[AFLP analysis on genetic structure of planted Acacia auriculiformis population in Heshan].

AFLP analysis of Acacia auriculiformis populations in 3 different planted communities with the same provenance and same planting time indicated that the genetic heterozygosity was quite high in the plantation population. 90.73% of total genetic variance distributed within a population, while only 9.27% among populations. The most distinct genetic structural difference was found between pure plantation and legume mixed plantation, next was between legume and non-legume mixed plantation, and the most similar was between pure and non-legume mixed plantation, which revealed that the genetic variation in this Acacia auriculiformis population was relatively higher than that from the natural forests by isoenzymic analysis data. The genetic differentiation was not the result of gene flow, but the result of the artificial selection and the microenvironmental changes.

[Experimental study on isolated testes with ischemia/reperfusion injury].

OBJECTIVE: To investigate the injury on isolated testes induced by ischemia/reperfusion(I/R), and the protective effect of Yisheng injection on the injury. METHODS: Twenty-six isolated cadaver testes contributed by 13 persons were preserved with 4 degrees C 250 ml hypertonic citrate alloxuric (HCA) solution and then reperfused with 37 degrees C 500 ml HCA. Solution of experimental group contained 500 micrograms/ml Yisheng injection. In simple cold preservation test, involving in 8 experimental and 8 control testes, a series of time points (6, 12, 18, 24, 36, 48, 60, 72 hours) were set to harvest. 10 testes (1 testis respectively on 6, 12, 18, 24 and 36 hours in experimental and control groups) were reperfused with 37 degrees C HCA for 6 and 12 hours. Histological and histochemical changes were observed. RESULTS: In the experimental testes, 4 degrees C cold preservation in 24 hours could not induce obvious pathologic changes. After 24 hours, changes such as swelling, vacuolar degeneration or detachment of endothelial cells (ECs), separation between basement membrane and seminiferous epithelium, mal-alignment of spermatogenous cell and edema of mesenchyme could be observed. In the testes preserved for 12 hours, the activity of lactic dehydrogenase(LDH) and succinic dehydrogenase (SDH) increased, then fallen after 24 hours. The activity of Nitric oxide synthetase(NOS) decreased after 18 hours. All changes were more obvious after following 37 degrees C reperfusion. In the control testes, swelling and vacuolar degeneration of ECs occurred on 12 hours cold preservation, and injury was worse along with the prolongation of cold preservation time. Pathologic changes of ECs, seminiferous epithelium and mesenchyme were serious after 37 degrees C reperfusion. CONCLUSION: 4 degrees C cold preservation in 24 hours can only cause mild ECs' injury, and obvious abnormal testes' histological profile can be observed beyond 24 hours. 37 degrees C reperfusion will make injury worse. Yisheng injection can keep isolated testes histologic structure well in 24 hours cold preservation, and it has protective effect on I/R injury.

[Early postoperative complications of choledochal cyst excision and reconstruction of biliary tract].

OBJECTIVE: To study the early postoperative complications of choledochal cyst excision with reconstruction of the biliary tract. METHODS: In last 10 years, 173 patients aged from 27 days to 14 years (mean = 2.4 years) were operated on with procedures of choledochal cyst excision and biliary tract reconstruction by jejunointerposition between the liver hilum and duodenum or Roux-en-Y hepaticojejunostomy in our hospital. Amongst these 173 patients, 16 were found to suffer from postoperative complications. Therefore, their clinical data were retrospectively analyzed, and the prevention and treatment of the early postoperative complications discussed. RESULTS: The morbidity of early postoperative complications was 9.3% (16/173). The early complications included biliary leakage in 10 patients, wound dehiscence of abdominal wall in 3, hepatic failure in 1, pancreatic fistula in 1, and postoperative enteric intussusception in 1. The morbidity of early postoperative complications in infants younger than 1-year old was significantly higher than that in children(chi 2 = 15.78, P < 0.01). However, there was no significant difference in the morbidity of early postoperative complications between patients undergoing biliary tract reconstruction by jejunal segment interposition and those undergoing that by Roux-en-Y hepaticojejunostomy (chi 2 = 0.07, P > 0.05). The total mortality in all the operated patients was 2.3% (4/173). Three patients died from biliary leakage and 1 from postoperative hepatic failure. CONCLUSIONS: Biliary leakage, wound dehiscence of abdominal wall and hepatic failure are major early postoperative complications. The morbidity in infants younger than 1-year old is higher than in children. The preoperative treatment of biliary tract infection, hypohepatia, and surgical skills during operation should be emphasized. Reoperation for biliary leakage should not be delayed and mycotic cholangitis after biliary leakage needs more attention. Tension suture in the fascial space of the abdominal wall is useful for the prevention and treatment of abdominal wall dehiscence.

