Transport and budding at two distinct sites of visible nucleocapsids of West Nile (Sarafend) virus.

It has been difficult to detect and visualize the physical nucleocapsid particles during the replication process of the flaviviruses. The use of cryo-immunoelectron microscopy has clearly revealed the capsid proteins and nucleocapsid particles of West Nile (Sarafend) virus (a flavivirus) for the first time. Physical nucleocapsid particles accumulated in large numbers from 8 hr postinfection. Double immunolabeling of the envelope and capsid proteins showed a close association of these structural proteins for most of the replication cycle. By 10 hr postinfection, budding of nucelocapsids from the plasma membrane was very obvious. Although maturation at the plasma membrane was the dominant mode, during late infection, intracellular maturation into large vacuoles was also observed.

High therapeutic index of factor C Sushi peptides: potent antimicrobials against Pseudomonas aeruginosa.

Factor C protein isolated from the horseshoe crab, Carcinoscorpius rotundicauda, has endotoxin binding capability. Synthetic peptides of 34 amino acids based on the sequence of two regions of factor C (Sushi 1 and Sushi 3) as well as their corresponding mutants exhibited activities against 30 clinical isolates of Pseudomonas aeruginosa. Collectively, all four peptides demonstrated exceptionally effective bactericidal activity against P. aeruginosa with 90% minimal bactericidal concentrations (MBC(90)s) in the range of 0.06 to 0.25 microg/ml (16 to 63 nM). Viable bacteria were reduced by 90% after 7 min and were totally eradicated within 40 to 50 min. These peptides are minimally hemolytic against both rabbit and human erythrocytes even at concentrations up to 1,600-fold their MBC(90)s. Both in vitro and in vivo studies indicate that cytotoxic effects are small even at 1,000-fold their MBC(90)s. Furthermore, the Sushi peptides are tolerant of high-salt and adverse pH conditions. These findings demonstrate the promising therapeutic potential of the Sushi peptides.

Preliminary criteria for the definition of allergic rhinitis: a systematic evaluation of clinical parameters in a disease cohort (I).

BACKGROUND: The aim of this study is to formulate criteria for the definition of allergic rhinitis. Other studies have sought to develop scoring systems to categorize the severity of allergic rhinitis symptoms but it was never used for the formulation of diagnostic criteria. These other scoring systems were arbitrarily chosen and were not derived by any statistical analysis. To date, a study of this kind has not been performed. OBJECTIVE: The hypothesis of this study is that it is possible to formulate criteria for the definition of allergic rhinitis. This is the first study to systematically examine and evaluate the relative importance of symptoms, signs and investigative tests in allergic rhinitis. We sought to statistically rank, from the most to the least important, the multiplicity of symptoms, signs and test results. METHODS: Forty-seven allergic rhinitis and 23 normal subjects were evaluated with a detailed questionnaire and history, physical examination, serum total immunoglobulin E, skin prick tests and serum enzyme allergosorbent tests (EAST). RESULTS: Statistical ranking of variables indicated rhinitis symptoms (nasal, ocular and oronasal) were the most commonly occurring, followed by a history of allergen provocation, then serum total IgE, positive skin prick tests and positive EAST's to house dust mite, perennial rye and bermuda/couch grass. Throat symptoms ranked even lower whilst EAST's to cat epithelia, plantain and cockroach were the least important. Not all symptoms, signs and tests evaluated proved to be statistically significant when compared to a control group; this included symtoms and signs which had been considered historically to be traditionally associated with allergic rhinitis, e.g. sore throat and bleeding nose. CONCLUSION: In performing statistical analyses, we were able to rank from most to least important, the multiplicity of symptoms signs and test results. The most important symptoms and signs were identified for the first time, even though some of these were not included in our original selection criteria for defining the disease cohort i.e. sniffing, postnasal drip, oedematous nasal mucosa, impaired sense of smell, mouth breathing, itchy nose and many of the specific provocation factors.

