[Digital subtraction angiography of the hand].

The report deals with 66 digital subtraction angiograms of the hand performed on 61 patients; 60 were carried out by direct puncture of the brachial artery and six by trans-femoral catheterisation. In 39 patients a peripheral organic vascular abnormality was demonstrated (usually arterial sclerosis, thromboangiitis obliterans or collagenosis). Purely functional flow abnormalities were found in 17 patients. In 15 patients an organic abnormality was demonstrated, but its type could not be identified. Intra-arterial DSA is able to differentiate between functional abnormalities of blood flow and peripheral organic vascular abnormalities.

Localization of phycoerythrin at the lumenal surface of the thylakoid membrane in Rhodomonas lens.

The thylakoids of cryptomonads are unique in that their lumens are filled with an electron-dense substance postulated to be phycobiliprotein. In this study, we used an antiserum against phycoerythrin (PE) 545 of Rhodomonas lens (gift of R. MacColl, New York State Department of Health, Albany, NY) and protein A-gold immunoelectron microscopy to localize this light-harvesting protein in cryptomonad cells. In sections of whole cells of R. lens labeled with anti-PE 545, the gold particles were not uniformly distributed over the dense thylakoid lumens as expected, but instead were preferentially localized either over or adjacent to the thylakoid membranes. A similar pattern of labeling was observed in cell sections labeled with two different antisera against PE 566 from Cryptomonas ovata. To determine whether PE is localized on the outer or inner side of the membrane, chloroplast fragments were isolated from cells fixed in dilute glutaraldehyde and labeled in vitro with anti-PE 545 followed by protein A-small gold. These thylakoid preparations were then fixed in glutaraldehyde followed by osmium tetroxide, embedded in Spurr, and sections were labeled with anti-PE 545 followed by protein A-large gold. Small gold particles were found only at the broken edges of the thylakoids, associated with the dense material on the lumenal surface of the membrane, whereas large gold particles were distributed along the entire length of the thylakoid membrane. We conclude that PE is located inside the thylakoids of R. lens in close association with the lumenal surface of the thylakoid membrane.

Identification of a single nucleotide C-to-T transition and five different deletions in patients with severe hemophilia B.

DNA of 70 unrelated hemophilia B patients, including three inhibitor patients, was analyzed by using various restriction enzymes and was hybridized with both a factor IX cDNA and 3'- and 5'-flanking probes. When the gene was mapped this way, six patients all afflicted with severe hemophilia B were shown to have a deviating hybridization pattern. One inhibitor patient showed a partial deletion of about 9 kb that removes exons a-c. A partial deletion of at least 11 kb that removed exon a and that had a maximum size of 35 kb in the 5'-flanking region could be identified in a patient of unknown status. In another three noninhibitor patients a complete deletion of the factor IX gene and two partial deletions could be observed. The partial deletions are of approximately 8 kb and approximately 1.5 kb, removing exons d and e and exon g, respectively. As detected by oligonucleotide probing, a C-to-T transition at amino acid 338 gave rise to an altered TaqI restriction pattern that could be observed in a sixth patient. The other 64 hemophilia B patients, including two inhibitor patients, showed a hybridization pattern indistinguishable from a normal one.

Haemophilia A: carrier detection by DNA analysis.

From 46 families of predominantly German origin, afflicted with haemophilia A, 178 females were tested for carrier status. Two polymorphic restriction endonuclease sites, the extragenic marker locus DXS 52 (St 14 probe) and the intragenic Bcl I RFLP were investigated in these families. In some cases the results were corroborated by identifying (i) deletions within the factor VIII:C gene and (ii) eliminating a restriction endonuclease site. Two new alleles of the DXS 52 marker locus were found. According to this strategy, 27 women were classified as carriers and 74 as non-carriers. Forty-six women were classified as carriers according to pedigree analysis. Twenty-five females of families with sporadic cases and 6 test persons, who had mothers who where homozygous for the marker alleles, were diagnosed by additional use of conventional carrier detection.

[Comparative study of the amplitude, surface and duration of the pre- and post-imperative waves of the CNV (contingent negative variation) in a group of 40 schizophrenic and schizo-affective patients as compared with normal subjects].

In 27 normal adult subjects, pre- and postimperative phases of the CNV had an homogeneous pattern. The return to the baseline was of short duration (320 +/- 170 msec) whatever the amplitude and surface. This is in favor of an active mechanism terminating the CNV. In 27 schizophrenic and 13 schizoaffective patients, pre- and postimperative phases of the CNV were heterogeneous in character suggesting the possibility of multiple disturbances. When present, the preimperative negativity was of lower amplitude and had a delayed return to the baseline with a wider dispersion. In the patients with preimperative negativity followed by a PINV (postimperative negative variation), a linear relationship has been found between the amplitudes of the two waves.

[Prostaglandin E1 in stage III and IV arterial occlusive diseases. results of a multicenter study].

In a controlled randomized trial at four centers, using a common protocol, 57 patients with advanced chronic arterial occlusive disease (21 in stage III, 36 in stage IV) were treated with prostaglandin E1 (PGE1) or adenosine triphosphate (ATP) for three weeks. Both substances were administered intraarterially over 60 min. Daily dose of PGE1 was 20 micrograms, of ATP 30 mg. Both produced a significant reduction in resting pain at the end of the treatment phase, in stage III significantly better with PGE1. There was also a clear reduction in the use of analgesics, significantly more so with PGE1. Healing or improvement of ulcers was significantly better with PGE1, while there was no significant differences between the two drugs as regards stage improvement. Three amputations had to be performed in the PGE1 group, nine in the ATP group, a significant difference. Side effects in the form of reddening, pain and swellings occurred in 15 patients of the PGE1 group and six of the ATP group. Final verdict by the treating doctor about the success of treatment was significantly more favorable for PGE1.

[Two-dimensional ultrasonics--a non-invasive method for the diagnosis of arteriosclerotic changes in the region of the supra-aortic branches].

2-D ultrasound examinations were performed on 98 patients suspected of arteriosclerotic changes in the region of the extracranial arteries of the neck. The findings were compared with those obtained by angiography, including digital subtraction angiography. In 84 patients there was concordance between the results of 2-D ultrasound and angiography. In six patients the B-scan alone gave false results. In eight patients no definite diagnosis could be made. The addition of 2-D ultrasound with bidirectional Doppler reduced false diagnoses to three and uncertain diagnoses to five. The combination of 2-D ultrasound and bidirectional Doppler thus increases the diagnostic possibilities to detect arteriosclerotic changes in the region of the extracranial cervical arteries of the neck.

[Treatment of Raynaud's syndrome with captopril].

Twenty patients with primary or secondary Raynaud's phenomenon were, after a 4-week treatment-free interval, given captopril ( Lopirin ) at a dosage of 25 mg three times daily for 84 days. Marked subjective improvement was reported by 14 patients. Digital-artery pressure, blood flow at rest and blood flow during reactive hyperaemia increased. Blood flow at rest increased significantly after cooling (10 degrees C for three minutes).