[Analysis of pregnancy rates after treatment of tubal pregnancy with systemic methotrexate].

OBJECTIVE: To analyse the pregnancy rate after treatment of tubal pregnancy with the systemic methotrexate (MTX) injection. METHODS: From March 1985 to August 1999, 129 women with confirmed unruptured tubal pregnancy, and desiring to conceive were selected. Among them 60 women were successfully treated with systemic MTX, and 69 with unilateral salpingectomy. All cases were followed up for 1-15 years. RESULTS: The rates of subsequent intrauterine pregnancies (IUP) in the MTX group was 73% (44 cases) and of recurrent extrauterine pregnancies (EP) was 8% (5 cases). Among 69 patients treated by salpingectomy, the rates of IUP and EP was 70% (48 cases) and 4% (3 cases) respectively. The differences between the two groups were not statistically significant. CONCLUSIONS: The effect of conservative management with MTX was similar to those of salpingectomy. The rate of subsequent pregnancy did not increased.

[Effect of growth hormone on the outcome of ovulation induction in patients with polycystic ovary syndrome].

OBJECTIVE: To study the effect of growth hormone (GH) on the outcome of ovulation induction in patients with polycystic ovary syndrome (PCOS). METHODS: We examined serum sex hormone, GH and insulin like growth factor-II (IGF-II) basal levels by radioimmunoassay in 130 PCOS patients and 107 normal women. In addition, we observed the effect of GH on the outcome of ovulation induction in 7 poor responders to human menopausal gonadotropin (hMG) treatment. RESULTS: The mean serum GH level is (2.50 +/- 1.33) micrograms/L and (1.04 +/- 0.47) micrograms/L respectively in nonobese and obese PCOS patients which were significantly lower than those in controls [(5.30 +/- 2.26) micrograms/L, (2.95 +/- 1.49) micrograms/L respectively, P < 0.05]. The mean serum IGF-II level is (136 +/- 27) nmol/L in the obese PCOS patients, significantly greater than those in nonobese PCOS and controls (P < 0.05). When we used GH with hMG in 7 poor responders, the total amount of hMG required decreased from 1 to 12 amples and the duration of treatment shortened as compared with hMG alone. CONCLUSION: There is abnormal GH secretion in patients with PCOS, GH may improve the outcome of ovulation induction by gonadotropin.

[Clinical significance of sentinel lymph node biopsy for breast cancer].

OBJECTIVE: To study the significance of sentinel lymph node(SLN) biopsy (SLNB) in determining the extent of axillary dessection for cN0 breast cancer patients. METHODS: In 96 patients with cN0 breast cancer, sentinel lymph nodes identified by patent blue-V or methylene blue staining were excised for biopsy. patients then received operations for breast cancer including axillary dessection. RESULTS: SLN was identified in 91 patients. The total number of SLN identified was one in 54 cases(59.3%), 2 in 23 cases(25.3%) and > or = 3 in 14 cases(15.4%). In 24 of the 91 patients, SLN was positive for metastasis. In 13 the 24 patients, only SLN provided evidence of lymph node metastasis. The sensitivity of SLN biopsy was 87.5%, the specificity was 100%. It had 100% positive predictability and 95.7% negative predictability of lymph node metastasis. The accuracy rate of intraoperative imprint cytology examination of SLN was 92.1%, with a false negative rate of 10.0% and a false positive rate of 7.1%. The accuracy rate of frozen-section examination of SLN was 98.7% during operation, with a false negative rate of 5.0% but without false positive result. Immunohistochemical assay did not help demonstrate metastasis in SLN negative on routine pathologic examination. CONCLUSION: The SLN status can generally be a representation of axillary metastasis, SLNB is useful to determine the extent of axillary dessection in cN0 breast cancer patients. Methylene blue is as effective as patent blue-V in the identification of SLN.