Preliminary criteria for the definition of allergic rhinitis: a systematic evaluation of clinical parameters in a disease cohort (II)

BACKGROUND: This study is a development of the work done in 'Preliminary criteria for the definition of allergic rhinitis: a systematic evaluation of clinical parameters in a disease cohort (I)'. OBJECTIVE: We sought to develop criteria which could be routinely used to define allergic rhinitis. METHODS: A total of 47 allergic rhinitis and 23 normal subjects were evaluated with a detailed questionnaire and history, physical examination, serum total immunoglobulin (Ig) E, skin prick tests and serum EAST's. RESULTS: Based on this data, we developed a preliminary scoring system by which a reliable diagnosis of allergic rhinitis can be made, which is based on the prevalence adjusted strength of association delta ( partial differential) rank values described in this report. Cumulative frequency histograms were constructed for allergic rhinitis and normal subjects in a variety of configurations. CONCLUSION: A simple scoring system which can be used to diagnose allergic rhinitis on a routine basis was developed in this study.

Definition of endotoxin binding sites in horseshoe crab factor C recombinant sushi proteins and neutralization of endotoxin by sushi peptides.

Three truncated fragments, harboring different sushi domains, namely, sushi123, sushi1, and sushi3 domains, of Factor C were produced as biologically active secreted recombinant proteins. Sushi1 and 3 each has a high-affinity LPS binding site with K:(d) of 10(-9) to 10(-10) M. Positive cooperativity in sushi123 resulted in a 1000-fold increase in K:(d)2. The core LPS binding region of sushi1 and 3 reside in two 34-mer peptides, S1 and S3. A rigidly held disulfide-bonded structure is not essential but is important for LPS binding, as confirmed by a 100- to 10000-fold decrease in affinity. Both S1 and S3 can inhibit LAL reaction and LPS-induced hTNF-alpha secretion with different potency. LAL assay revealed that at least two molecules of S1 bind cooperatively to one LPS molecule, with Hill's coefficient of 2.42. The LPS binding by S3 is independent and noncooperative. The modified SDelta1 and SDelta3 peptides exhibited increased LPS neutralization potential although its LPS binding affinities indicated only a 10-fold improvement. Hence, the structural difference of the four sushi peptides conferred different efficiencies in LPS neutralization without altering their binding affinity for LPS. Circular dichroism spectrometry revealed that the four peptides underwent conformational change in the presence of lipid A, transitioning from a random coil to either an alpha-helical or beta-sheet structure. Two factors are critical for the sensitivity of Factor C to LPS: 1) the presence of multiple binding sites for LPS on a single Factor C molecule; and 2) high positive cooperativity in LPS binding. The results showed that in the design of an improved LPS binding and neutralizing peptide, charge balance of the peptide is a critical parameter in addition to its structure.

Microalbuminuria measurements by two in-house ELISA methods.

Detection of microalbuminuria is important in the management of diabetic patients since it is predictive of development of proteinuria and nephropathy. Two sensitive and specific in-house ELISAs for microalbuminuria were established and validated. One of the ELISAs was based on antigen coating while the other employed antibody coating. Recovery and linearity experiments gave acceptable results of 100 +/- 10%, while precision results were <10% for intra-assay and <12% for inter-assay coefficients of variation (CVs). The standard curve ranged from 10-625 ug/l, equivalent to 0.2-12.5 mg/l for urine samples diluted 1:20 fold. When the antibody coated ELISA was compared to antigen coated ELISA, a correlation of r=0.996 was obtained. When compared to commercial kits, the in-house ELISAs gave good correlations of r=0.961 versus the Boehringer Mannheim Micral Test strips and r=0.940 versus Ames Microalb Turbidimetry. The normal microalbumin reference ranges determined for 12h, first morning and random urine samples were 0.7-5.3 mg, 0.1-10.2 mg/l and 0.8-26.1 mg/l respectively. The normal albumin excretion rate (AER) was 1.0-7.3 ug/min while untimed urine samples gave results of 0.1-0.9 and 0.2-1.6 mg/mmol after dividing by creatinine concentrations. The ELISAs were used to detect microalbuminuria in 338 random urine samples from diabetic patients. A high percentage 47.9% was found to be positive for microalbuminuria and 18.0% had macroalbuminuria >25 mg/mmol. Thus screening for microalbuminuria together with creatinine measurements using random urine samples can be used for management of diabetic patients.

A different mode of entry by dengue-2 neutralisation escape mutant virus.

