Driving convection in a fluid layer by a temperature gradient or a heat current.

Bifurcation properties, stability behavior, dynamics, and the heat transfer of convection in a horizontal fluid layer that is driven away from thermal equilibrium by imposing a vertical temperature difference are compared with those resulting from imposing a heat current. Similarities and differences are elucidated. In particular various paradigmatic backwards bifurcating convection structures that occur, e.g., in binary fluid mixtures are determined numerically for the two different driving mechanisms. Conditions are given under which current driven convection is stable when temperature driven convection is unstable.

Measuring the transverse magnetization of rotating ferrofluids.

We report on measurements of the transverse magnetization of a ferrofluid rotating as a rigid body in a constant magnetic field, H0, applied perpendicular to the axis of rotation. The rotation of the fluid leads to a nonequilibrium situation, where the ferrofluid magnetization M and the magnetic field within the sample, H, are no longer parallel to each other. The off-axis magnetization perpendicular to H0 is measured as a function of both the applied magnetic field H0 and the angular frequency Omega. The latter ranges from a few hertz to frequencies well above a characteristic inverse Brownian relaxation time. Our experimental results strongly indicate that the transverse magnetization is caused only by a small fraction of the colloidal ferromagnetic particles. The effect of the polydispersity of the ferrofluid is discussed. Experimental results are compared to predictions based on several theoretical models. A single-time relaxation approach for the so-called effective field and a field-dependent Debye relaxation of M yield reasonably good shapes of the curves of transverse magnetization vs Omega. However, like the other models, they overestimate their magnitudes.

Competition between traveling fluid waves of left and right spiral vortices and their different amplitude combinations.

Stability, bifurcation properties, and the spatiotemporal behavior of different nonlinear combination structures of spiral vortices in the counterrotating Taylor-Couette system are investigated by full numerical simulations and by coupled amplitude-equation approximations. Stable cross-spiral structures with continuously varying content of left- and right-spiral modes are found. They provide a stability transferring connection between the initially stable, axially counterpropagating wave states of pure spirals and the axially standing waves of so-called ribbons that become stable slightly farther away from the onset of vortex flow.

Spiral vortices traveling between two rotating defects in the Taylor-Couette system.

Numerical calculations of vortex flows in Taylor-Couette systems with counter rotating cylinders are presented. The full, time-dependent Navier-Stokes equations are solved with a combination of a finite difference and a Galerkin method. Annular gaps of radius ratio eta=0.5 and of several heights are simulated. They are closed by nonrotating lids that produce localized Ekman vortices in their vicinity and that prevent axial phase propagation of spiral vortices. The existence and spatiotemporal properties of rotating defects, modulated Ekman vortices, and the spiral vortex structures in the bulk are elucidated in quantitative detail.

Traveling wave fronts and localized traveling wave convection in binary fluid mixtures.

Nonlinear fronts between spatially extended traveling wave (TW) convection and quiescent fluid and spatially localized traveling waves (LTWs) are investigated in quantitative detail in the bistable regime of binary fluid mixtures heated from below. A finite-difference method is used to solve the full hydrodynamic field equations in a vertical cross section of the layer perpendicular to the convection roll axes. Results are presented for ethanol-water parameters with several strongly negative separation ratios where TW solutions bifurcate subcritically. Fronts and LTWs are compared with each other and similarities and differences are elucidated. Phase propagation out of the quiescent fluid into the convective structure entails a unique selection of the latter while fronts and interfaces where the phase moves into the quiescent state behave differently. Interpretations of various experimental observations are suggested.

Oscillatory convection in binary mixtures: thermodiffusion, solutal buoyancy, and advection.

The role of thermodiffusive generation of concentration fluctuations via the Soret effect, their contribution to the buoyancy forces that drive convection, the advective mixing effect of the latter, and the diffusive homogenisation are compared and elucidated for oscillatory convection. Numerically obtained solutions of the field equations in the form of spatially extended relaxed traveling waves, of standing waves, and of the transient growth of standing waves and their transition to traveling waves are discussed as well as spatially localized convective states of traveling waves that are surrounded by the quiescent fluid.

Standing-wave oscillations in binary mixture convection: from the onset via symmetry breaking to period doubling into chaos.

Oscillatory solution branches of the hydrodynamic field equations describing convection in the form of a standing wave (SW) in binary fluid mixtures heated from below are determined completely for several negative Soret coefficients psi. Galerkin as well as finite-difference simulations were used. They were augmented by simple control methods to obtain also unstable SW states. For sufficiently negative psi, unstable SWs bifurcate subcritically out of the quiescent conductive state. They become stable via a saddle-node bifurcation when lateral phase pinning is exerted. Eventually their invariance under timeshift by half a period combined with reflection at midheight of the fluid layer gets broken. Thereafter, they terminate by undergoing a period-doubling cascade into chaos.

Spiral vortices and Taylor vortices in the annulus between rotating cylinders and the effect of an axial flow.