[Effects of ovulation induction on expression of integrins alpha 4 beta 1 in endometrium].

OBJECTIVE: To study the effect of clomiphene citrate (CC) and human menopausal gonadotropin (hMG) on the expression of integrins alpha 4 beta 1 during implantation window in endometrium. METHODS: Endometrium integrin alpha 4 beta 1 expression was detected by immunohistochemistry method in the mid-secretory phase of 48 normal natural cycles. CC + hCG or CC + hMG + hCG cycles in 30 polycystic ovary syndrome (PCOS) patients and 48 normal women. RESULTS: The expression of endometrial integrin alpha 4 beta 1 was stronger in the natural cycles than those of ovulation induction cycles with CC + hCG or CC + hMG + hCG. So was the pregnancy cycles as compared with the non-pregnant cycles. CONCLUSION: The endometrial receptivity in the min luteal phase and pregnant rate declined during ovulation induction by CC + hCG or CC + hMG + hCG in either normal women or PCOS patients.

[Nonperforating trabecular surgery with reticulated hyaluronic acid implant].

OBJECTIVE: To evaluate the clinical results of nonperforating trabecular surgery (NPTS) with reticulated hyaluronic acid implant. METHODS: Twenty cases (28 eyes) of advanced open angle glaucoma (C/D = 0.86 +/- 0.14) underwent NPTS with reticulated hyaluronic acid implant. Postoperatively, the intraocular pressure (IOP), intraocular reactions and the bleb appearance were recorded, gonioscopy and ultrasound biomicroscopy were performed. RESULTS: The mean follow-up was (9.2 +/- 3.4) months. The mean IOP was (28.1 +/- 10.2) mm Hg preoperatively and (17.5 +/- 4.1) mm Hg postoperatively, the difference being significant (t = 5.776, P < 0.001). The number of drugs used for anti-glaucoma was (2.04 +/- 0.74) preoperatively, and (0.71 +/- 0.71) postoperatively (t = 8.103, P < 0.001). On the last visit, in 27 eyes (96.4%) the IOP was < or = 21 mm Hg, and among them, there were 12 eyes (42.8%) without anti-glaucoma medicine and 15 eyes with anti-glaucoma medicine. In 13 eyes (46.4%) the IOP was decreased > or = 10 mm Hg in which 6 eyes did not take any anti-glaucoma medicine and 7 eyes used anti-glaucoma medicine. There were no flare and cells in the anterior chamber in 14 eyes. In 10 eyes there was only slight flare that disappeared in 2-6 days. In 4 eyes there was a small amount of hyphema that was absorbed in 3-7 days. After operation, there was an obvious diffuse bleb in each eye, at the last visit, there were functional blebs in 19 eyes, the successful filtration was ceased in 9 eyes within 1.5-3 months. Intra-operatively, in 5 eyes microperforations occurred in the trabecular meshwork without iris prolapse, and after operation, under the gonioscope, the perforation could be observed in 3 eyes. In another 4 eyes, there was no intra-operative perforation, but a small fissure was seen by gonioscopy. In the post-operative 1-8 months, ultrasound biomicroscopy was performed, and it was shown that the implant was not degraded. CONCLUSIONS: The elevated IOP in patients with open angle glaucoma can be effectively reduced by NPTS, and the number of drugs used for anti-glaucoma was decreased. The post-operative complications were much less because of no intra-operative penetration of the anterior chamber. Therefore, NPTS is a very hopeful and practicable new anti-glaucoma surgery.

[Positive immunoglobulin A expression in human epithelial carcinoma cell lines].