Entry processes were compared between dengue-2 (DEN-2) virus and a DEN-2 neutralisation escape mutant virus in baby hamster kidney (BHK) cells. The mutant virus (mu6B2) was resistant to neutralisation by a monoclonal antibody, MAb 6B2. Infection of BHK cells by the wild-type DEN-2 virus resulted in direct penetration of the virions into the cytoplasm whereas the mutant virus entered cells by endocytosis. The continual presence of the monoclonal antibody might have asserted some pressure for the mutant to adopt a different mode of entry.

Acoustic, aerodynamic, physiologic, and perceptual properties of modal and vocal fry registers.

The purpose of the study was to examine the acoustic, aerodynamic, physiologic, and perceptual characteristics of modal and vocal fry production. Twenty normal speakers (10 males, 10 females) participated in the study. Speech material included four sustained vowels (/i/, /a/, /ae/, /u/), and syllable strings of /pi/ repetitions produced in both modal and vocal fry registers. Acoustic data (fundamental frequency, jitter, shimmer, and signal-to-noise ratio), aerodynamic data (airflow and air pressure), and electroglottographic (EGG) data were obtained simultaneously. Results demonstrated considerable differences across voice parameters for the modal and vocal fry registers. Fundamental frequency was significantly lower in vocal fry than in modal register for both males and females, however, significant gender differences existed only in modal register. For both males and females, measurements of jitter and shimmer were significantly higher and signal to noise ratio was significantly lower in vocal fry. In addition, airflow rate in modal register was almost three times as high as the airflow rate in vocal fry register during sustained vowel production. During syllable string production, subglottal air pressure values in modal register were approximately 1.5 times higher than that in the vocal fry register. In general, these data emphasize that the aeromechanical mechanisms of vocal fold vibratory behavior are substantially different between modal and vocal fry registers. A model of vocal fry phonation is presented to account for the present results.

Perceptions of tonal changes in normal laryngeal, esophageal, and artificial laryngeal male Cantonese speakers.

Listeners were asked to identify six tonal variations in the production of the syllables /ji/ and /si/ produced by 10 normal laryngeal (NL), 10 standard esophageal (SE), and 10 Servox-type electrolarynx (EL) Cantonese speakers. The purpose of the present study was to determine if alaryngeal Cantonese speakers could differentiate meanings associated with different tone levels. Results indicated that listeners' identification of the six tones for each syllable produced by the SE speakers was similar in pattern to those produced by the NL speakers of Cantonese. For both NL and SE speech, the high-level, low-falling, and high-rising tones were perceived more accurately than the mid-level, low-level, and low-rising tones. Identification of the six tones for each syllable as produced by the EL speakers was significantly poorer due to the limitations of the electrolarynx.

Interaction of kunjin virus with octyl-D-glucoside extracted Vero cell plasma membrane.

Initial experiments using whole cells have shown that there were specific and saturable interactions between kunjin (KUN) virus and receptor molecules on the Vero cell surfaces. Solubilisation of Vero cell plasma membranes with octyl-D-glucoside (OG) yielded an extract which also interacted specifically with KUN virus. This was proven using electron microscopy. When the virus-OG-extract complex was exposed onto Vero cell monolayers, no KUN virus was observed to enter into the whole cells. This would imply that there was virus-receptor interaction with the OG-extract leaving no free virus to attach to the whole cells. The attachment kinetics of KUN virus was studied further using the Scatchard analysis which indicated the involvement of more than one interactive macromolecule in the attachment event.

Speech performance of adult cantonese-speaking laryngectomees using different types of alaryngeal phonation.

The purpose of the present study was to compare the speech performance of four types of alaryngeal phonation-electrolaryngeal (EL), pneumatic artificial laryngeal (PA), tracheoesophageal (TE), and standard esophageal (SE) speech-by adult Cantonese-speaking laryngectomees. Subjective ratings of (1) voice quality, (2) articulation proficiency, (3) quietness of speech, (4) pitch variability, and (5) overall speech intelligibility were given by eight naive individuals who had no prior experience with any form of alaryngeal speech. Results indicated that SE and TE speech was perceived to be more hoarse than PA and EL speech. EL speech was associated with significantly less pitch variability, and PA speakers produced speech with the least amount of perceived noise. However, articulation proficiency and overall speech intelligibility were found to be comparable in all four types of alaryngeal speakers.

