Expression of arginase by mouse myeloid leukemic cell differentiation in vitro induced with tumor necrosis factor.

Induction of arginase activity in mouse myeloid leukemic M1 cells by treatment with recombinant human tumor necrosis factor (rH-TNF) or TNF-elicited mouse serum (TNS) were examined in vitro. M1 cells differentiated into macrophage-like cells by addition of rH-TNF or TNS. The differentiated cells expressed phagocytic function and did not grow anymore. Cytolytic effect of rH-TNF or TNS was not observed. The differentiation of M1 cells into phagocytic cells by the TNS treatment was more rapidly than that by the rH-TNF treatment though TNS contained 25-time less amounts of TNF indicating species specificity of TNF action on myeloid leukemic cells or TNS containing other differentiation factor(s). 3H-ornithine formation from 3H-arginine is catalyzed by arginase (EC. 3.5.3.1). The enzyme product increased in the M1 cell culture medium by the treatment with rH-TNF or TNS. The arginase activity statistically correlated with the appearance percentage of differentiated cells with phagocytic function. These results suggest that in vitro differentiation of M1 cells is accompanied by induction of arginase activity.

[Clinical and pharmacokinetics studies of ceftriaxone upon 2g once daily administration in respiratory tract infections].

Clinical studies on ceftriaxone (CTRX) were conducted with 2 g once daily administration to respiratory tract infections. In addition, CTRX concentrations in serum, sputum and urine were determined. The results obtained are summarized as follows: 1. Clinical responses to CTRX in a total of 29 cases with respiratory tract infections were excellent in 7 cases, good in 13, fair in 8 and poor in 1 with a response rate of 69.0%. 2. CTRX concentrations in serum, sputum and urine (total and free body) were determined in 3 cases after intravenous drip infusion of 2g CTRX. Peak levels in sputum were 2.6 to 7.8 micrograms/ml, and CTRX maintained high sputum levels for 12 to 24 hours after administration.

[Left atrial myxoma in a 16-year-old girl following acute myocardial infarction--a case report and a review of 17 cases in the world].

A 16-year-old girl had clinical signs of an acute myocardial infarction. Echocardiography revealed left atrial myxoma and coronary angiography demonstrated a complete obstructive lesion at LAD. Coronary arterial embolization from myxoma was strongly suggested. The tumor was removed successfully 32 days after acute coronary embolization. Coronary embolization from left atrial myxoma is extremely rare. There are only 17 reports in the world and we reviewed these reports.

[Mitral and aortic valve replacement with tricuspid annuloplasty in a patient suffering from hereditary spherocytosis].

A 51-year-old woman was admitted with increasing shortness of breath. Preoperative evaluation disclosed MSr + ASr + TR. She had been undergone a splenectomy at 35-year-old for hereditary spherocytosis (HS). The patient underwent successful mitral valve replacement with a No. 25 Björk-Shiley (BS) valve, aortic valve replacement with a No. 21 BS valve, and tricuspid valve annuloplasty by means of membrane oxygenator (COBE). Haptoglobin and poloxamer 188 were used for prevention of hemolysis during extracorporeal circulation. No significant hemolysis occurred intra-or postoperatively. A very rare case of open heart surgery associated with HS was reported.

[Immunotherapy using high concentration purified antigen showed remarkable effect in all cases].

Hoya (sea-squirt) asthma is a typical type I occupational asthma. A total of 22 females with this disease whose age ranged from 22 to 69 years were treated for two years with high concentration purified sea-squirt antigen named Ei-M having a molecular weight of 22,800. One case who developed ordinary bronchial asthma abandoned the immunotherapy during the early stage of the therapy, but in the remaining 21 cases remarkable effects were rapidly observed from the early stage of treatment. These cases did not develop asthmatic attacks even through they engaged in oyster shucking work and no symptomatic therapy was indicated. Serum anti-Ei-M IgG antibody was significantly elevated after the therapy. None of the treated cases developed any side effects.

