Hematologic and enzymatic analysis of Ctenopharyngodon idella x Hypophthalmichthys nobilis F1 hybrids.

1. Erythrocyte counts, hemoglobin concentrations and hematocrit values were determined for diploid and triploid Ctenopharyngodon idella X Hypophthalmichthys nobilis hybrids and the parental species. 2. Comparisons of diploid and triploid hybrids with the parental species revealed low erythrocyte counts for triploids, high mean corpuscular hemoglobin values for triploids, elevated hematocrits for diploids and triploids and similar hemoglobin concentrations for all fish. 3. Alkaline phosphatase, aldolase, and lactate dehydrogenase specific activities were determined spectrophotometrically. Levels of specific activity of these enzymes in the hybrids were consistently elevated above that of the parental species. These higher levels of enzyme activities in hybrids were probably the result of a breakdown in gene regulation.

Altered copper metabolism in heterozygous tortoiseshell (Moto/+) female mice, Mus musculus.

1. The copper concentration of various tissues and excrement of normal (+/+) and heterozygous tortoiseshell (Moto/+) female mice were examined by atomic absorption spectrophotometry (AAS). 2. The kidney, duodenum and spleen of Moto/+ female mice exhibited increased copper levels. 3. A reduced copper content was detected in the plasma and erythrocytes of Moto/+ females. 4. The feces and urine of Moto/+ females exhibited elevated copper concentrations. 5. The other tissues examined (e.g. liver, lung) showed no significant copper level alterations.

SEM analysis of body hairs and whiskers of heterozygous tortoiseshell (Moto/+) female mice (Mus musculus).

Back hairs of +/+ and Moto/+ female Mus musculus generally exhibited identical form when examined by SEM. However, the hair shafts of Moto/+ female mice were beaded in appearance (monilethrix), twisted (pili torti) or exhibited a rough nodular appearance. Also, some hairs of Moto/+ female mice which were devoid of pigment appeared enlarged and bitubular. The whiskers of +/+ and Moto/+ female mice were identical in form. The hair abnormalities of Moto/+ female mice resulted from a copper deficiency and were similar to those hair anomalies observed in other mottled mice and in copper-deficient crinkled mice.

Characterization of human erythrocyte alloantibodies by IgG subclass and monocyte interaction.

Human blood group antibodies of 11 specificities were examined for ability to mediate interaction with normal blood monocytes, IgG subclass composition, and titer score. Most antibodies were normally non-complement-binding (except anti-JKa) and clinically significant (except anti-Lub). A simple and relatively rapid in-vitro assay for the visualization of sensitized erythrocyte-monocyte interaction is described. Capillary procedures were adapted for IgG subclass composition determinations. Standard serologic tests were used to assess antibody titer score. A relationship appeared to exist between increased antibody titer score and increased interaction of sensitized erythrocytes (RBC) with normal blood monocytes (MNL), insofar as the mean titer score for antibodies that did mediate significant interaction was 52, whereas titer scores for antibodies that did not averaged 25. One or more examples within all antibody specificity, mediated significant RBC-MNL interaction. No relationship was observed between IgG subclass composition and mediation of RBC-MNL interaction except that all antibodies that did mediate interaction contained IgG1 or IgG3. Characteristic patterns of IgG subclass composition were observed for many antibody specificity groups, although too few examples were tested for valid statistical analysis. This study identifies two features of common blood group antibody populations that are instrumental in mediating erythrocyte destruction by monocytes in vitro. Similar information may be of value in predicting the result of erythrocytes transfused to patients with antibodies of unknown clinical significance.

BS II lectin: a second hemagglutinin isolated from Bandeiraea simplicifolia seeds with affinity for type III polyagglutinable red cells.

BS II lectin, a second hemagglutinin isolated from Bandeiraea simplicifolia seeds, agglutinated the red cells from 11 patients manifesting the 'acquired B' antigen phenomenon. The results of serological studies indicate that this lectin has specificity for the type III polyagglutinable receptors rather than for the 'B-like' antigens present on 'acquired B' red cells. Two other blood samples which did not possess 'acquired B' antigens were also found to react with BS II lectin. Both of these samples were shown to have properties similar to those reported for Tk red cells. The serological characteristics of red cells modified in vitro by a culture broth of Bacteroides fragilis suggest that type III polyagglutinable receptors are identical to those associated with Tk red cells.