Some aerodynamic and acoustic characteristics of acute laryngitis.

The purpose of the present study was to describe the effects of acute laryngitis on some aerodynamic, acoustic, and perceptual measures. Eleven subjects with diagnosed acute laryngitis due to upper respiratory infection were recorded during a laryngitic episode and 1 week to 10 days after amelioration of the laryngitic condition. Fundamental frequency, values, collapsed across the five vowels, were significantly reduced in the laryngitic compared with the normal speaking condition. The decrease in fundamental frequency associated with acute laryngitis suggests an increase in the mass of the vocal folds. In addition, aerodynamic values differed significantly for the laryngitic condition compared with the normal speaking condition, suggesting the presence of laryngeal hypofunction. Perceptual data indicated that speakers in the laryngitic condition were judged to have a hoarse voice when compared with the normal speaking condition.

A use study of speech pathology and audiology periodicals at Illinois State University.

No core list of periodicals exists for speech pathology and audiology. Faced with the prospect of having to cancer periodicals for all subjects, the science librarians at Illinois State University decided to determine which science periodicals were used most heavily. A one-year study of science periodical reshelving and interlibrary loan requests yielded ranked lists of periodicals important to speech pathology and audiology faculty and students at Illinois State University. The three most heavily used journals were the Journal of Speech and Hearing Research, ASHA, and Topics in Language Disorders. Most of the periodicals on the lists were indexed by either MEDLINE or UnCover, or by both. While the lists of journals developed in the study are not sufficient to serve as true core lists, they should be useful to libraries supporting comparable programs in speech pathology and audiology.

Perinatal vitamin A (retinol) status of northern Canadian mothers and their infants.

Vitamin A (retinol) status was determined in two groups living in the northern part of Canada: native (Indian and Inuit) and non-native (Caucasian). The dietary intake of vitamin A and its plasma concentration were measured prenatally, at delivery and postnatally in mothers. Plasma concentrations were also measured at birth and postnatally in their infants. The mean vitamin A intake of native mothers was significantly lower than that of non-native mothers, 661 +/- 485 versus 1,377 +/- 1,418 retinol equivalents (p < 0.00005), with a higher risk of deficiency without supplementation, 35% versus 8%. Plasma retinol concentrations, although not in the deficient range, were significantly lower in native than non-native mothers prenatally and postnatally. Infant mean plasma retinol concentrations at birth averaged only 52% of those of their mothers and were significantly lower among native than non-native infants although no clinical evidence of vitamin A deficiency was noted. We speculate that vitamin A supplementation in native Northern Canadian mothers during pregnancy and in their neonates during infancy may have a role to play in the prevention of vitamin A deficiency. We also postulate that plasma retinol concentrations of 50-60% of maternal levels and between 0.7 and 2.5 mumol/l represent a 'normal' range for newborn infants.

Usefulness and limitations of an in-house direct radioimmunoassay for 17-hydroxyprogesterone in serum.

Since conventional radioimmunoassays (RIA) for measurement of 17-hydroxyprogesterone (17-OHP) in serum samples require a laborious solvent extraction step, a direct and rapid in-house RIA was developed for early diagnosis and management of congenital adrenal hyperplasia (CAH). In-house rabbit anti-17-OHP antiserum, tritium labelled 17-OHP and dextran-coated charcoal were used in assay buffer with low pH 5.1 and preheated serum samples. Both inter- and intra-assay CVs were < 10% and the sensitivity was 1.2 nmol/l or 12 fmol/tube. Results from the direct assay correlated well with values from an extraction assay, r = 0.88 in samples from CAH patients, r = 0.85 in adults and children, 0.69 and 0.40 in term and preterm neonates respectively, 0.66 and 0.63 in luteal phase and third trimester pregnancy; p < 0.001 in all groups except p < 0.05 in preterm neonates. However, results from the direct assay were two to three times higher in serum samples from CAH patients, normal adults and children, but were five to seven times higher in pregnancy and term neonates and thirty times higher in preterm neonates. The markedly elevated levels measured by the direct assay are probably due to cross-reactivities with water-soluble steroid metabolites such as 17-hydroxypregnenolone sulphate and dehydroepiandrosterone sulphate (DHEAS). Although the direct assay is only useful as a screening test for preterm babies, it can be used for both diagnosis and monitoring of treatment of CAH in all other age groups.