OBJECTIVE: In the present study, we try to further elucidate which class of immunoglobulin is expressed in epithelial carcinoma cell lines. METHODS: Three assays, immunohistochemistry, Western blot and enzyme-linked immunosorbent assay (ELISA) were used to detect the cellular protein extracts and the cultivated supernatant. RESULTS: Seven human epithelial carcinoma cell lines, MCF-7 (human breast carcinoma cell line), SW480 (human colon carcinoma cell line), MGC (human gastric carcinoma cell line), HeLa (human cervical carcinoma cell line), HNE2, CNE1-LMP1 and Tet-on-LMP1-HNE2 (three human nasopharyngeal carcinoma cells) expressed IgA protein in their cellular extracts and cultivated supernatants. CONCLUSION: Immunoglobulin is expressed in epithelium-derived carcinoma cell lines, which is in conflict with the traditional immunoglobulin theory. Our findings imply that the immunoglobulin expressed in epithelial carcinoma cells might play a role in the carcinogenesis of epithelium-derived carcinoma. Understanding the mechanism of immunoglobulin expression by cancer cells may possibly further facilitate the improvement techniques in cancer diagnosis, therapy or the mechanism of carcinogenesis.

[Dexamethasone inhibits trabecular meshwork cell growth and epidermal growth factor mRNA expression in vitro].

OBJECTIVE: To investigate the effect of dexamethasone on cultured trabecular meshwork cell growth and the cell expression of epidermal growth factor (EGF) mRNA in vitro. METHODS: Human trabecular meshwork cells were cultured in vitro. The third passage cells were used in this experiment; with the addition of 300 microgram/ml dexamethasone in the culture medium in the test group, the culture was carried out as routine. After 5 days, the cell growth conditions were observed. After 7 days, the cells in the test and the control group were collected, and mRNA was drawn from them in both groups. EGFcDNA probe with alpha-(32)P isotope labeling was used to proceed dot blot hybridization and autoradiography to detect EGFmRNA of the cells. A computerized laser instrument was used to scan the size of dot blot hybridization, the relative value of the optical density was measured in the autoradiographs, and the inter-group comparison was made. RESULTS: Trabecular meshwork cells were inhibited by dexamethasone in the test group. The cells in the control group had been confluent at the 5th day, but the cells in the test group still grew like tribes. The total RNA was 14 microgram in the test group, while 22.5 microgram in the control group. In both groups, 14 microgram RNA was used for dot blot hybridization test with EGFcDNA probe. Autoradiographic image was positive in the two groups. The results of the scanning and the measurement of the autoradiograph density showed that dexamethasone obviously decreased the EGFmRNA expression of trabecular meshwork cells. CONCLUSIONS: Dexamethasone can inhibit trabecular meshwork cell growth in vitro. At first it affects the cell total RNA transcription and the second affects the EGFmRMA expression. It is suggested that steroid glaucoma be caused by the inhibition of various physiological and metabolic functions of trabecular meshwork cells induced by dexamethasone.

[The retinal nerve fiber layer defect and its related clinical features in early primary open-angle glaucoma].

OBJECTIVE: To investigate the distribution of diffuse and local retinal-nerve-fiber-layer defects (RNFLD) and their clinical features in early primary open-angle glaucoma (POAG) and normal-tension glaucoma (NTG). METHOD: The types of RNFLD in 81 cases with POAG and 70 cases with NTG were observed, the distribution of diffuse and local RNFLD were analyzed, and the maximum untreated intraocular pressure (IOP) and the history of optic disc hemorrhage between the two types of RNFLD in the two glaucomatous groups were compared. RESULTS: RNFLD appeared in 78 of 81 cases with POAG, 50 of them were diffuse and 28 were local; 27 of diffuse and 43 of local RNFLD were found in 70 cases with NTG. The IOP was (28.78 +/- 5.41) mm Hg in the diffuse and (26.2 +/- 5.2) mm Hg in the local type in POAG (t = 2.04, P < 0.05), and (19.2 +/- 1.9) mm Hg and (17.8 +/- 2.0) mm Hg, respectively in NTG (t = 2.94, P < 0.05). The incidence of disc hemorrhage was 10/78 and 18/70 in POAG and NTG, respectively (chi(2) = 4, P < 0.05). That was 3/50 in the diffuse and 7/28 in the local type in POAG (chi(2) = 4.22, P < 0.05), and 3/27 and 15/43, in NTG respectively (chi(2) = 4.91, P < 0.05). There were no statistic differences in the diffuse type between the POAG and NTG groups (chi(2) = 0.15, P > 0.05), and neither were in the local type (chi(2) = 1.31, P > 0.05). CONCLUSION: The diffuse RNFLD tends to be distributed in POAG, while the local RNFLD tends to be distributed in NTG. The maximum IOP in the eye with diffuse RNFTD is higher than that in the eye with local RNFLD, either in POAG or NTG group. The occurrence of disc hemorrhage is higher in the eye with local RNFLD than that in the eye with diffuse RNFLD in the two groups respectively. There are differences in clinical features between the two types of RNFLD in early POAG and NTG.

