The effect of a single oral dose of 2.5 mg Bromocriptine on intraocular pressure over a 12 hour period in a group of 20 puerperal women with normal eyes, is reported. Intraocular pressure was significantly reduced, the maximal response being 3-4 hours after administration. Possible explanations for the reducing effect of Bromocriptine on intraocular pressure are discussed.
The effect of a single oral dose of 2.5 mg bromocriptine on intraocular pressure in 20 glaucoma patients is reported. Intraocular pressure was significantly reduced (31%), the maximal response being after 4-6 hours. A decrease in blood pressure was also noticed, maximal reduction (5%) at 4 hours. Three patients experienced nausea and dizziness. Possibly the reducing effect of bromocriptine on intraocular pressure is mediated by the stimulation of intraocular dopamine receptors (DA2).
PTH is initially synthesized as a larger precursor, containing a 25 amino acid signal sequence. Modification of cDNA encoding the hormone precursor resulted in the synthesis of proteins whose signal sequences were shortened at their amino termini. The effects of these mutations were analyzed using a cell-free translation system and rat pituitary GH4 cells in culture. Removal of the first six amino acids of the signal sequence had no effect on the efficiency or kinetics of protein processing as measured in the two assay systems. Mutants lacking 10 or 13 amino acids were not processed efficiently in the cells, nor were they translocated across microsomes in the cell-free translation system. These studies suggest that a modest change in the hydrophobic domain of the signal sequence, which might not have been predicted to alter function, led to a dramatic decline in signal activity.
cDNA clones corresponding to subunits of neuron-specific (gamma gamma and alpha gamma) and non-neuronal (alpha alpha) enolase isozymes were characterized from two mouse brain cDNA libraries. Our hybridization data revealed a partial homology of the coding sequences of mouse alpha, mouse gamma and rat gamma mRNAs. The noncoding sequences, however, appear to be specific for each mouse mRNA. Although coding for two polypeptides of the same molecular weight, the mRNA for the gamma subunit (2600 bases) is larger than that for the alpha subunit (1900 bases). The noncoding sequences for neuron-specific gamma mRNA (about 1300 bases) are therefore longer than those of the non-nervous tissue specific alpha mRNA (about 600 bases).
In the last decade, laser therapy has revolutionized the treatment of eye diseases. In many conditions, it has replaced conventional surgery. In others, it has opened up new avenues for the management of diseases that were considered untreatable before the laser era. Figure 1 summarizes the clinical applications of lasers in ophthalmology.
Two families with vitelliform macular dystrophy were investigated ophthalmologically and genetically. The pedigree examination verified the usual expressivity. Macular pigmentary abnormalities or extramacular vitellin deposits were disclosed in some asymptomatic patients.
Two concentrations of levobunolol (0.5% and 1%) and one concentration of timolol (0.5%) were evaluated for the control of elevated intraocular pressure (IOP) in a double-masked, randomized study. Fifty-one patients received one of the three study treatments in both eyes bid for one year. Both drugs were equally effective in reducing IOP: The overall reduction in mean IOP was slightly more than 9 mm Hg in all three treatment groups. Levobunolol was as safe and effective as timolol for the long-term control of elevated IOP.
A 73-year-old man was treated with oral glycerol solution for elevated intraocular pressure. Forty-five minutes later, he developed severe pulmonary edema. To our knowledge, this is the first documented case of this severe complication.

The significance of the time interval between topical instillation of two different ocular preparations in 63 normal subjects was investigated. The experimental model selected was pupillary dilatation by mydriatic agents. The results indicated that two drugs applied at the "same time" had an equal effect to drugs applied 10 minutes apart. The implications of the study to clinical practice are discussed.
A transconjunctival cryotechnique for the management of retrobulbar tumours is described. Fourteen retrobulbar tumours, evaluated by CT scan, were treated by this simple and minimally invasive operation. Patient's recovery is prompt and uneventful.
Retinal detachment secondary to choroidal nevus may be caused by subretinal fluid accumulation or neovascularization. Foveal subretinal fluid or leakage of choroidal neovascularization may impair visual acuity. 10 cases of nevus with serous sensory retinal detachment successfully treated with laser photocoagulation are reviewed. 2 additional cases of successful laser therapy for retinal detachment secondary to choroidal neovascularization with nevus of the choroid are presented.
The symptomatology of congenital fibrosis of extraocular muscles, markedly restricted eye movements, blepharoptosis and chin elevation were found in 10 individuals and three generations of one family. The mode of inheritance is compatible with a variable expressive dominant trait. Our experience with the surgical management of the condition using more aggressive procedures than currently employed is described.
We have used the method of heavy isotope labeling to study the metabolic turnover of acetylcholinesterase forms in the neuroblastoma-derived T 28 hybrid cells in their differentiated state. These cells contain mostly G1 and G4 forms, together with a small proportion of G2, and secrete all these forms into the culture medium. The cells maintained constant and equal levels of acetylcholinesterase, with the same proportions of molecular forms, in a medium containing heavy isotope-labeled amino acids and in a control light medium of similar composition. In addition, they secreted acetylcholinesterase at the same rate in both media. After transfer of the cells into the heavy medium, heavy isotope-labeled acetylcholinesterase molecules progressively replace preexisting light molecules. We analyzed heavy and light components of acetylcholinesterase for each of the two major G1 and G4 forms, by reconstructing the pattern obtained in sucrose gradient differential sedimentation, using combinations of weighted elementary distributions. Heavy molecules were detected in cellular extracts after about 30 min for G1 and 3 h for G4. Both heavy forms also appeared in the medium after a lag of about 3 h. The cellular complement of G1 was renewed much faster than that of G4, the levels of the light forms being reduced to 50% of the original level after 3.5 and 40 h, respectively. Each of these forms appeared to consist of several metabolic pools, and we present simplified models which describe their possible relationships.

Heart rate, systolic time intervals (pre-ejection period, left ventricular ejection time , ejection friction, stroke volume and QT interval of two cosmonauts (Leonid Popov--L.P. and Dumitru Prunariu--D.P.) were studied before, during, and after an ergometric bicycle exercise test performed before and after the seven-day Soviet-Romanian orbital flight on the Soyuz 40-Salyut 6 Complex in May 1981. For this purpose one precordial electrocardiogram (ecg) and the ear photodensitopram (den) were recorded stimulaneously. The method used permitted recording even during exercise, Ecg and den signals were stored on magnetic tape, processed in an analogue device and in a digital computer. The data obtained after landing suggest a slight cardiac deconditioning in L.P., demonstrated especially by augmentation of the pre-ejection period, which was unchanged in D.P. corresponding to a sympathoadrenergic hypertonia. The seven-day orbital flight has not produced important cardiovascular changes.
