[Effect of zhuyu huatan decoction for cellular and humoral immunity in cerebral hemorrhage].

Fifty-one patients of cerebral hemorrhage were observed on controlled study which were treated with the Zhu-Yu Hua-Tan Decoction (ZYHTD) and Western medicine. The results revealed that ZYHTD could reduce the serum IgG (P < 0.05), and lymphocyte transformation rate (P < 0.05). Meanwhile, the patient's health condition of treated group was better than the control group in the fields of eliminating brain hematoma, evaluating patients condition score in TCM and compromised neurologic function (P < 0.05). It is assumed that there is positive significance in anti-inflammatory effect and promoting the absorption of brain hematoma in the acute phase of cerebral hemorrhage.

Expression of a viral oncoprotein during mammary gland development alters cell fate and function: induction of p53-independent apoptosis is followed by impaired milk protein production in surviving cells.

The disruption of cell cycle regulation is associated with developmental abnormalities and tumorigenesis. The SV40 large T antigen (Tag) interferes with cell cycle control by interacting with the pRb family and p53. Mice carrying a transgene composed of the whey acidic protein (WAP) gene promoter and the Tag coding sequence express Tag during pregnancy and are unable to nurse their young. Tag expression induced apoptosis in mammary epithelial cells during late pregnancy. At least 5% of mammary epithelial cells were undergoing apoptosis at any one time. In contrast, less than 0.2% of mammary epithelial cells in nontransgenic littermates was undergoing apoptosis. Apoptosis in Tag mice was associated with increased steady-state RNA levels of bax and bcl-xL + S, with a relative increase in bcl-xs expression. Since p53 was sequestered by Tag, it is likely that p53-independent mechanisms precipitated apoptosis. The Tag-expressing mammary alveolar cells that did not undergo apoptosis continued to differentiate through late pregnancy, as measured by the sequential activation of milk protein gene expression. However, milk protein production, processing, and secretion was impaired, resulting in lactation failure.

Apoptosis and remodeling of mammary gland tissue during involution proceeds through p53-independent pathways.

Mammary gland involution is a physiological process that follows lactation and results in the rapid disappearance of the entire lobulo-alveolar compartment. Coincident with the onset of involution, milk protein gene expression ceases and alveolar cells undergo programmed cell death. In mammary epithelial tissue culture cells in vitro, both p53-dependent and p53-independent apoptosis pathways have been identified. We investigated whether p53 induces apoptosis during mammary gland involution in vivo and participates in tissue remodeling. Toward this end, we examined the process of involution in the presence and absence of functional p53 in mouse models: wild-type, transgenic mice that express SV40 T-antigen specifically in mammary tissue during pregnancy; and mice that carry nonfunctional p53 alleles in their germ line. Mammary gland whole-mount and histological analyses revealed that involution and remodeling, with the concomitant disappearance of the lobulo-alveolar structures, proceeded normally in the absence of functional p53. In addition, the absence of functional p53 did not alter the involution related pattern of bax (death inducer) gene expression or the ratio of RNAs encoding bcl-xs (death inducer) to bcl-xL (survival inducer).

Beneficial effects of captopril on prognosis in patients with acute myocardial infarction. Shanghai Secondary Prevention of Acute Myocardial Infarction Study Group.

OBJECTIVES: To assess the effects of early and long-term angiotensin-converting enzyme inhibitor treatment with captopril on clinical outcome in patients with acute myocardial infarction (AMI). METHODS: Eight hundred and twenty-two patients with AMI who were hospitalised within 72 hours of symptoms and had no cardiogenic shock were randomly allocated to captopril (n = 478, Group I) and conventional treatment (n = 344, Group II). Cardiac events including congestive heart failure, reinfarction, severe arrhythmias and cardiac death during hospitalization and follow-up period (average 20 months) were determined. RESULTS: The overall mortality rate during hospitalization was lower in group I than in group II (P = 0.0001), this was true for patients with anterior (P = 0.0003), inferior (P = 0.0411) and anterior inferior AMI (P = 0.0232). During follow-up, despite similar occurrence rate of reinfarction and severe arrhythmias in the two groups, the mortality rate (P = 0.0324) and total cardiac event rate (P = 0.055) were lower in group I than in group II. CONCLUSIONS: After AMI, early and long-term treatment with captopril exerts a beneficial effect on the prognosis of patients.

Cryopreservation of embryos in an IVF-ET program.