[The influence of surface roughness on bacteria adhesion on titanium].

OBJECTIVE: To investigate the correlation between surface roughness and bacteria adhesion. METHODS: Surface roughness of four abutments were determined by Talysurf/6p-120 topographer. Ra, Rz, Rp, Rv, Ry and Sm values were gathered. Commercial pure titanium and titanium alloy discs, S. mutans and A. viscosus were used in adhesion experiment. RESULTS: Ra values (in microns) of the tested abutments were: 0.1556, 0.2073, 0.6976, and 0.3811. No statistical difference was observed in bacterial quantity when Ra < 0.4528 microns and Rz < 3.3882 microns in adhesion experiment. CONCLUSIONS: The surface roughness of Ra < 0.4 micron and Rz < 3.4 microns is recommended for implant abutment.

[Determination of jujubaside A in semen ziziphi spinosae by RP-HPLC].

OBJECTIVE: To develop a new method for the determination of Jujubaside A in Semen Ziziphi Spinosae. METHOD: A RP-HPLC method was set up, using Hypersil-18 column, acetonitrile: water(30:70) as mobile phase with a flow rate of 0.8 ml.min-1. RESULT: The calibration curve was linear in the range of 0.076-0.456 mg. The average recovery was 93.5%. CONCLUSION: The method is appropriate for the determination of Jujubaside A.

[Experimental study of restiffening of the rigor mortis].

OBJECTIVE: To observe changes of the length of sarcomere of rat when restiffening. METHODS: We measured the length of sarcomere of quadriceps in 40 rats in different condition by scanning electron microscope. RESULTS: The length of sarcomere of rigor mortis without destroy is obviously shorter than that of restiffening. CONCLUSION: The length of sarcomere is negatively correlative to the intensity of rigor mortis. Measuring the length of sarcomere can determine the intensity of rigor mortis and provide evidence for estimation of time since death.

[The application of vascular endothelial growth factor in forensic science].

Vascular Endothelial Growth Factor (VEGF) is a potent angiogenic factor with a key role in several pathological processes, including wound repair as well as a effective vascular permeability factor. This article review the present study of VEGF in molecular biology, the connection with repair and expression regulation and so on.

[Effects of clasp designs of Kennedy Class II RPD on the displacement of alveolar bone around the abutment tooth--an under load hologram study].

OBJECTIVE: This article is to investigate the effects of different clasp designs of Kennedy class II RPD on the displacement of alveolar bone around the abutment tooth. METHODS: This experiment was carried out by using a separate human mandible. Five clasp designs were examined in this experiment, including RPA, RPI, modified RPI (mRPI), three-arm clasps and buccal wrought wired three-arm clasps. Hologramatic measurement was applied to record the different effects of these five designs of clasps on buccal and lingual alveolar bone. RESULTS: The RPI clasp was found to cause the smallest bone displacement. Generally, the clasps with the mesial occlusal rest (RPA, RPI, mRPI) caused smaller bone displacements than those with distal occlusal rests. The bar clasps brought about evenly spread bony displacements, however, the circumferential clasp caused highly concentrated displacement around apical site of the abutment tooth. CONCLUSION: The results suggest that mesial occlusal rests and bar clasps are preferable designs in this kind of prostheses.
