Iodine deficiency and brain development in the rat.

An assessment was made of the influence of low-iodine diet on somatic and brain development at birth (day 0) and 21 days postnatally in the rat. The rat mothers were proven to be iodine-deficient by assay of plasma thyroxine and thyroid stimulating hormone prior to mating, an at 21 days postnatally, when maternal thyroids were removed, weighed and stored for subsequent iodine analysis, along with those of the offspring. There were no significant differences in body weight or brain weight of the offspring at birth, or in the content of DNA or protein. However, at 21 days there was a significant reduction in body weight (21.7%) and whole brain weight (7.9%, P less than 0.2) which was associated with a significant fall n cholesterol content (12.4%, P less than 0.05) and protein level (9.6 %, P less than 0.01), while DNA was not significantly affected (6%). The greatest reduction in weight was seen in the cerebellum. The thyroids in these rats were double normal size, showed follicular cell hypertrophy and absence of colloid histologically, and contained 8% of the iodine content of controls. It is concluded that iodine deficiency retards both somatic and brain development, the change in the latter case being expressed as a reduction in cell size in the cerebral hemispheres and cerebellum, along with reduced myelination throughout the brain.

Evaluation of a new free-thyroxin assay.

The Amerlex Free Thyroxin (T4) Radioimmunoassay Kit (Amersham International Ltd.) is a new direct equilibrium radioimmunoassay for free T4 based on an antiserum with very high affinity for T4, and a unique 125I-labeled T4 analog as tracer. It is a very simple single-tube radioimmunoassay, making use of Amerlex particles to separate antibody-bound from free species. Interassay precision (CV) is 3.7% at 13 pmol/L and 2.3% at 30 pmol/L; within-assay precision is 4.2% at 21 pmol/L. The reference interval is 11-22 pmol/L. The assay did not misclassify any patients tested who had untreated myxedema or untreated thyrotoxicosis. The free T4 assay excelled both the free T4 index and the T4/T4-binding globulin ratio in correcting for increased thyroxin-binding globulin from pregnancy, and it was better than the index but not better than the ratio in correcting for increased thyroxin-binding globulin in users of oral contraceptives.

The effects of iopodate on the serum iodothyronine pattern in normal subjects.

The ingestion by normal subjects of 3 g of sodium iopodate, which is widely used in routine oral cholecystography, resulted in significant decreases of serum total and free T3 to a nadir on day 4 which averaged 43% and 40%, respectively, below initial mean values. Total and free rT3 increased markedly to a peak on day 3, 244% and 189%, respectively, above initial mean values. Total and free T4 and free T4 index rose to a maximum on day 4, but these changes were not statistically significant. A marked TSH increase was also seen, most evident on day 3. All these changes reverted to baseline values by day 14 at a time when serum total iodide was still markedly elevated. It is concluded that the changes observed after iopodate were not due to alterations in serum binding proteins nor to an effect on thyroid gland by the large iodine component of iopodate, but were consistent with an effect on the peripheral metabolism of T4. Difficulty in interpreting routine thyroid function tests may occur for up to 14 days after oral cholecystography with iopodate.

Humoral and cellular immunity in asthma.

Parameters of humoral and cellular immunity have been measured in 91 asthmatic patients. Mean serum levels of IgG and IgE were raised. IgG levels were higher in those with a family history of asthma. IgE levels were higher in those with a past history of atopic eczema, but intrinsic and extrinsic asthma could not be differentiated on the basis of IgE levels. Thirteen of 74 patients failed to respond to tetanus immunization, while only 1 failed to respond to Salmonella typhi H antigen. Tetanus nonresponders had a raised mean serum IgA level, reduced spontaneous lymphocyte tritiated thymidine uptake, and reduced thymidine uptake in fetal calf serum. Eight of 87 patients failed to mount delayed hypersensitivity reactions to a battery of five intradermal antigens. The tritiated thymidine uptake of lymphocytes stimulated with phytohemagglutinin was normal in autologous serum, but reduced in fetal calf serum. The data support the hypothesis that asthma may be associated with immunodeficiency states.