OBJECTIVE: To evaluate the use of cryopreservation of embryos in an in vitro fertilization and embryo transfer (IVF-ET) program. METHODS: Three cases of successful pregnancy after frozen-thawed embryo transfer were reviewed. The Testart's slow freezing and rapid thawing method was chosen. RESULTS: The first case was a Turner's syndrome patient and her husband had azoospermia. Donated frozen-thawed embryo transfer was done after hormonal replacement therapy. Pregnancy was achieved and a normal baby was delivered. The other two cases of IVF-ET did not achieve pregnancy during the stimulated cycle but became pregnant with frozen-thawed embryos in the natural cycle 4-5 months later. CONCLUSIONS: Pregnancy could be achieved in couples with incurable reproductive defects with donated embryos cryopreserved and ready for use whenever the endometrium is prepared by hormonal replacement therapy to be synchronous with the embryo development. In an IVF-ET program, one egg-retrieval may serve for many future chances of embryo transfer and therefore increase the cumulative pregnancy success rate.

[Changes in intracellular K+ concentration and ATPase activity of myocardiocytes in early stage of burn injury].

In this experiment, intracellular K+ concentration ([K+]i) and ATPase activity of myocardiocytes were measured in early stage of burn injury. Comparing with control group, it was found that, 1. [K+]i were decreased after burn injury, [K+]i of 1st, 3rd, 8th and 24th hours were decreased to 96.2 +/- 1.3%, 85.8 +/- 1.3%, 65.9 +/- 1.0% and 73.7 +/- 1.1% of normal, respectively. 2. Cardiac sarcolemma total ATPase, Mg(2+)-ATPase and Na(+)-K(+)-ATPase activities were all reduced significantly at 8th hour after injury. These results suggest that, burn injury accelerates K+ efflux current, but inhibits K+ influx current, and the reduction of Na(+)-K(+)-ATPase activity is one reason of decrease of [K+]i after injury.

[Establishment of in vitro culture of bovine trabecular meshwork cells and their phagocytosis].

OBJECTIVE: To study the role of abnormality of phagocytosis of trabecular meshwork cells in the pathogenesis of glaucoma. METHOD: In vitro trabecular meshwork cell culture was successfully established from excised bovine aqueous outflow pathway. Using latex microspheres 0.6 micron in diameter as markers, we investigated the phagocytosis of cultured bovine trabecular cells and its influential factors qualitatively and quantitatively. RESULTS: Immunohistochemical staining studies have shown that trabecular cells secrete fibronectin and laminin. It is shown that bovine trabecular cells in vitro had strong phagocytic ability to ingest the particles, the number of latex beads in cells was increased with the prolongation of time in culture, and by 24 hours of incubation, the average number of beads in each cell was 40.5 +/- 6.7, in the mean time marked morphological and damaging changes, even death, of trabecular cells occurred. Epinephrine and cortisone inhibited significantly the phagocytosis of cells (P < 0.05), while epidermal growth factor had no such effect. CONCLUSION: The phagocytosis of trabecular cells is important to keep aqueous humour outflow patent, and abnormal phagocytosis of trabecular cells might be related to the pathogenesis of primary glaucoma.

[Sequencing of right terminal fragment of canine adenovirus type 1 vaccine strain DNA].

Current studies show that adenoviruses are more advantageous as expression vectors than other viruses. Canine adenovirus type 1 (CAV-1) vaccine strain, CLL (cannaught laboratory limited), as vectors will play more important role for the prophylaxis of zoonosis than human adenoviruses. Studies on molecular biology of CAV-1 or CLL are necessary for construction of vectors of CLL. In this paper, the right terminal of CLL DNA cleaved by Hind II was sequenced. Authors found that it contained a 238 bp inverted terminal repeats (ITR). There are three copies of 40 bp short repeat sequences in the ITR. According to the DNA structural model of human adenovirus type 2, the 40 bp short repeats are the sites that are combined with nuclear factor 1 (NF-1) that is an initiation factor of DNA replication of virus, so they are essential structure of CLL DNA.

[Experimental study and clinical application of primary choledochorraphy after choledochotomy].

An animal model was established in dogs to compare primary choledochorraphy with the traditional T-tube method. Authors observed cholangic histologic changes under microscope and electron-microscope between the two groups. The result showed that primary choledochorraphy was better than the traditional method. The advantage of primary choledochorraphy lies in its simplicity, safety and reliability, which have been shown in 30 patients of cholelithiasis in clinical application. It is suggested that for patients primary choledochorraphy may clesirably be used for patients who are indicated for this approach.