We present numerical simulations of vortices that appear via primary bifurcations out of the unstructured circular Couette flow in the Taylor-Couette system with counter rotating as well as with corotating cylinders. The full, time dependent Navier Stokes equations are solved with a combination of a finite difference and a Galerkin method for a fixed axial periodicity length of the vortex patterns and for a finite system of aspect ratio 12 with rigid nonrotating ends in a setup with radius ratio eta=0.5. Differences in structure, dynamics, symmetry properties, bifurcation, and stability behavior between spiral vortices with azimuthal wave numbers M=+/-1 and M=0 Taylor vortices are elucidated and compared in quantitative detail. Simulations in axially periodic systems and in finite systems with stationary rigid ends are compared with experimental spiral data. In a second part of the paper we determine how the above listed properties of the M=-1, 0, and 1 vortex structures are changed by an externally imposed axial through flow with Reynolds numbers in the range -40< or =Re< or =40. Among other things we investigate when left handed or right handed spirals or toroidally closed vortices are preferred.

Magnetization of concentrated polydisperse ferrofluids: cluster expansion.

The equilibrium magnetization of concentrated ferrofluids described by a system of polydisperse dipolar hard spheres is calculated as a function of the internal magnetic field using the Born-Mayer or cluster expansion technique. This paper extends the results of Huke and Lücke [Phys. Rev. E 62, 6875 (2000)] obtained for monodisperse ferrofluids. The magnetization is given as a power series expansion in two parameters related to the volume fraction and the coupling strength of the dipolar interaction, respectively.

Noise sensitivity of sub- and supercritically bifurcating patterns with group velocities close to the convective-absolute instability.

The influence of small additive noise on structure formation near a forwards and near an inverted bifurcation as described by a cubic and quintic Ginzburg Landau amplitude equation, respectively, is studied numerically for group velocities in the vicinity of the convective-absolute instability where the deterministic front dynamics would empty the system.

Spiral and Taylor vortex fronts and pulses in axial through flow.

The influence of an axial through flow on the spatiotemporal growth behavior of different vortex structures in the Taylor-Couette system with radius ratio eta=0.5 is determined. The Navier-Stokes equations (NSE) linearized around the basic Couette-Poiseuille flow are solved numerically with a shooting method in a wide range of through flow strengths Re and different rates of co-rotating and counter-rotating cylinders for toroidally closed vortices with azimuthal wave number m=0 and for spiral vortex flow with m=+/-1. For each of these three different vortex varieties we have investigated (i) axially extended vortex structures, (ii) axially localized vortex pulses, and (iii) vortex fronts. The complex dispersion relations of the linearized NSE for vortex modes with the three different m are evaluated for real axial wave numbers for (i) and over the plane of complex axial wave numbers for (ii) and (iii). We have also determined the Ginzburg-Landau amplitude equation (GLE) approximation in order to analyze its predictions for the vortex structures (ii) and (iii). Critical bifurcation thresholds for extended vortex structures are evaluated. The boundaries between absolute and convective instability of the basic state for vortex pulses are determined with a saddle-point analysis of the dispersion relations. Fit parameters for power-law expansions of the boundaries up to Re4 are listed in two tables. Finally, the linearly selected front behavior of growing vortex structures is investigated using saddle-point analyses of the dispersion relations of NSE and GLE. For the two front intensity profiles (increasing in positive or negative axial direction) we have determined front velocities, axial growth rates, and the wave numbers and frequencies of the unfolding vortex patterns with azimuthal wave numbers m=0,+/-1, respectively.

Localized waves without the existence of extended waves: oscillatory convection of binary mixtures with strong soret effect.

Spatially confined solutions of traveling convection rolls are determined numerically for binary mixtures such as ethanol-water. The appropriate field equations are solved in a vertical cross section of the rolls perpendicular to their axes subject to realistic horizontal boundary conditions. The localized convective states are stably and robustly sustained by strongly nonlinear mixing and complex flow-induced concentration redistribution. We elucidate how this enables their existence for strongly negative separation ratios at small subcritical heating rates below the saddle node of extended traveling convection rolls where the quiescent fluid is strongly stable.

Growth of binary fluid convection: role of the concentration field.

The growth of convection in binary fluid mixtures out of different perturbations of the quiescent conductive state is investigated using finite-difference numerical simulations for realistic ethanol-water parameters with strong negative Soret coupling between temperature and concentration fluctuations. Several different analysis tools are used to elucidate the complex spatiotemporal behavior associated with the dramatic concentration redistribution during the transients. It shows first the competition between counterpropagating waves that initially superimpose to form standing wave perturbations. Having reached a critical amplitude an advective breaking of the concentration wave triggers a very fast flow-induced transition from standing to traveling wave convection with large phase velocity and large concentration field amplitudes. Strongly nonlinear advective mixing and weak long-time diffusive homogenization then slow down the waves.

[Influence of postoperative radiation therapy of cerebral metastases on survival time and disease related to living condition].