Free-radical reactions induced by OH-radical attack on cytosine-related compounds: a study by a method combining ESR, spin trapping and HPLC.

Free-radical reactions induced by OH-radical attack on cytosine-related compounds were investigated by a method combining ESR, spin trapping with 2-methyl-2-nitrosopropane and high-performance liquid chromatography (HPLC). Cytidine, 2'-deoxycytidine, cytidine 3'-monophosphate, cytidine 5'-monophosphate, 2'-deoxycytidine 5'-monophosphate and their derivatives, of which 5,6-protons at the base moiety were replaced by deuterons, and polycytidylic acid (poly(C] were employed as samples. OH radicals were generated by X-irradiating an N2O-saturated aqueous solution. Five spin adducts were separated by HPLC. Examination of them by ESR spectroscopy and UV photospectrometry showed that spin adducts assigned to C5 and C6 radicals due to OH addition to the 5,6 double-bond, a deaminated form of the spin adduct derived from a C5 radical due to the cyclization reaction between C5' of the sugar and C6 of the base, and a spin adduct assigned to the C4' radical due to H abstraction by OH radicals were produced. From these results the sites of OH-radical attack and the subsequent radical reactions in cytosine-related compounds were clarified.

Purification of staphylococcal exfoliative toxin by high pressure liquid chromatography.

Exfoliative toxin (ET) isolated from a clinical strain of Staphylococcus aureus was purified to homogeneity, using a 3-step HPLC system. NH2-terminal 20 amino residues of purified ET was found to be identical with ETA of S. aureus TA (7), S. aureus TC16 (9) and S. aureus ZM (10), but stability of purified ET was completely different from that of ETA. This purification system gave a high yield of pure ET, which exhibited higher purity than specimens purified by more complicated and time-consuming procedures. It is useful for small-scale purification for the comparative study of ET and easy to scale up for preparative purification.

Dose and dose-splitting effects of X-rays on lung tumour induction in mice.

This paper reports lung-tumour induction 12 months after single or split doses of X-rays in C3H/He male mice. The early proliferative response of lung cells after doses which induced lung tumours was also examined after single X-irradiation. The lung-tumour incidence tended to increase with increasing dose after a single irradiation and peaked at 5 Gy. At more than 10 Gy it decreased sharply to the control level. The mean tumour diameters tended to increase with doses up to 7.5 Gy and then decreased beyond 10 Gy. These results suggested that suppression of tumour growth reduced the tumour incidence at doses of over 10 Gy. The lung-tumour incidence decreased with increasing intervals between two equal doses of 2.5 or 5 Gy. The decrease was thought to be caused by the repair of the tumorigenic injury. However, the tumour incidence after two 2.5 Gy irradiations at 1 day intervals or two 5 Gy irradiations at 6 h intervals was higher than that observed after a single dose. This phenomenon was regarded as a progression of the tumorigenic injury. The labelling indices of the lung cells, determined using tritiated thymidine after a single irradiation, were higher than those of non-irradiated control cells. This response was delayed as the dose increased. The responses versus days after irradiation could be classified into three patterns on the basis of their peaks. The possibility that the larger delay after higher doses had some relation to the reduction of target cells for tumour incidence is suggested.

[Clinical features of 36 cases of psittacosis].

Clinical features of 36 cases of psittacosis experienced from 1980 to 1988 were analyzed. The following results were obtained. 1) Sixteen patients were male; twenty were female. The thirty-six patients ranged in age from 29 to 76 years. 2) Fifteen patients had been exposed to Parakeets prior to the onset of illness. 3) High fever (94.1%) and cough (94.4%) were prominent clinical symptoms. 4) The number of pneumonic foci per lung field by chest X-ray, showed the right inferior lobe was most frequently involved (45.2%). According to the nature of roentgenological shadows, the ground-glass-like shadow was most frequent (70.6%). 5) Leucocytosis was present in only three patients. The erythrocyte sedimentation rate and C-reactive protein were high in many patients. Half of the patients had elevated serum glutamate oxaloacetate transaminase and glutamate pyruvate transaminase. 6) All cases were treated with antibiotics and cured. The incidence of psittacosis may be increasing. Therefore, it is useful for clinicians to know the clinical features of this disease. These results may be useful in early diagnosis.