[Monoclonal antibodies to human progesterone receptors: production, indentification and clinical application].

A peptide with 15 amino acid of human progesterone receptor (PR) was synthesized and used as antigen. Five hybridoma cells were obtained by cloning technique. In tests to hybridoma cell H4, the site specificity of the antibody to PR was found. The antibody reacted specifically with PR, but did not cross-react with ER, AR, or GR. Five women who used clomiphene citrate (CC) 100mg/d x 5 days were involved in a study of serum pregesterone (P) and endometrial histology in early luteal phase. There was no difference in serum P level in two groups. Three cases in CC group showed abnormal secretory endometrium. PR concentration in the nuclei of glandular cells of endometrium in CC group was significantly lower than that in the control group. In women with abnormal secretory endometrium, PR concentration in the nuclei of glandular cells was lower than that in women with normal endometrial histology. CC can influence endometrial histology by changing-PR concentration in the nuclei of glandular cells. The endometrium can not respond to the stimulus of P at normal levels and thus results in luteal phase defect.

[A study on the mechanism of severe hypoxemia associated with acute unilateral atelectasis in dogs].

OBJECTIVE: To study the mechanism of severe hypoxemia associated with acute unilateral atelectasis (AUA). METHODS: Six dogs with acute left side atelectasis were studied during the period of rest and exercise. The pulmonary shunt and PaO2 were measured at different levels of ventilation of right lung. RESULTS: After the left side ateloctasis during resting condition, the right lung could compensate to produce only mild hypoxemia (PaO2 9 kPa). But with further increased ventilation by exercise the shunt increased (from 19% to 29%) and led to severe hypoxemia (PaO2 8 kPa). When an artificial pneumothorax was instituted on the left lung, the shunt decreased (from 29% to 18%) and PaO2 increased (from 8 kPa to 9 kPa). CONCLUSIONS: After AUA, whether severe hypoxemia would occur is determined by the volume of ventilation of the normal side lung. The excess increase of ventilation of the normal side lung by any reason may result in severe hypoxemia.

[Cytoreductive surgery plus combined chemotherapy for the treatment of advanced ovarian cancer].

OBJECTIVE: To evaluate the impact of cytoreductive surgery plus combined chemotherapy on advanced ovarian cancer. METHODS: From Jan. 1980 to Dec. 1992, 76 patients admitted to our hospital for primary treatment, were eligible for retrospective evaluation. 26 patients with stage II and 50 with stage II. All patients with ovarian cancer were given cytoreductive surgery followed by systemic and intraabdominal chemotherapy. Regimens were CAP (Cyclophosphamide, Adriamycin, Cisplatin), AP (Adriamycin, cisplatin) or CE (Carboplatin, Epi-adriamycin). RESULTS: In 52 patients no residual tumor was found in 24 patients there were residual lesions less than 2cm in diameter. Postoperative chemotherapy ranged from 1 to 12 courses. The overall 5-year-survival rate was 33.6%, with 34.9% for stage II and 29.5% for stage II (P > 0.1). The 5-year-survival rates of cases with and without residual tumor were 16.5% and 37.6% respectively, the rates of those cases receiving less or more than 8 courses of chemotherapy were 20.0% and 60.1% respectively. CONCLUSION: The prognosis of ovarian cancer following cytoreductive surgery is influenced by residual tumor, and the number of courses of chemotherapy.

[The effects of lipoteichoic acid and calcium on the adherence of oral streptococci in vitro].

In this study, the effects of lipoteichoic acid (LTA) and calcium on adherence of oral streptococci in vitro were investigated. The results revealed that the adherence of streptococcus sobrinus to a glass surface was increased by the addition of calcium, in the condition of the same calcium ion concentration, LTA inhibited adherence, suggesting that LTA and the bacterial cell surface LTA competed for the same site of calcium ion, providing some evidence to support calcium-bridging theory. The result also showed that calcium ion not only influenced sucrose-independent adherence, but also infulenced sucrose-dependent adherence. The effect of calcium ion on the adherence has a threshold, over which the adherence was reduced.

[Detection of bcl-2/JH fusion gene in patients with non-Hodgkin's lymphoma by semi-nested polymerase chain reaction].