Cerebral metastases are frequently observed in patients with systemic carcinoma as indication for new progress of the disease. Neurological deficits or seizures indicating cerebral metastases reduce the disease-related living conditions of the patients. Improving quality of life and survival time is the overriding goal of an early treatment after the diagnosis of cerebral metastases. Contemporary treatment include surgical removal of the cerebral metastases followed by whole brain irradiation and in some cases additional systemic chemotherapy for the primary tumor. This study was conducted to test the hypothesis that whole brain radiation following surgical removal improves the quality of life and the survival time in patients with cerebral metastasis. From January 1, 1994 to December 31, 2000, a total of 139 patients (mean age 59 +/- 2.3 years, m : f = 84 : 55) with cerebral metastases were investigated. Disease-related living conditions were assessed by Karnofsky score, the median time of follow-up was 11 months. For the analysis, patients were divided into groups with and without radiation therapy. Additionally, groups of patients with singular and two brain metastases were defined. In patients with singular brain metastases neither the survival time nor disease-related living conditions during the remaining life time was increased by postoperative whole brain irradiation. Almost all patients died due to the progression of the primary tumor. Patients with more than one metastases seemed to have a slight but not significant benefit from irradiation therapy after surgical removal of all metastases. In conclusion, these results indicate that an uncritical irradiation therapy of neurocranium after surgical removal of cerebral metastases is not beneficial in terms of survival time or disease-related living conditions.

Influence of inlet and bulk noise on Rayleigh-Bénard convection with lateral flow.

Spatiotemporal properties of convective fluctuations and of their correlations are investigated theoretically in the vicinity of the threshold for onset of convection in the presence of a lateral through-flow using the full linearized equations of fluctuating hydrodynamics. The effect of external forcing by inlet boundary conditions on the downstream evolution of convective fields is separated from the effect of internal bulk thermal forcing with the use of spatial Laplace transformations. They show how the spatial variation of fluctuations and of their correlations are governed by the six spatial characteristic exponents of the field equations.

Localized perturbations in binary fluid convection with and without throughflow.

Dynamics and structure of spatially localized convective perturbations in binary fluid layers heated from below and the effect of a plane horizontal Poiseuille throughflow on them are investigated. Fronts and pulse-like wave packets formed out of the three relevant perturbations-two oscillatory ones and a stationary one-are analyzed after evaluating the appropriate saddle points of the three respective dispersion relations of the linear field equations over the complex wave number plane. Front and pulse properties are elucidated in quantitative detail as a function of small throughflow Reynolds numbers for different Soret coupling strengths psi including the pure fluid limit psi=0 in comparison with the appropriate Ginzburg-Landau amplitude equation approximations. Furthermore, small amplitude pulses and fronts obtained from solving the full nonlinear field equations numerically are presented to check and compare with the linear results.

The rate of homozygous CDKN2A/p16 deletions in glioma cell lines and in primary tumors.

The rate of homozygous deletions of CDKN2A/p16 is variable between different tumor entities, and in addition it is higher in established cell lines in comparison with primary tumors. Such incongruencies may reflect statistical sampling errors, true differences depending on tissue derivatisation and CDKN2A/p16 loss under selective pressure in tissue culture. Clarification of these issues is warranted in the context of defining tumor suppressor genes such as CDKN2A/p16 as targets for gene replacement therapies. We therefore compared established cell lines derived from human glioblastomas and their corresponding primary tumors by multiplex PCR methodology. Archival early passages were included to determine the time point at which the p16 status of a cell line changes if it is different from the original tumor. It was found that in 2 of 11 cases (18%) the primary tumor had no p16 alteration whereas the corresponding cell lines had a homozygous p16 deletion. Tracking the in vitro evolution of these two cell lines we found that CDKN2A/p16 was lost already in the earliest passages. This suggests a clonal outgrowth advantage of a subpopulation of p16 deleted tumor cells rather than instability of the CDKN2A/p16 genotype in vitro. Including 20 additional glioblastoma-derived cell lines we detected that in 19 of the total 31 lines at least one exon was lost bringing the rate of p16 loss in the whole panel to 61%. This compares to a rate of 49% which was found in original glioma tissue from 47 unselected other patients. It is concluded, that in cell culture selective pressure favours the outgrowth of pre-existing CDKN2A/p16 negative clones, which account for the difference of CDKN2A/p16 status between cell lines and tumors. In no case did we see a change of the CDKN2A/p16 status during prolonged tissue culture periods of up to 8 years.

Foamy virus vectors for suicide gene therapy.

To improve the delivery of so-called suicide genes into tumors, recombinant retroviruses were constructed by inserting the herpes virus type 1 (HSV-1) thymidine kinase (tk), the E. coli cytosine deaminase (cd) and polynucleoside phosphorylase (pnp), or the jellyfish gene for the green fluorescent protein (gfp) into a foamy virus (FV)-derived replication-competent vector (pFOV-7). Expression and stability of the inserted foreign gene was analyzed by immunoblot and polymerase chain reaction (PCR). The functionality of the suicide genes was determined by a metabolic assay on virus vector infected cells and treatment with the respective prodrugs. In terms of vector stability and effectiveness of specific cell killing a virus transducing the pnp gene (FOV-7/pnp) was superior to those using the other two suicide genes. FOV-7/pnp is a candidate virus for suicide gene delivery into solid tumors.
