenia which occurs commonly in Taiwan aborigines was used to establish an experimental infection in a human volunteer. Symptomatic effects attributed to the infection included diarrhoea, upper abdominal pain, and increase or loss of appetite over a four-month period. After an expelled proglottid was observed 122 days post-exposure, eggs and proglottids were found continuously until the patient was treated with anthelmintics. Antibody titres measured by enzyme-linked immunosorbent assay and levels of eosinophilia seemed to correlate with symptoms. Haematological analyses revealed an abnormal lipid metabolism during the entire symptomatic period.

A potential diagnostic reagent for bovine cysticercosis.

A fraction of larval Taenia hydatigena cyst fluid was shown to have high sensitivity and specificity in the enzyme-linked immunosorbent assay (ELISA) for the detection of bovine antibodies to the heterologous parasite Taenia saginata. This antigenically active lipoprotein fraction was isolated by ultracentrifugal density flotation using either ammonium sulfate (specific gravity = 1.231 g per ml) or NaCl/KBr (specific gravity = 1.225 g per ml), followed by ion-exchange chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) indicated that this fraction was composed of high molecular weight (65,000 to 77,000 Mr) and low molecular weight (9,500 to 16,000 Mr) proteins. Electrophoresis under non-denaturing conditions in either acrylamide (5%) or agarose (1%) resulted in 1 major diffuse band staining for both protein and lipid. The high and low molecular weight proteins observed on SDS-PAGE under reducing conditions could not be resolved by gel filtration chromatography and emerged as a single lipoprotein peak. This T. hydatigena cyst fluid fraction appears promising as a diagnostic reagent in the ELISA for bovine cysticercosis.

Sensitivity and specificity of the indirect-fluorescent antibody test and two enzyme-linked immunosorbent assays in canine dirofilariasis.

Ninety dogs naturally infected with Dirofilaria immitis and 103 noninfected dogs, as determined by necropsy, were used to compare the sensitivity and specificity of the cuticular and somatic reactions of the indirect fluorescent antibody test (IFA-C and IFA-S, respectively) and 2 enzyme-linked immunosorbent assays (ELISA). In microfilaremic and amicrofilaremic heartworm-infected dogs, negative results were common for all serotests. In dogs without adult heartworms at necropsy, 32% to 49% were positive, using 1 ELISA, 27% to 29% were positive with the other ELISA, 15% to 36% were positive with the IFA-S, and 0% to 1% were positive using the IFA-C, depending on the classification of borderline reactions. The prevalence of false positive serotests was probably not due to the detection of precardial stages of D immitis in dogs obtained from areas of low endemicity. Until the causes of the false-positive tests are resolved, the use of currently available serotests for routine diagnostic screening or as criteria for instituting treatment is not recommended.

Effects of verapamil on coronary hemodynamic function and vasomobility relative to its mechanism of antianginal action.

The effect of intravenous verapamil on systemic and coronary hemodynamic function was studied at cardiac catheterization in 12 patients with coronary artery disease. Verapamil was administered as a 2-minute bolus (0.145 mg/kg) followed by an infusion (0.005 mg/kg/min). Cardiac output and coronary sinus blood flow were measured by thermodilution techniques. Caliber of the large coronary arteries and of diseased segments was determined from the coronary angiogram using a computer-assisted method. Verapamil reduced mean arterial pressure 14% (p less than 0.001), systemic vascular resistance 21% (p less than 0.01), and stroke work index 16% (p less than 0.001). Coronary vascular resistance decreased 24% (p less than 0.01) with a small increase in coronary sinus blood flow (+13%, difference not significant [NS]). Myocardial oxygen consumption determined in 5 patients showed no significant change with verapamil. Luminal area in 39 coronary lesions was measured in the "normal" portion of the diseased segment and at its maximal constriction, and an estimate of flow resistance in the stenosis was computed. Overall, 50% of "normal" and of diseased coronary segments dilated significantly with verapamil. Stenosis dilation resulted in an average 14% reduction (p less than 0.01) in estimated flow resistance. In 8 patients, the luminal changes (n = 27) induced by sublingual nitroglycerin were compared with those induced by verapamil. Nitroglycerin induced a significantly greater increase in coronary caliber in both normal and diseased segments; estimated stenosis flow resistance decreased 28% with nitroglycerin compared with 14% with verapamil (p less than 0.01). Thus, verapamil moderately dilates the systemic and coronary small vessel resistance bed without apparently increasing myocardial metabolic demand. Furthermore, verapamil mildly dilates large coronary conductance vessels in both "normal" and diseased segments, although significantly less than does nitroglycerin.

Projecting average family size in rural India by the analytic hierarchy process.

"In this study, cultural, economic as well as certain crucial demographic factors are considered as the determinants for projecting the average family size in rural India.... The Analytic Hierarchy Process [is used] to analyze influences of the factors which enter implicitly in a rural couple's decision-making to determine the number of children they want to have as time goes by. [The authors] did not attempt to make distinctions among the regional differences in rural India. The outcome projected in [this] analysis compares favorably with the results of other demographic studies."

Light and electron microscopic pulmonary lesions associated with retained microfilariae in canine occult dirofilariasis.

Lungs from dogs infected with adult, gravid Dirofilaria immitis but without microfilaria circulating in systemic vessels were studied by light and transmission electron microscopy. Microfilariae were seen within the pulmonary microvasculature and in interstitial areas of interalveolar septa. An acute inflammatory reaction characterized by aggregates of eosinophils, neutrophils, macrophages and lymphocytes was associated with intact microfilariae. Necrotic and fragmented microfilariae were surrounded by macrophages and epithelioid cells. Other lesions in interalveolar septa of dogs with chronic occult dirofilariasis included endothelial degeneration and hypertrophy, perivascular aggregates of plasma cells, hypertrophy and hyperplasia of smooth muscle cells, and interstitial fibrosis. The findings suggest that a cell-mediated hypersensitivity reaction is at least partially responsible for retention of microfilariae in pulmonary parenchyma of dogs with occult dirofilariasis.