Measurement of androstenedione levels by an in-house radioimmunoassay.

An in-house radioimmunoassay (RIA) for the measurement of androstenedione levels in serum was established and validated. Levels of androstenedione were measured by RIA using serum samples from various normal population groups and patients with congenital adrenal hyperplasia (CAH). Analytical recovery and linearity results were > 95%, while intra- and inter-assay CVs were < 10% and < 22% respectively. The assay sensitivity was 0.5 nmol/l or 25 fmol/tube. In normal population groups, the highest androstenedione levels were found in preterm neonates (1.6-12.4 nmol/l), followed by adult females (1.5-10.2 nmol/l), adult males (1.6-8.0 nmol/l) and term neonates (0.8-8.8 nmol/l), while the lowest values were observed in prepubertal children (0.5-3.4 nmol/l). There were no significant differences in diurnal variation and between follicular and luteal phases. The range of androstenedione levels in untreated or poorly controlled CAH patients (7.6-355.0 nmol/l, median 42.5 nmol/l, n = 20) were significantly higher (p < 0.001) than the upper normal limit of 3.4 nmol/L for prepubertal children. The normal androstenedione reference ranges for paediatric and adult groups have thus been established.

Presence of pro-opiomelanocortin peptides and corticotropin-releasing factor in human placenta.

Immunoreactive adrenocorticotropin (ACTH), beta-endorphin (BEP) and corticotropin-releasing factor (CRF) were detected in human term placenta obtained from elective Caesarian surgery. The concentrations of ACTH, BEP and CRF in placenta detected by radioimmunoassay (RIA) were 2.83 +/- 0.36, 0.52 +/- 0.05 and 0.56 +/- 0.15 ng/g wet weight of tissue respectively. Pro-opiomelanocortin (POMC) peptides were also detected in the amnion and chorion membranes and in the decidua. The concentrations of ACTH were 1.72 +/- 0.20, 4.43 +/- 0.39 and 5.80 +/- 0.17 ng/g and the levels of BEP were 0.42 +/- 0.18, 0.65 +/- 0.20 and 3.66 +/- 1.10 ng/g in the amnion, chorion and decidua respectively. In contrast to placenta, immunoreactive CRF was not detected in the amnion, chorion and decidua. Immunoreactive N-acetylated BEP was also not detected in all the placental subfractions. Comparison of the amounts of both ACTH and BEP in the various placental components indicated the following distribution: decidua > chorion > placenta > amnion. In decidua, POMC peptides were present in an equi-molar ratio but in the other three placental fractions, ACTH levels were three to five-fold higher than BEP. In immunohistochemical studies, only a positive staining for ACTH was obtained for decidua. Our results confirm the presence of POMC peptides and CRF in placenta and their physiological roles in pregnancy and parturition.

Immunochemical and genetic analysis of the p53 gene in liver preneoplastic nodules from aflatoxin-induced rats in one year.

Mutations of the p53 tumour-suppressor gene in human hepatocellular carcinomas from certain geographic areas appear to be associated with high dietary exposure to aflatoxin B1 (AFB1). In this study, the effects of AFB1 on p53 locus at the preneoplastic stage of rat liver oncogenesis were assessed. Male Wistar rats were treated with a single dose of 1.5 mg AFB1/kg body weight by a gastric tube. Liver biopsies over a period of one year were examined for aberrations of the p53 gene together with the expression of placental glutathione-S transferase (GST-P), a marker for preneoplasia. Immunohistochemistry, Western blot, polymerase chain reaction-single-strand conformation polymorphism (PCR-SSCP) and DNA sequencing techniques were used. AFB1 induction resulted in GST-P overexpression, forming GST-P-positive multi-foci and nodules of hepatocytes, but no aberrations in the p53 expression and the microstructure of exons 5-8 of the p53 gene. These results suggested that p53 mutation(s) might not occur at this early stage of AFB1-induced hepatocarcinogenesis.