Effects of 3'-deoxyadenosine (cordycepin) on the repair of X-ray-induced DNA single- and double-strand breaks in Chinese hamster V79 cells.

The ability of cordycepin to inhibit the repair of DNA strand breaks was examined with X-irradiated Chinese hamster V79 cells in log-phase culture. A filter elution technique revealed that 70 microM cordycepin did not inhibit the repair of single-strand breaks but inhibited the repair of double-strand breaks. These findings confirmed the fact that the increase in the lethality of cordycepin in X-irradiated cultured mammalian cells was attributable to unrepaired DNA double-strand breaks.

Effect of radical scavengers on canine peripheral blood mononuclear lymphocyte-mediated cytotoxicity and production of active oxygen.

Production of active oxygen by canine peripheral blood mononuclear lymphocytes (PBLs) from beagle dog was examined by luminol-dependent chemiluminescence production. The canine PBLs rapidly produced the active oxygen in parallel with the number of cells when PBLs were cocultured with canine leukemia-derived CL-1 cells as target cells. Cytolysis of the target cells and active oxygen production were inhibited linearly by the addition of benzoic acid and n-propyl gallate as hydroxyl radical scavenger. However, superoxide dismutase and tiron which are scavengers of superoxide anion did not inhibit the cytotoxicity so much at low concentrations that inhibited the induction of luminol-dependent chemiluminescence. These results suggest that hydroxyl radical production by stimulated PBLs might be playing a major role of cytotoxic action in the case of canine system.

[A case of mediastinitis caused by rupture of esophageal diverticulum].

A 42-year-old male student from abroad visited a department of internal medicine of a hospital because of fever and dyspnea. A diagnosis of purulental mediastinitis with empyema was made. After drainage of the right thoracic cavity, the patient was referred to our department. Although consecutive treatment consisting of administration of antibiotics and thoracic drainage was conducted, dyspnea became worse, and widening of the mediastinum become severer. Mediastinal drainage was done via the parasternal approach under general anesthesia. Thereafter, the patient temporarily suffered from respiratory failure, and controlled respiration by a respirator was needed. Since the amount of discharge from the mediastinal and thoracic drains gradually decreased, and clinical symptoms disappeared, the drain tubes were removed. During the clinical course, laryngoscopy, bronchoscopy, esophago-gastro-endoscopic examination and esophagography were performed. Esophagoscopy revealed a diverticulum, and esophagography showed a fistula from the esophageal diverticulum to mediastinum. From these examination results, a diagnosis of mediastinitis caused by rupture of esophageal diverticulum was established.

[Septic pulmonary emboli caused by parenteral nutrition catheter infection].

A case of septic pulmonary emboli due to parenteral nutrition catheter infection was reported. Characteristic radiologic features were recognized. A 50-year-old man, who was receiving parenteral nutrition after total gastrectomy, consulted our department with complaints of fever and general malaise. A chest radiograph showed scattered ill-defined small peripheral nodules, which were not present before parenteral nutrition, and these nodules were quickly formed cavities + in 2nd day. He was suffering from high fever, hemo-sputum and dyspnea after removal of the parenteral nutrition catheter. Pseudomonas aeruginosa was isolated from the tip of parenteral nutrition catheter and sputum cultures. Septic pulmonary emboli were diagnosed and antibiotic therapy was performed. Bacterial endocarditis and septic thrombophlebitis were ruled out. The multiple cavity nodules extended to involve the peripheral areas of the lung and invasive shadows appeared on the chest radiograph in 8th day. Then, the invasive shadows disappeared and the walls of the cavitary lesions became thinner. After 2 months, all cavitary lesions disappeared with only linear shadows remaining.