To study the incidence and clinical significance of bcl-2/JH fusion in 45 Chinese patients with non-Hodgkin's lymphoma, semi-nested polymerase chain reaction (PCR) was used to detect the fusion gene of non-Hodgkin's lymphoma. A total of 12 patients gave positive results, they include 9 of the 15 patients with follicular lymphoma and 3 of the 30 patients with diffuse B-cell lymphoma. The results were negative in 5 patients with reactive hyperplasia. The breakpoint was located within the major breakpoint region in 11 of the 12 patients, and the remaining patient had bcl-2 translocation in the minor cluster region. The results of the study show that bcl-2/JH fusion gene in non-Hodgkin's lymphomas is an important molecular biological marker and has a significant role in differentiating benign or malignant hyperplasia, determination of clonality, predicting prognosis and in the discovery of minor residual disease.

[Inhibitory effects of malignant phenotype of human bladder cancer cell line by c-Ha-ras, c-myc antisense oligodeoxynucleotide].

The effects of two antisense oligodeoxynucleotide on expression of c-Ha-ras, c-myc proto-oncogene and the growth of human bladder cancer cell line were observed. Synthetic 15-mer directed at the region of the translational initiation site of c-Ha-ras, c-myc proto-oncogene (ASO-r, ASO-m) greatly inhibited the proliferation and DNA synthesis of T24 cell line, inhibited the expression of rasp21, mycp62 protein. The power of tumorigenesis of cell line treated with ASO was decreased. The results implicate the potential value in bladder cancer gene therapy.

[Simple artificial mouth model].

A simple artificial mouth model is established under our laboratory condition. Development of monobacterial plaque and mixed bacterial plaque was studied in this artificial mouth model. The samples were subjected to viable count, microhardness measurement, etc. The result showed that the controlled conditions of the model can be used to study plaque development and earlier enamel lesion production on a time-dependent basis. It is concluded that the simple artificial mouth model is suitable for a wide range of dental applications.

Relationship between plasma membrane Ca(2+)-ATPase activity and acrosome reaction in guinea pig sperm.

The results obtained by biochemical measurement demonstrated for the first time that significant decrease of the plasma membrane Ca(2+)-ATPase activity occurred during capacitation and acrosome reaction of guinea pig sperm. Ethacrynic acid, one kind of Ca(2+)-ATPase antagonists, inhibited the plasma membrane Ca(2+)-ATPase activity, but calmodulin (50 micrograms/mL) and trifluoperazine (200-500 mumol/L) did not, suggesting that calmodulin is not involved in ATP-driven Ca2+ efflux from sperm. However, calmodulin is involved in the control of Ca2+ influx. TFP, one kind of calmodulin antagonists, accelerated the acrosome reaction and Ca2+ uptake into sperm cells significantly. Ca(2+)-ATPase antagonists, quercetin, sodium orthovandate, furosemide and ethacrynic acid promoted the acrosome reaction, but inhibited Ca2+ uptake, which cannot be explained by their inhibitory effects on the plasma membrane Ca(2+)-ATPase activity. It is speculated that this phenomenon might be caused by simultaneous inhibitions of the activities of Ca(2+)-ATPase present in the plasma membrane, the outer acrosome membrane and the outer mitochondrion membrane resulting in Ca2+ accumulation in the cytoplasm, which in turn blocks further Ca2+ entry through some negative feedback mechanism(s). The inhibitory effect of Ca(2+)-ATPase antagonist on glycolytic activity may also be the reason for Ca2+ accumulation in cytoplasm and inhibition of Ca2+ uptake.

[Cloning and sequence analysis of the gene encoding the second region of MSA1 from three Chinese isolates of Plasmodium falciparum].

Gene encoding the second region of merozoite surface antigen (MSA1-R2) of three Chinese isolates of Plasmodium falciparum from Hainan, Yunnan and Anhui were amplified by PCR, cloned into M13 and sequenced. Comparison among MSA1-R2 sequences from different isolates of P. falciparum shows that there is no difference among the three Chinese isolates. The sequence of MSA1-R2 of the Chinese isolates are highly homologous with that of WELLCOME from West Africa. The sequence flanking the upstream of the tandem repeats is exactly the same as that of MAD-20, but sequence downstream the repeats exhibit the characteristics of K1 allele, indicating that intragenic recombination of the two MSA1 allelic families exists in the Chinese isolates.
